The possibility of differentiating Brucella species and biovars by Southern blot hybridization of agarose gel-electrophoresed HindIII-digested genomic DNA with membrane protein- or cytoplasmic protein-specific gene probes was investigated on 92 reference and field strains representative of all known species and biovars. Based on the RFLP pattern observed, three gene probes, i.e. br25, 39ugpa and omp16 coding for membrane or cytoplasmic proteins differentiated B. melitensis, B. ovis and B. suis biovar 2 strains from each other and from the other Brucella species and biovars. Thus, the use of these specific gene probes could contribute, in addition to previously identified species- or biovar-specific markers, to the molecular identification and typing of Brucella isolates.  相似文献

Invasive pulmonary aspergillosis (IPA) is an infectious complication appearing mainly in immunosuppressed patients, whose diagnosis is often difficult and lately made, and that usually bears a dismal prognosis. Patients diagnosed as having IPA from 1989 to 1994 were retrospectively analyzed. Probable IPA was diagnosed on the basis of a positive culture for Aspergillus together with a consistent radiological image. Confirmed IPA was diagnosed if there was, in addition to the former, a pathological examination showing Aspergillus hifae invading pulmonary parenchyma and/or pulmonary vessels. There were 25 men and 8 women with a mean age of 53.7 +/- 16.9 years (range: 22-86 years). IPA was confirmed in 11 cases and probable in 22. Sixty three percent of the patients had hematologic malignancy or solid cancer, whereas 30.3% did not have prior granulocytopenia or immunosuppressive therapy. The mean (SD) interval between admission and diagnosis was 40.2 (37.1) days (range: 1-180 days), and the diagnosis was made while the patient was still alive in 75% of the cases. Fifteen percent of the patients had extrapulmonary aspergillosis. The most frequent finding both on X-ray film of the chest and pulmonary computed tomography were bilateral multiple pulmonary nodules. Thirteen patients were treated with itraconazole, 6 with amphotericin B, 5 received both drugs, and 2 received fluconazole. Nineteen patients (57.6%) died and the case-fatality rate among treated patients was 46.1%. IPA presents mainly in immunosuppressed patients, but there was a not negligible proportion of patients lacking the classical risk factors. IPA is often a lately made diagnosis and in a quarter of the patients it is not made when the patient is alive. The most frequent radiological presentation are multiple bilateral nodules. The case-fatality rate of IPA is exceedingly high, even when if the patient has been adequately treated.  相似文献

In the present investigation, the effects of annealing in oxygen atmosphere on the electrical and optical properties of pulsed direct-current (DC) magnetron-sputtered ZnO thin films have been studied. With annealing, the electrical conductivity was found to increase from 2.3 S cm to 123 S cm. The optical transparency was also found to improve from 83% to 90%. The improvement in the electrical properties with annealing is attributed to the reduction of the grain boundary potential from 34 meV to 8 meV. X-ray photoelectron spectroscopy (XPS) measurements revealed oxygen deficiency in as-deposited films, whereas adequate incorporation of oxygen was observed in the annealed films. Ultraviolet photoelectron spectroscopy (UPS) measurements showed a shift of +0.8 eV in the Fermi level with annealing. This shift indicates significant changes in the electrical conductivity with annealing, which is due to an increase in the carrier concentration.  相似文献

We evaluated phospholipase activity in the intestine of rats and other species. Phospholipase activity was assayed by a surface barostat technique or an egg yolk titration system. Mucosal activity was found only by the surface barostat technique with phosphatidylglycerol as substrate; it was not found with phosphatidylcholine as substrate in assays by either technique. In gut luminal fluid activity was found when both phosphatidylcholine and phosphatidylglycerol were used as substrate in assays by the surface barostat technique, and phosphatidylcholine as substrate yielded activity in egg yolk titration. In rats in which pancreatic juice had been diverted, mucosal and gut luminal phospholipase activity was greater than in controls, thus demonstrating that enzyme activity was not due to pancreatic phospholipase. Bacterial origin of phospholipase activity was excluded in that phospholipase activity was found in germ-free rats; gastric and salivary gland origins were excluded in that continued phospholipase activity was found in rats with gastric fistula. The physiological importance of the enzyme was established by the finding that rats with pancreatic fistula absorbed 111 mumol of phosphatidylcholine and that controls absorbed 119 mumol of a 135-mumol load. Activity was found to be three times greater in the distal than in the proximal intestine; in cryptal cells it was 10 times greater than in villus tip cells. 65% of the activity in the gut lumen was tightly bound to particulate matter. We propose that intestinal phospholipase may be important in gut bacterial control, in the digestion of vegetable matter (phosphatidylglycerol is a major phospholipid in both plants and bacteria), and in the digestion of phospholipids in the gut lumen.  相似文献

This paper evaluates the performances of a neural network approach to estimate LAI from CYCLOPES and MODIS nadir normalized reflectance and LAI products. A data base was generated from these products over the BELMANIP sites during the 2001-2003 period. Data were aggregated at 3 km × 3 km, resampled at 1/16 days temporal frequency and filtered to reject outliers. VEGETATION and MODIS reflectances show very consistent values in the red, near infrared and short wave infrared bands. Neural networks were trained over part of this data base for each of the 6 MODIS biome classes to retrieve both MODIS and CYCLOPES LAI products.Results show very good performances of neural networks to estimate the original LAI products with an overall root mean square error (RMSE) around 0.5 for MODIS LAI from both MODIS and CYCLOPES normalized reflectances and a RMSE ranging between 0.12 (CYCLOPES reflectances) and 0.29 (MODIS reflectances) for CYCLOPES LAI. A drop of 15% of performance was found by training MODIS biome dependant algorithm by a single network over all the classes at the same time. More detailed analyses show that CYCLOPES and MODIS LAI values are very consistent for grasses and crops. Conversely, other biomes including shrubs, savanna, needleleaf and broadleaf forests show significant discrepancies, mainly due to differences between LAI definitions used between CYCLOPES (closer to effective LAI) and MODIS (closer to true LAI). However, products derived from the original CYCLOPES LAI products show a better agreement with both effective and true LAI ground measurements values. MODIS LAI products show more instability, partly because of the slightly shorter temporal resolution as compared to CYCLOPES.These results confirm the interest and versatility of neural networks for operational algorithms. This approach could be extended to other products or sensors, and may constitute a step forward for the fusion of data from several sensors, hence contributing to develop ‘virtual constellations’.  相似文献

