The electrodiffusion method has been used in fluid dynamic research for the past 50 years. It allows the measurement of wall shear stress, a crucial parameter, e.g., for the cleaning of membrane modules used in water filtration. Various authors have published articles dealing with the theory behind this technique. But no paper collects all the knowledge assembled over five decades of application. Here, comprehensive summary of the theory of steady flow, unsteady flow, and transient voltage step experiments is given. Factors influencing the accuracy of the measurements are discussed. Furthermore, a new approach to calibrate the system from voltage step experiments is introduced, and practical issues related to its application in flow measurements are discussed for an exemplary signal response to a near‐wall flow. 相似文献

Despite decades of skin research, regulation of proliferation and homeostasis in human epidermis is still insufficiently understood. To address the role of mitoses in tissue regulation, we utilized human long-term skin equivalents and systematically assessed mitoses during early epidermal development and long-term epidermal regeneration. We now demonstrate four different orientations: (1) horizontal, i.e., parallel to the basement membrane (BM) and suggestive of symmetric divisions; (2) oblique with an angle of 45°–70°; or (3) perpendicular, suggestive of asymmetric division. In addition, we demonstrate a fourth substantial fraction of suprabasal mitoses, many of which are committed to differentiation (Keratin K10-positive). As verified also for normal human skin, this spatial mitotic organization is part of the regulatory program of human epidermal tissue homeostasis. As a potential marker for asymmetric division, we investigated for Numb and found that it was evenly spread in almost all undifferentiated keratinocytes, but indeed asymmetrically distributed in some mitoses and particularly frequent under differentiation-repressing low-calcium conditions. Numb deletion (stable knockdown by CRISPR/Cas9), however, did not affect proliferation, neither in a three-day follow up study by life cell imaging nor during a 14-day culture period, suggesting that Numb is not essential for the general control of keratinocyte division. 相似文献

In flame spray pyrolysis (FSP), the evolution of metal oxide nanoparticles relies on quite a number of droplet (liquid) and vapor phase related physical mechanism as for instance precursor evaporation, oxidation, nucleation via gas-to-particle conversion mechanism, and subsequent particle (solid) growth mechanisms based on coagulation, sintering/coalescence, and agglomeration. The liquid precursor and dispersion oxygen feed rates are relevant control parameters of the FSP process for tailoring the nanoparticle size (diameter) and structure as well as the atomizer nozzle configuration. Sophisticated nonintrusive, laser-based in situ and ex situ diagnostics with multiscale spatial resolution (micrometer to meter range) are applied for analyzing droplet formation and size, gas velocity, temperature, species concentration, as well as primary and agglomerate diameters along the flow direction. Computational fluid dynamics (CFD) are coupled with population balance modeling (PBM) to elucidate the nanoparticle dynamics within the reactive spray. It is found that the CFD-PBM approach allows estimations of primary and agglomerate nanoparticle diameters within 80 and 75% accuracy compared to experimental data, suggesting that the methods presented could pave the way for designing next-generations of flame reactors. 相似文献

Based on the previously reported potent and selective sulfone hydroxamate inhibitors SC-76276, SC-78080 (SD-2590), and SC-77964, potent MMP inhibitors have been designed and synthesized to append a boron-rich carborane cluster by employing click chemistry to target tumor cells that are known to upregulate gelatinases. Docking against MMP-2 suggests binding involving the hydroxamate zinc-binding group, key H-bonds by the sulfone moiety with the peptide backbone residues Leu82 and Leu83, and a hydrophobic interaction with the deep P1’ pocket. The more potent of the two triazole regioisomers exhibits an IC50 of 3.7 nM versus MMP-2 and IC50 of 46 nM versus MMP-9. 相似文献

