Several reasons for the use of multidisciplinary teams composed of individuals with natural science and engineering background in problem‐solving processes exist. The most important are the integration of science‐based technologies into products and processes, and benefits for the problem‐solving process thanks to new knowledge and new perspectives on problems. In this study we analyse the implications of interdisciplinary (science – engineering) group problem solving from a managerial as well as from a cognitive perspective. We then report on an experiment investigating the impact of problem‐relevant disciplinary group composition and methodological support on the problem‐solving process and its outcome. The findings of the experiment have managerial, theoretical, and pedagogical implications related to early phases of New Product/Process Design processes in high‐technology and scientific knowledge‐related domains. 相似文献

Many e-commerce sites present additional item recommendations to their visitors while they navigate the site, and ample evidence exists that such recommendations are valuable for both customers and providers. Academic research often focuses on the capability of recommender systems to help users discover items they presumably do not know yet and which match their long-term preference profiles. In reality, however, recommendations can be helpful for customers also for other reasons, for example, when they remind them of items they were recently interested in or when they point site visitors to items that are currently discounted. In this work, we first adopt a systematic statistical approach to analyze what makes recommendations effective in practice and then propose ways of operationalizing these insights into novel recommendation algorithms. Our data analysis is based on log data of a large e-commerce site. It shows that various factors should be considered in parallel when selecting items for recommendation, including their match with the customer’s shopping interests in the previous sessions, the general popularity of the items in the last few days, as well as information about discounts. Based on these analyses we propose a novel algorithm that combines a neighborhood-based scheme with a deep neural network to predict the relevance of items for a given shopping session. 相似文献

Many enterprises that use expensive capital goods outsource the process of spare parts supply and stocking to a maintenance-, repair-, and overhaul provider (MRO-provider). Because of the high value, these spare parts are stocked in repairable item systems, in which defective parts are repaired after removal from the machine and put back into stock. When setting inventory levels, the providers do not consider all of the spare parts that are stocked in the system because customers hold own stock to bridge the delivery time from the MRO-provider. As these spare parts are not property of the MRO-provider, the provider does not take them into account when planning inventory, incurring overstocking at the central depot. A model, which considers customer stock when determining depot inventory levels, is developed by extending the METRIC model with a heuristics. The heuristics calculates backorder levels according to the depot inventory levels as well as customer stock levels and allows for an inventory reduction. Using the proposed model, savings of depot inventory can be up to 100 %, while keeping the backorder level of part requests below a preset threshold. 相似文献

The application of different methods for a microstructural analysis of functional catalysts is reported for the example of different Cu/ZnO-based methanol synthesis catalysts. Transmission electron microscopy and diffraction were used as complementary techniques to extract information on the size and the defect concentration of the Cu nano-crystallites. The results, strengths and limitations of the two techniques and of different evaluation methods for line profile analysis of diffraction data including Rietveld-refinement, Scherrer- and (modified) Williamson–Hall-analyses, single peak deconvolution and whole powder pattern modeling are compared and critically discussed. It was found that in comparison with a macrocrystalline pure Cu sample, the catalysts were not only characterized by a smaller crystallite size, but also by a high concentration of lattice defects, in particular stacking faults. Neutron diffraction was introduced as a valuable tool for such analysis, because of the larger number of higher-order diffraction peaks that can be detected with this method. Tumor growth and survival requires a particularly effective immunosuppressant tumor microenvironment (TME) to escape destruction by the immune system. While immunosuppressive checkpoint markers like programmed cell death 1 ligand (PD-L1) are already being targeted in clinical practice, lymphocyte-activation-protein 3 (LAG-3), T-cell immunoglobulin and mucin-domain containing-3 (TIM-3) and V-domain Ig suppressor of T cell activation (VISTA) inhibitors are currently under investigation in clinical trials. Reliable findings on the expression status of those immune checkpoint inhibitors on tumor-infiltrating lymphocytes (TILs) in the TME of oropharyngeal squamous cell carcinoma (OPSCC) are lacking. This work aims to describe the expression of LAG-3, TIM-3, and VISTA expression in the TME of OPSCC. We created a tissue microarray of paraffin-embedded tumor tissue of 241 OPSCC. Expression of the immune checkpoint protein LAG-3, TIM-3, and VISTA in OPSCC was evaluated using immunohistochemistry and results were correlated with CD8+ T-cell inflammation and human papillomavirus (HPV)-status. 73 OPSCC stained positive for LAG-3 (31%; HPV+:44%; HPV-:26%, p = 0.006), 122 OPSCC stained positive for TIM-3 (51%; HPV+:70%; HPV-:44%, p < 0.001) and 168 OPSCC (70%; HPV+:75%; HPV-:68%, p = 0.313) for VISTA. CD8+ T-cells were significantly associated with LAG-3, TIM-3 and VISTA expression (p < 0.001, p < 0.001, p = 0.007). Immune checkpoint therapy targeting LAG-3, TIM-3, and/or VISTA could be a promising treatment strategy especially in HPV-related OPSCC. Future clinical trials investigating the efficacy of a checkpoint blockade in consideration of LAG-3, TIM-3, and VISTA expression are required. 相似文献

