The enzymatic activity of four commercial enzymatic preparations (Peelzym I, II, III and IV) on citrus pectin, polygalacturonic acid and carboxymethylcellulose was determined (measured as the decrease in relative viscosity). In addition, the effectiveness of these preparations in the enzymatic degradation of the albedo and the segment membrane from Cimboa fruits was assessed. The highest activity on citrus pectin was shown by Peelzym II, although Peelzym I and IV activities were also elevated, 94.5 ± 6.2% and 88.7 ± 8.3% respectively of Peelzym II activity, and no relevant differences were found between them. Peelzym II also showed the highest activity for polygalacturonic acid, which was approximately 25% more than that of Peelzym I and IV, and more than double that of Peelzym III. Peelzym IV showed 40% more EM‐cellulase activity than Peelzym I, II and III. Segment membrane solution was degraded mainly by the enzymatic preparations Peelzym I and II. Thus, the most effective activities for the degradation of the carpelar membrane from Cimboa were those activities which act mainly on pectin and especially on polygalacturonic acid. However, the albedo was degraded to the greatest extent by Peelzym II and, in turn, the most important activities for albedo degradation were those which act on polygalacturonic acid. In addition, the concentration of the enzymatic preparation for the degradation of the carpelar membrane was lower than that required for albedo degradation. The effect of the calcination conditions on the properties and catalytic performance of hierarchical ZSM-5 zeolite has been investigated using different reactions as catalytic tests: acylation of aromatic substrates and rearrangement of linear and cyclic epoxides. The hierarchical h-ZSM-5 material was prepared by crystallization of silanized protozeolitic units. The removal of the organics present in the as-synthesized ZSM-5 samples has been carried out using two different calcination procedures: (i) directly under air atmosphere; (ii) following a two-step (nitrogen-air) treatment.Clear differences are noticed related to the textural properties of the ZSM-5 samples, since the h-ZSM-5 presents higher BET and external surface area than standard ZSM-5 zeolite. Whereas the calcination method does not affect the properties of the reference ZSM-5 sample, noticeable changes were observed over the hierarchical zeolite. The concentration of Brønsted acid sites decreases by half for one-step air calcination, but only a quarter when using a two-step nitrogen/air calcination, showing that the aluminium present in hierarchical ZSM-5 is very sensitive to the calcination conditions as it may undergo framework extraction and dehydroxylation phenomena. For all the studied reactions, the hierarchical sample, calcined by a two-step treatment, presents higher activity than when using direct air calcination.  相似文献

The FIPA Agent Communication Language includes a library of communicative acts supporting generic types of social interaction, such as information exchange and action performing. However, when developing specific applications, it is desirable to support agent interaction by means of more expressive constructs. In this paper, a structured approach to the design of communicative act libraries is put forward, which takes into account both the criteria of reusability and expressiveness. A linguistic approach is taken to ensure the expressiveness requirement. The proposal is illustrated by the design of an advisory sub-catalogue of reusable communicative acts.  相似文献

This paper studies the synthesis of structured triacylglycerols (STAGs) rich in palmitic and docosahexaenoic acids (PA and DHA) at sn-2 position and oleic acid (OA) at sn-1,3 positions by a four step process. First, triacylglycerols (TAGs) were obtained with 63–66 mol PA/100 mol total fatty acids and 10 mol DHA/100 mol by acidolysis of tuna oil and commercial PA, catalyzed by the non-positionally specific lipase Novozym 435. Then these TAGs were purified neutralizing the free fatty acids (FFAs) by KOH hydroethanolic solutions and extracting TAGs with hexane; these TAGs were completely recovered as pure TAGs (without FFAs). The third step involved the displacement of fatty acids located at sn-1,3 positions by acidolysis of PA and DHA enriched TAGs with OA rich FFAs, catalyzed by the sn-1,3 specific lipase DF from Rhizopus oryzae, immobilized on Accurel MP-1000; TAGs with 67 mol OA/100 mol at sn-1,3 positions and 52.1 and 15.4 mol PA and DHA, respectively, per 100 mol at sn-2 position were obtained. Both acidolysis reactions were carried out in stirred tank reactors (STRs) with lipase both dispersed in the reaction medium and contained in a cartridge filter attached to the stirrer rod. Finally STAGs were purified and obtained with yields of over 80 mol STAGs/100 mol STAGs in the reaction product (no FFAs were detected).  相似文献

