Bacterial adhesion onto abiotic surfaces is an important issue in biology and medicine since understanding the bases of such interaction represents a crucial aspect in the design of safe implant devices with intrinsic antibacterial characteristics. In this framework, we investigated the effects of nanostructured metal substrates on Escherichia coli adhesion and adaptation in order to understand the bio-molecular dynamics ruling the interactions at the interface. In particular, we show how highly controlled nanostructured gold substrates impact the bacterial behavior in terms of morphological changes and lead to modifications in the expression profile of several genes, which are crucially involved in the stress response and fimbrial synthesis. These results mainly demonstrate that E. coli cells are able to sense even slight changes in surface nanotopography and to actively respond by activating stress-related pathways. At the same time, our findings highlight the possibility of designing nanoengineered substrates able to trigger specific bio-molecular effects, thus opening the perspective of smartly tuning bacterial behavior by biomaterial design. 相似文献

In this work, we present a novel trained method for combining biometric matchers at the score level. The new method is based on a combination of machine learning classifiers trained using the match scores from different biometric approaches as features. The parameters of a finite Gaussian mixture model are used for modelling the genuine and impostor score densities during the fusion step.Several tests on different biometric verification systems (related to fingerprints, palms, fingers, hand geometry and faces) show that the new method outperforms other trained and non-trained approaches for combining biometric matchers.We have tested some different classifiers, support vector machines, AdaBoost of neural networks, and their random subspace versions, demonstrating that the choice for the proposed method is the Random Subspace of AdaBoost. 相似文献

Fluorescent silica nanoparticles (SiNPs) appear to be a promising imaging platform, showing a specific subcellular localization. In the present study, we first investigated their preferential mitochondrial targeting in myeloid cells, by flow cytometry, confocal microscopy and TEM on both cells and isolated mitochondria, to acquire knowledge in imaging combined with therapeutic applications. Then, we conjugated SiNPs to one of the most used anticancer drugs, doxorubicin (DOX). As an anticancer agent, DOX has high efficacy but also an elevated systemic toxicity, causing multiple side effects. Nanostructures are usually employed to increase the drug circulation time and accumulation in target tissues, reducing undesired cytotoxicity. We tested these functionalized SiNPs (DOX-NPs) on breast cancer cell line MCF-7. We evaluated DOX-NP cytotoxicity, the effect on the cell cycle and on the expression of CD44 antigen, a molecule involved in adhesion and in tumor invasion, comparing DOX-NP to free DOX and stand-alone SiNPs. We found a specific ability to release a minor amount of CD44+ extracellular vesicles (EVs), from both CD81 negative and CD81 positive pools. Modulating the levels of CD44 at the cell surface in cancer cells is thus of great importance for disrupting the signaling pathways that favor tumor progression. 相似文献

The paper presents a virtual diving system based on a virtual reality (VR) application for the exploitation of the Underwater Cultural Heritage. The virtual diving experience has been designed to entertain users, but its added pedagogical value is explicitly emphasized too. In fact, the ludic activities, consisting in the simulation of a real diving session from the point of view of a scuba diver, are following a storyline described by a virtual diving companion who guides users during the exploration of the underwater archaeological site. The virtual diving system provides general and historical-cultural contents, but also information about the flora and fauna of the specific submerged site to the users. The results collected through user studies demonstrate that the proposed VR system is able to provide a playful learning experience, with a high emotional impact, and it has been well appreciated by a large variety of audiences, even by younger and inexperienced users. 相似文献

Poly(meta‐phenylene isophtalamide) (PMIA) was processed via electrospinning to provide nanofibrous membranes with randomly and aligned fibers. Mechanical performance of such membranes was evaluated, applying a normalization procedure that takes into account the peculiar morphology of such complex substrate where voids can sum up to almost 80% of the sample volume. Random and aligned fibers membranes are applied onto wood panels to test their fire resistance in cone calorimetry when coated in polyaramidic thin nanofiber mats. Tests highlighted that random fibers provide a better fire protection, increasing Time to Ignition and decreasing the Fire Performance Index. Another important parameter affecting the performance is the adhesive system used to apply the nanofibers onto wood that is able to significantly modify the fire performance of the polyaramidic‐coated wood panels. Nanofibrous nonwovens show high versatility and outstanding properties, with reduced weight. Porous morphology, high material flexibility and deformability challenge their mechanical testing, severely affecting results reliability. Still today, a specific technical standard method to carry out tensile testing of nonwoven nanofibrous mats is lacking, as well as studies concerning tensile test data reliability. In this work, an accurate, systematic, and critical study is presented concerning tensile testing of nonwovens, using electrospun Nylon 66 random nanofibrous mats as a case study. Nanofibers diameter and specimen geometry are investigated to thoroughly describe the nanomat tensile behavior, also considering the polymer thermal properties, and the nanofibers crossings number as a function of the nanofibers diameter. Below a threshold value, which lies between 150 and 250 nm, the overall mat mechanical behavior changes from ductile to brittle, showing enhanced elastic modulus for a high number of nanofibers crossings. While specimen geometry does not affect tensile results. Stress–strain data are analyzed using a phenomenological data fitting model to better interpret the tensile behavior. The experimental results demonstrate the high reliability of the proposed mass-based load normalization, providing a simple, effective, and universally suitable method for obtaining high reproducible tensile stress–strain curves. 相似文献
