Characterization of single-nucleotide polymorphisms is a major focus of current genomics research. We demonstrate the discrimination of DNA mismatches using an elegantly simple microcantilever-based optical deflection assay, without the need for external labeling. Gold-coated silicon AFM cantilevers were functionalized with thiolated 20- or 25-mer probe DNA oligonucleotides and exposed to target oligonucleotides of varying sequence in static and flow conditions. Hybridization of 10-mer complementary target oligonucleotides resulted in net positive deflection, while hybridization with targets containing one or two internal mismatches resulted in net negative deflection. Mismatched targets produced a stable and measurable signal when only a four-base pair stretch was complementary to the probe sequence. This technique is readily adaptable to a high-throughput array format and provides a distinct positive/negative signal for easy interpretation of oligonucleotide hybridization.  相似文献

Two processing routes, namely sonochemical and microemulsion have been exploited to prepare bismuth ferrite powders. Phase pure nanosized BiFeO3 powders are prepared by sonochemical and microemulsion techniques in the temperature range of 400 °C and at 500 °C which is much lower than that of the conventional preparation methods. The XRD obtained from both cases was likely to be pure bismuth ferrite. Though the microemulsion based powders showed slightly better properties with respect to particle size, surface area, and final density, sonochemically prepared powders may be more acceptable considering the ease of preparation and cost effectiveness.  相似文献

The present study highlights application of Taguchi’s robust design coupled with fuzzy based desirability function approach for optimizing multiple bead geometry parameters of submerged arc weldment. Fuzzy inference system has been adapted to avoid uncertainly, imprecision and vagueness in experimentation as well as in data analysis by traditional Taguchi based optimization approach. Detailed methodology and unique features of the proposed method has been highlighted through a case study. The said approach can efficiently be used in off-line quality control of any production process as well as automation of the process.  相似文献

OBJECTIVE: The aim of this study was to demonstrate the MR characteristics of non-Hodgkin lymphoma of the skull base to help in the differential diagnosis of this neoplasm from other conditions. MATERIALS AND METHODS: MR of five patients, 7-64 years old, with pathologically proved lymphomas of the skull base were reviewed. Three cases had primary skull base lesions involving the sphenoid bone and the cavernous sinus. One case with a nasal cavity lesion involving the skull base and one with a relapsing skull base lesion of previously treated tonsillar lymphoma were included. RESULTS: The lesions had signal intensities that were similar to that of gray matter of brain on both T1- and T2-weighted imaging. Bilateral cavernous sinuses were involved with encasement of internal carotid arteries in every case. Postcontrast MR showed homogeneous enhancement of the tumor with dural infiltration along the planum sphenoidale, clivus, or tentorium. The clivus was destroyed or replaced by tumors in adult cases but in two children the clivus was preserved with intact sphenooccipital synchondrosis. In one case the tumor extended to the extracranial portion through the jugular foramen. CONCLUSION: The MR findings of a permeative lesion of the skull base, invasion of the cavernous sinus without arterial narrowing, infiltration along the dural surface, and an iso- or hypointensity with brain on T2-weighted imaging should suggest lymphoma.  相似文献

A detailed understanding of the mechanism of unfolding, aggregation, and associated rheological changes is developed in this study for β-Lactoglobulin at different pH values through concomitant measurements utilizing dynamic light scattering (DLS), optical microrheology, Raman spectroscopy, and differential scanning calorimetry (DSC). The diffusion interaction parameter kD emerges as an accurate predictor of colloidal stability for this protein consistent with observed aggregation trends and rheology. Drastic aggregation and gelation were observed at pH 5.5. Under this condition, the protein’s secondary and tertiary structures changed simultaneously. At higher pH (7.0 and 8.5), oligomerizaton with no gel formation occurred. For these solutions, tertiary structure and secondary structure transitions were sequential. The low frequency Raman data, which is a good indicator of hydrogen bonding and structuring in water, has been shown to exhibit a strong correlation with the rheological evolution with temperature. This study has, for the first time, demonstrated that this low frequency Raman data, in conjunction with the DSC endotherm, can be been utilized to deconvolve protein unfolding and aggregation/gelation. These findings can have important implications for the development of protein-based biotherapeutics, where the formulation viscosity, aggregation, and stability strongly affects efficacy or in foods where protein structuring is critical for functional and sensory performance.  相似文献