External and internal quality parameters were measured in oranges (Citrus sinensis (L.) Osbeck cv. “Powell Summer Navel”) during on-tree ripening and at harvest using near-infrared reflectance (NIR) spectroscopy. The performance of two NIRS instruments was evaluated: a handheld microelectromechanical system spectrophotometer working in the 1,600- to 2,400-nm range, and a diode array visible–NIR spectrophotometer working in the 380- to 1,700-nm range. Spectra and analytical data were used to construct MPLS prediction models for quantifying weight, size (equatorial and axial diameters), color (L*, a*, b*, C*, h*, and color index), texture (firmness and maximum penetration force), yield (pericarp thickness, juice weight, and juice content), and chemical parameters (soluble solids content, pH, titratable acidity, and maturity index). Both instruments yielded promising results for on-tree and at-harvest quality measurements, but models constructed using the diode array instrument provided greater predictive capacity, particularly for fruit size (equatorial and axial diameters) and total soluble solids content. Subsequent evaluation of the LOCAL algorithm revealed that it increased the predictive capacity of models constructed for all the main parameters tested. These results confirm that noninvasive NIRS technology can be used to simultaneously evaluate external and internal quality parameters in intact oranges both during on-tree ripening and at harvest, thus making it easier for farmers to monitor the ripening process and also to optimize harvest timing in order to meet the demands of the citrus-fruit industry.  相似文献

The method exploits the correlation existing among the three colour planes by treating each pixel as a vector of three components, and performing region growing and difference operations using the vectors. The vaccinia virus 14-kDa protein (encoded by the A27L gene) plays an important role in the biology of the virus, acting in virus-to-cell and cell-to-cell fusions. The protein is located on the surface of the intracellular mature virus form and is essential for both the release of extracellular enveloped virus from the cells and virus spread. Sequence analysis predicts the existence of four regions in this protein: a structureless region from amino acids 1 to 28, a helical region from residues 29 to 37, a triple coiled-coil helical region from residues 44 to 72, and a Leu zipper motif at the C terminus. Circular dichroism spectroscopy, analytical ultracentrifugation, and chemical cross-linking studies of the purified wild-type protein and several mutant forms, lacking one or more of the above regions or with point mutations, support the above-described structural division of the 14-kDa protein. The two contiguous cysteine residues at positions 71 and 72 are not responsible for the formation of 14-kDa protein trimers. The location of hydrophobic residues at the a and d positions on a helical wheel and of charged amino acids in adjacent positions, e and g, suggests that the hydrophobic and ionic interactions in the triple coiled-coil helical region are involved in oligomer formation. This conjecture was supported by the construction of a three-helix bundle model and molecular dynamics. Binding assays with purified proteins expressed in Escherichia coli and cytoplasmic extracts from cells infected with a virus that does not produce the 14-kDa protein during infection (VVindA27L) show that the 21-kDa protein (encoded by the A17L gene) is the specific viral binding partner and identify the putative Leu zipper, the predicted third alpha-helix on the C terminus of the 14-kDa protein, as the region involved in protein binding. These findings were confirmed in vivo, following transfection of animal cells with plasmid vectors expressing mutant forms of the 14-kDa protein and infected with VVindA27L. We find the structural organization of 14kDa to be similar to that of other fusion proteins, such as hemagglutinin of influenza virus and gp41 of human immunodeficiency virus, except for the presence of a protein-anchoring domain instead of a transmembrane domain. Based on our observations, we have established a structural model of the 14-kDa protein.  相似文献

A comparison is made between the results obtained using the Hoek and Brown model, the characteristics method and the Meyerhof hypothesis, Serrano and Olalla (Ultimate bearing capacity at the tip of a pile in rock; theory (Part I). Int. J. Rock Mech. Min. Sci., 2002) and actual data from pile tip load tests. A comparison is also made with some of the theories postulated and recommendations given in this respect. It has been shown that the results obtained by this theory are similar when dealing with soft rocks (σc<20–30 MPa), but that the ultimate bearing capacity of hard rocks is overestimated. Different series of graphs are proposed for the application. A spreadsheet is also proposed for possible immediate application use with the aid of a personal computer. A specific calculation example is given for the ultimate tip bearing capacity of a pile.  相似文献

This study is concerned with the location of primary public schools. The public system should have the capacity to satisfy all the demand, although students may choose between public and private schools if they can afford the corresponding costs. A number of factors, such as questionable education quality, limited capacity, poor location and social preferences, secure a participation of about 30% to the private school system. The purpose of this study is both the evaluation of the existing public school network and a relocation proposal. The result of the former was the identification of areas with shortage and excess in school offer. The latter suggests school relocation using capacitated and uncapacitated models. ArcView, a software of the geographic information system (GIS) family, was employed, allowing the efficient handling of large problems and improving the presentation and evaluation of the findings. This methodology was applied to the primary public school network in the area of Vitoria, a state capital located in the southeast region of Brazil with about 300,000 inhabitants. Finally, the practical use of this study and its importance for planning purposes are discussed.  相似文献