Spirooxindole-1,3-oxazines are a small and structurally unique class of spirooxindole alkaloids. To date, only four of these compounds have been isolated from natural sources, and their biological properties remained unknown thus far. Dioxyreserpine is a synthetic spirooxindole-1,3-oxazine, that can readily be prepared from the Rauvolfia alkaloid (–)-reserpine by catalytic photooxygenation. While dioxyreserpine itself was now identified as a moderately effective antitumoral agent, structurally modified analogs of it emerged as a new class of highly potent and selective growth inhibitors of various human cancers, including pancreatic cancers. Systematic structural optimization ultimately led to an inhibitor displaying low-micromolar IC50-values against six cancer cell lines as well as selective apoptosis induction in vitro. 相似文献

Posttraumatic epilepsy (PTE) is a major public health concern and strongly contributes to human epilepsy cases worldwide. However, an effective treatment and prevention remains a matter of intense research. The present study provides new insights into the gamma aminobutyric acid A (GABAA)-stabilizing protein ubiquilin-1 (ubqln1) and its regulation in mouse models of traumatic brain injury (TBI) and in vitro epilepsy. We performed label-free quantification on isolated cortical GABAergic interneurons from GAD67-GFP mice that received unilateral TBI and discovered reduced expression of ubqln1 24 h post-TBI. To investigate the link between this regulation and the development of epileptiform activity, we further studied ubqln1 expression in hippocampal and cortical slices. Epileptiform events were evoked pharmacologically in acute brain slices by administration of picrotoxin (PTX, 50 μM) and kainic acid (KA, 500 nM) and recorded in the hippocampal CA1 subfield using Multi-electrode Arrays (MEA). Interestingly, quantitative Western blots revealed significant decreases in ubqln1 expression 1–7 h after seizure induction that could be restored by application of the non-selective monoamine oxidase inhibitor nialamide (NM, 10 μM). In picrotoxin-dependent dose–response relationships, NM administration alleviated the frequency and peak amplitude of seizure-like events (SLEs). These findings indicate a role of the monoamine transmitter systems and ubqln1 for cortical network activity during posttraumatic epileptogenesis. 相似文献

The growing demand for electro mobility together with advancing concepts for renewable energy as primary power sources requires sophisticated methods of energy storage. In this work, we present a Li ion battery based on Si nanowires, which can be produced reliable and cheaply and which shows superior properties, such as a largely increased capacity and cycle stability. The crack growth resistance behavior of polycrystalline Pb(Zr1?xTix)O3 has been characterized in a novel experimental arrangement between 24 and 140 °C. Experimental measurements were carried out on compact tension specimens submerged in a temperature-controlled silicone oil bath. The results show a decrease in the observed shielding toughness, leading to an overall reduction in the maximum toughness. The temperature-dependent stress–strain behavior and elastic properties were characterized, providing an insight into the effect of the changing ferroelastic properties on the temperature-dependent fracture behavior. 相似文献