KTCTL-26 and KTCTL-2 are renal cell carcinoma (RCC) lines with high and lowexpression of P-170 glycoprotein, respectively. Inherent differences between the two cell lines in terms of phosphate metabolites and growth characteristics in culture were examined for possible association with multidrug resistance (MDR). Differences in response to drug treatment were investigated for 40 h incubations with various doses of vinblastine (VBL) alone or as cotreatments with various concentrations of the calcium antagonist diltiazem (DIL) and/or interferon–α (IFN-α). Treatment effects were quantitated using the MTT survival assay and 31P magnetic resonance spectroscopy (MRS) to determine phosphate metabolite profiles in intact cells. KTCTL-2 and KTCTL-26 cells exhibited significant inherent differences in phosphocholine, glycerophosphocholine, glycerophosphoethanolamine, and phosphocreatine levels. KTCTL-26 cells were more sensitive than KTCTL-2 to 0.011μM VBL alone (87% vs. 102% survival) or to 0.011μM BL + 10μM DIL (55% vs. 80% survival). The latter treatment resulted in a significant decrease in the ratio of phosphocholine to glycerophosphocholine in KTCTL-26 cells but no significant changes in phosphate metabolites in KTCTL-2 cells. Metabolomic 31P MRS detects different metabolite profiles for RCC cell lines with different MDR phenotypes and may be useful for noninvasive characterization of tumors in a clinical setting.This revised version was published online in August 2005 with a corrected sequence of authors. 相似文献

Ba0.5Sr0.5Co0.8Fe0.2O3‐δ tubes, capillaries, capillary modules, and asymmetric membranes were prepared and tested for oxygen permeation in a dead‐end vacuum operation mode at temperatures up to 850°C. The capillary module was built up by reactive air brazing using seven capillaries and a supply tube. Two machined discs were used as an end cap and as a connector plate. The oxygen permeation behaves according to Wagner at small driving forces, but significant negative deviations were observed for asymmetric membranes and single capillaries at higher ones. This is caused by pressure drops at the vacuum side for single capillaries. The highest oxygen flux was revealed for the capillary module with 175.5 mL(STP)/min at a low‐vacuum pressure of 0.042 bar at 850°C, but the asymmetric membrane showing a little bit higher flux at moderate vacuum pressures above 0.07 bar. © 2012 American Institute of Chemical Engineers AIChE J, 58: 3195–3202, 2012 相似文献

In the 1980s, Bennett introduced computational depth as a formal measure of the amount of computational history that is evident in an object's structure. In particular, Bennett identified the classes of weakly deep and strongly deep sequences and showed that the halting problem is strongly deep. Juedes, Lathrop, and Lutz subsequently extended this result by defining the class of weakly useful sequences and proving that every weakly useful sequence is strongly deep. The present paper investigates refinements of Bennett's notions of weak and strong depth, called recursively weak depth (introduced by Fenner, Lutz, and Mayordomo) and recursively strong depth (introduced here). It is argued that these refinements naturally capture Bennett's idea that deep objects are those which “contain internal evidence of a nontrivial causal history.” The fundamental properties of recursive computational depth are developed, and it is shown that the recursively weakly (respectively, strongly) deep sequences form a proper subclass of the class of weakly (respectively, strongly) deep sequences. The above-mentioned theorem of Juedes, Lathrop, and Lutz is then strengthened by proving that every weakly useful sequence is recursively strongly deep. We apply the general framework developed by John et al. in Computing 64:307–321, 2000 to analyze the convergence of multi-level methods for mixed finite element discretizations of the generalized Stokes problem using the Scott–Vogelius element. The Scott–Vogelius element seems to be promising since discretely divergence-free functions are divergence-free pointwise. However, to satisfy the Ladyzhenskaya–Babu?ka–Brezzi stability condition, we have to deal in the multi-grid analysis with non-nested families of meshes which are derived from nested macro element triangulations. Additionally, the analysis takes into account an optional symmetric stabilization operator which suppresses spurious oscillations of the velocity provoked by a dominant reaction term. Usually, the generalized Stokes problems appears in semi-implicit splitting schemes for the unsteady Navier–Stokes equations, but the symmetric part of a stabilized discrete Oseen problem can be reguarded as a discrete generalized Stokes problem likewise. 相似文献
