DNA aptamer-based bioanalysis of IgE by fluorescence anisotropy   总被引:3,自引:0,他引:3  
A rapid, homogeneous aptamer-based bioanalysis is reported for the sensitive detection of immunoglobulin E (IgE) using fluorescence polarization (FP). 5'-End-labeled D17.4 DNA aptamer was used for IgE detection based on the anisotropy differences of the labeled ligand. Two different fluorophores, fluorescein and Texas Red, were used to analyze IgE in the low-nanomolar range with high specificity. Measurable anisotropy changes were observed with a short equilibration time. Analysis of the binding data reveals a possible cooperative binding process in solution. The nature of the fluorophore clearly influences the sensitivity of the analysis more than the tether length used for the dye conjugation. The local fluorophore motion is seen to influence the sensitivity of the FP probe significantly. Texas Red is seen to be relatively more sensitive for this approach and has apparently favorable dye-DNA interactions, and a limit of detection of 350 pM was obtained. Significant temperature dependence of the FP responses has been observed in this work. Ionic composition of the binding buffer also influences the assay sensitivity. The results confirm the promise and potential of similar homogeneous assays for aptamer-based bioanalysis.  相似文献

Cotton yarns covered with hard (barium hexaferrite – BaFe), and soft (Black Toner 6745 CP-313) magnetic particles are investigated for their application in the field of functional textile. Our results reveal a rather consistent dependence of the magnetic yarn properties on the fabrication parameters (nature of the magnetic powder, composition of the coating solution, etc.). The magnetic properties (residual magnetism and coercive field intensity) of the yarns are increasing with the magnetic powder content in the coating solution. However, this is at some point leading to the degradation of some yarn properties, e.g. elasticity. Bending tests revealed that several factors improve the bending behavior of the yarns and reduce cracks. For example, a higher number of cracks were exhibited by the hard magnetic yarns compared to the soft ones. The presence of polyurethane in the coating solution leads to no cracks on the surface of hard magnetic yarns or a small number in case of soft magnetic yarns. The tensile properties of the magnetic yarns are increasing with increasing magnetic powder content. Over a certain mass percentage (45% for BaFe and 30% for CP-313), the magnetic yarns tensile strength and elongation become lower than those of the support yarn.  相似文献

BACKGROUND: In this study the effects of pulsed electric field (PEF) pretreatment were evaluated during drying and rehydration of carrots. Carrots pretreated with an electric field intensity of 1 kV cm?1 (capacitance 0.5 µF, 20 pulses) or 1.5 kV cm?1 (capacitance 1 µF, 20 pulses) as well as blanched (100 °C, 3 min) carrots were used for the study. Following pretreatment, samples were oven dried at 70 °C and then rehydrated in distilled water (1:30 w/v) at room temperature (24 ± 1 °C). RESULTS: PEF pretreatment increased the drying rate of carrots. However, the rehydration rate of PEF‐pretreated carrots was lower than that of blanched carrots. There were no colour differences between PEF‐pretreated and blanched carrots before drying and after rehydration. In terms of texture, PEF‐pretreated carrots were firmer than blanched carrots. PEF pretreatment reduced the activity of peroxidase by 30–50%, while blanching completely inactivated the enzyme (>95%). CONCLUSIONS: Overall, the results suggest that PEF could be an effective pretreatment during drying and rehydration of carrots. Copyright © 2009 Society of Chemical Industry  相似文献

Omega-3 oil from fish can be stabilised against oxidation using a variety of microencapsulation technologies. Complex coacervation has been used and found to be commercially useful for fortifying foods and beverages with long-chain omega-3 containing oils. Here we report a comparative human bioavailability study of microencapsulated omega-3 fish oil and standard fish-oil soft-gel capsules. Phospholipid levels of long-chain omega-3 fatty acids increased equivalently in both subjects groups. Also, triacylglycerol levels were reduced similarly in both groups. These results indicate that omega-3 fatty acids have equivalent bioavailability when delivered as microencapsulated complex coacervates or as soft-gel capsules.  相似文献

通过常压等离子体处理进行PVA失重、PVA浆料在水中溶解率的变化、等离子体处理与常规H2O2退浆比较分析以及PVA薄膜的XPS和FT-IR分析,讨论常压等离子体对PVA作用的机理。结果表明,常压等离子体作用使部分PVA大分子氧化成小分子如CO2、H2O而直接消失在空气中,另一部分PVA大分子被氧化降解成分子链较短的分子,提高了其在水中的溶胀、溶解性。  相似文献

This article presents in closed form new influence functions for displacements and stresses to a boundary value problems (BVP) of thermoelasticity within a strip, caused by a unit point heat source. We also obtain the respective new integration formula of Green’s type, which directly determines the thermal stresses in the form of integrals of the products between specified internal heat sources, temperature, or heat flux prescribed on boundary and derived thermoelastic influence functions (kernels). The general Green’s type integral formula permits to derive new solution to one particular BVP of thermoelasticity for a strip in the form of elementary functions. Graphical representation of thermal stresses, created by an internal point heat source and by a boundary temperature, is included.  相似文献

Resource sharing among co-located firms--referenced in the industrial ecology literature as "industrial symbiosis"--engages traditionally separate industries in a collective approach to business and environmental management involving the physical exchanges of materials, energy, water, and byproducts. While industrial symbiosis is seen hypothetically as a win-win situation, there are few analyses of the economic and environmental consequences for the individual participants in multi-faceted exchanges. In this article, the nascent industrial symbiosis network in Guayama, Puerto Rico, is explored from environmental, economic, and regulatory perspectives of the individual participants and the community. A coal-fired power plant, owned and operated by the AES Corporation, draws five million gallons per day of process water from nearby sources thus avoiding freshwater withdrawals and, through steam sales, significantly reduces emissions from a nearby refinery. This article quantifies economic and environmental costs and benefits for the symbiosis participants, concluding that there are substantial benefits to engaging in symbiosis, although these benefits fall unevenly on participating organizations. Policy intervention can be a viable means of motivating more regular occurrences of resource exchanges among groups of firms.  相似文献

Preharvest contamination of field crops may have many sources, including feces, soil, and irrigation water. In March 2000, a sewage spill released unchlorinated tertiary-treated effluent into a creek used to irrigate commercial produce. A field of young cabbage transplants was irrigated with creek water as the contaminated water flowed past this land. Cabbage samples were taken from plots within this field, and Escherichia coli was isolated from the roots of these plants but not from the edible portion of the cabbage. No E. coli was isolated from water samples or from control samples taken from a nearby cabbage field watered with chlorinated municipal water. The cabbage field under study had not been fertilized with manure for at least 2 years prior to the contamination incident. Six different E. coli serotypes were identified, although none of them proved to be pathogenic. These serotypes were separated into five groups by a RiboPrinter; the resulting groups correlated well with the serotypes and the locations in the field from which these strains were isolated. We previously found that certain nonpathogenic E. coli strains displayed lower levels of adherence to lettuce seedling roots in a hydroponic adherence assay. The E. coli field strains displayed variable patterns of adherence to lettuce seedlings: strain MW421 showed significantly lower root and shoot adherence levels than did the other field strains, while strains MW423 and MW425 showed significantly higher root and shoot adherence levels. These data suggest that water quality is of paramount importance for the food safety of growing crops.  相似文献