Ultraviolet protection factor (UPF) of fabrics made of 100% cotton and 100% bamboo viscose yarns were studied and a comparative analysis carried out using curve fitting technique. Bamboo viscose fabrics showed higher shrinkage, cover percentage, areal density and UPF compared to its cotton counterpart woven with identical yarn counts and fabric sett. However, the predictive model of cotton fabric UPF using fabric areal density as the input was able to estimate the UPF of bamboo viscose fabrics with very good accuracy. Furthermore, the 100% cotton and 100% bamboo viscose fabrics showed the same UPF if their cover percentage and areal density is similar. It is inferred from the analysis that the apparently higher UPF of bamboo viscose fabrics can be attributed to their higher cover percentage and areal density instead of bamboo’s inherent UV protective property which has been claimed in various literatures.  相似文献

Light amplification by stimulated emission of radiation (laser) is a coherent and monochromatic beam of electromagnetic radiation that can propagate in a straight line with negligible divergence and occur in a wide range of wave-length, energy/power and beam-modes/configurations. As a result, lasers find wide applications in the mundane to the most sophisticated devices, in commercial to purely scientific purposes, and in life-saving as well as life-threatening causes. In the present contribution, we provide an overview of the application of lasers for material processing. The processes covered are broadly divided into four major categories; namely, laser-assisted forming, joining, machining and surface engineering. Apart from briefly introducing the fundamentals of these operations, we present an updated review of the relevant literature to highlight the recent advances and open questions. We begin our discussion with the general applications of lasers, fundamentals of laser-matter interaction and classification of laser material processing. A major part of the discussion focuses on laser surface engineering that has attracted a good deal of attention from the scientific community for its technological significance and scientific challenges. In this regard, a special mention is made about laser surface vitrification or amorphization that remains a very attractive but unaccomplished proposition. The present study concerns development of a hard in situ boride-dispersed composite layer on the surface of AISI 304 stainless steel substrate to improve the wear resistance property. Laser processing was carried out by melting the surface of sand-blasted AISI 304 stainless steel substrate using a continuous wave CO2 laser and simultaneous deposition of a mixture of K2TiF6 (potassium titanium hexafluoride) and KBF6 (potassium hexafloroborate) (in the weight ratio of 2:1) using Ar as shrouding environment. Powder feed rate was maintained constant at 4 g/min. Irradiation results in dissociation of a pre-deposited mixture along with a part of the stainless steel substrate, intermixing and rapid solidification to form the composite layer on the surface. The micro-structure of composite layer consists of dispersion of titanium boride particles in AISI 304 stainless steel matrix. Volume fraction of particles is found to be uniform throughout the composite layer, though varied with laser parameters. The micro-hardness of the surface was improved 250–350 VHN as compared to 220 VHN of the AISI 304 stainless steel substrate with a significant improvement in wear resistance property. The mechanism of wear was found to be a combination of adhesive and abrasive in as-received stainless steel. However, it was predominantly abrasive for laser composite surfaced stainless steel.  相似文献

Chronic caloric restriction has been shown to inhibit mammary tumor promotion in the 7,12-dimethyl-benz[a]anthracene (DMBA) rat mammary tumor model. The objectives of this study were to determine (i) the effects of chronic caloric cycling (yo-yo dieting) on mammary tumor promotion by high fat diets and (ii) the effect of three dietary regimens +/- superimposed mammary tumor burden on plasma endothelin-1,2 (ET) levels. Female Sprague-Dawley rats were treated with DMBA (5 mg/rat) and divided into three dietary groups: ad libitum (AL) (containing 15% corn oil); 40% calorie restricted (CR) (containing 20% corn oil so consumption of fat was equivalent between AL and CR); a calorie cycled (CC) group fed alternatively AL and CR diets each 48 h period. After 10 weeks, tumor incidences were: AL, 63%; CR, 27%; CC, 57% (AL versus CR, P < 0.05; CC versus CR, P < 0.05; AL versus CC, NSD). ET levels (pg/ml plasma) were: AL, 16.0 +/- 6.54; CR, 32.31 +/- 0.34; CC, 23.44 +/- 5.04 (AL versus CR, P < 0.01; CC versus CR, P < 0.01; AL versus CC, P < 0.05). Plasma ET levels were independent of tumor incidence and tumor burden, but plasma ET levels were significantly increased in rats with a prior history of calorie restriction. As expected, maintained caloric restriction reduced mammary tumor incidence but intermittent caloric restriction (caloric cycling or yo-yo dieting) was without similar benefit.  相似文献