The Codex Alimentarius in its General Principles committee proposes the definitions of risk assessment and risk management linked to ingestion of food additives and food contaminants. Moreover, three European directives imply for all member states an evaluation of food additives intakes by general population and by possible "at risk groups of consumers". This work is conducted in the general case of European Scientific Cooperation and is coordinated by United-Kingdom. This paper describes the French approach, using a step-by-step procedure to evaluate food additives consumption. It emphasizes the importance of scientific works in this field of research to obtain french datas about french consumptions.  相似文献

Auger electron-emitters increasingly attract attention as potential radionuclides for molecular radionuclide therapy in oncology. The radionuclide technetium-99m is widely used for imaging; however, its potential as a therapeutic radionuclide has not yet been fully assessed. We used MDA-MB-231 breast cancer cells engineered to express the human sodium iodide symporter-green fluorescent protein fusion reporter (hNIS-GFP; MDA-MB-231.hNIS-GFP) as a model for controlled cellular radionuclide uptake. Uptake, efflux, and subcellular location of the NIS radiotracer [99mTc]TcO4 were characterised to calculate the nuclear-absorbed dose using Medical Internal Radiation Dose formalism. Radiotoxicity was determined using clonogenic and γ-H2AX assays. The daughter radionuclide technetium-99 or external beam irradiation therapy (EBRT) served as controls. [99mTc]TcO4 in vivo biodistribution in MDA-MB-231.hNIS-GFP tumour-bearing mice was determined by imaging and complemented by ex vivo tissue radioactivity analysis. [99mTc]TcO4 resulted in substantial DNA damage and reduction in the survival fraction (SF) following 24 h incubation in hNIS-expressing cells only. We found that 24,430 decays/cell (30 mBq/cell) were required to achieve SF0.37 (95%-confidence interval = [SF0.31; SF0.43]). Different approaches for determining the subcellular localisation of [99mTc]TcO4 led to SF0.37 nuclear-absorbed doses ranging from 0.33 to 11.7 Gy. In comparison, EBRT of MDA-MB-231.hNIS-GFP cells resulted in an SF0.37 of 2.59 Gy. In vivo retention of [99mTc]TcO4 after 24 h remained high at 28.0% ± 4.5% of the administered activity/gram tissue in MDA-MB-231.hNIS-GFP tumours. [99mTc]TcO4 caused DNA damage and reduced clonogenicity in this model, but only when the radioisotope was taken up into the cells. This data guides the safe use of technetium-99m during imaging and potential future therapeutic applications.  相似文献

The French manufacture of Sèvres, famous for its production of fine porcelain artifacts, has been synthesizing pink pigments mainly composed of the spinel phase ZnAl2?xCrxO4 with x varying from 0.25 to 0.41 since the middle of the 19th century. This kind of pigment is mixed with an uncolored frit to obtain decorations for porcelain artifacts. However, the pink color of the pigment is altered in a particular uncolored frit and a brownish color appears. The mechanism of this color change was investigated. Observations under a scanning electron microscope revealed the formation of a phase rich in Cr resulting from reactions between the uncolored frit and the pigment during firing. X‐ray diffraction combined with Rietveld refinements and X‐ray absorption near edge structure measurements at the Cr K‐edge showed that the new formed phase belongs to the same spinel phase ZnAl2?xCrxO4 than the pigment, but with a higher Cr content x. We showed that its formation and thus the stability of the pigment is driven by the Al content in the uncolored frit.  相似文献

In order to fully validate actively cooled tungsten plasma facing components (industrial fabrication, operation with long plasma duration), the implementation of a tungsten axisymmetric divertor structure in the tokamak Tore-Supra is studied. With this major upgrade, so-called WEST (Tungsten Environment in Steady state Tokamak), Tore-Supra will be able to address the problematic of long plasma discharges with a metallic divertor target.To do so, it is planned to install two symmetric divertor coils inside the vacuum vessel. This assembly, called divertor structure, is made up of two stainless steel casings containing a copper winding pack cooled by a pressurized hot water circuit (up to 180 °C, 4 MPa) and is designed to perform steady state plasma operation (up to 1000 s).The divertor structure will be a complex assembly ring of 4 m diameter representing a total weight of around 20 tons. The technical challenge of this component will be the implementation of angular sectors inside the vacuum vessel environment (TIG welding of the coil casing, induction brazing and electrical insulation of the copper winding). Moreover, this complex assembly must sustain harsh environmental conditions in terms of ultra high vacuum conditions, electromagnetical loads and electrical isolation (13 kV ground voltage) under high temperature.In order to fully validate the assembly and the performance of this complex component, the production of a scale one dummy coil is in progress.The paper will illustrate, the technical developments performed in order to finalize the design for the call for tender for fabrication. The progress and the first results of the simplified dummy coils will be also addressed.  相似文献
