A quadratic cost functional for reconstruction of a high-resolution wave front from a coarse wave front is presented. The functional uses as data the position and the direction of the coarse wave front that had previously been computed with a ray-tracing method. This functional uses an optical relationship between the ray information and the wave front's shape to reconstruct a high-density wave front. The performance of the proposed functional is illustrated by reconstruction of complicated wave fronts for which this functional has an accuracy that is superior to that of conventional interpolation methods.  相似文献

Laser-induced fluorescence of the CH radical is used to determine the flame-front temperature of an 8-Torr premixed CH(4)/O(2) flame. The A (2)D-X (2)Delta (0, 0) and B (2)Sigma- - X (2)II (0, 0) bands give values of 1960 +/- 60 and 1920 +/- 70 K, respectively. This is an improvement over a previous study that found discrepancies up to 20% between these bands. New rotational-level-dependent transition probabilities are the main reason for this improvement. The weaker off-diagonal bands A-X (0, 1) and B-X (0, 1) yield temperatures of 1930 +/- 90 and 1830 +/- 100 K, respectively. The influence of rotational transfer on the predissociated levels that have N' > in B (2)Sigma-, v'= 0 was investigated with fluorescence scans and a statistical power-gap law model of the relaxation; deviations up to 8% in temperature can occur because of this process.  相似文献

The differential pulse polarography has been utilized to carry out the study of the catalytic hydrogen currents in which the 2-benzoylpyridine oximecobalt ion system acts as catalyst. The current is produced by species in adsorbed state as well as in solution, as the extensive study performed has shown, which has also allowed us to attain data to propose a possible mechanism by which the electrodic reaction takes place. The purpose of this research was to show that controlled centrifugal instabilities-Dean vortices-produced by solutions and suspensions from typical biotechnology applications flowing through curved tubes can be used to reduce concentration polarization and/or fouling in pressure-driven ultrafiltration (UF) and microfiltration (MF) processes. Experiments were conducted to (i) evaluate the ultrafiltration performance of hollow fiber membranes in linear and helical configurations with dextran (low fouling) and bovine serum albumin (high fouling) solutions and (ii) compare the performance of linear and helical coiled UF hollow fiber modules with that of similar MF modules using baker's and beer yeast (Saccharomyces cerevisiae) suspensions as feed. Both constant transmembrane pressure (TMP) and constant permeation flux (J) experiments were utilized here. The membrane material was polyether sulfone. For the ultrafiltration experiments, the helical module performed consistently better than the linear module with dextran T500 and BSA solutions, resulting in performance improvements (helical versus linear) from 20 to 200% and up to 85%, respectively. For the comparative experiments between UF and MF, the helical module again performed better than the linear module for low concentration baker's yeast suspensions (0.5-1% dry wt). At constant TMP, the flux improvements for UF were 30-120%, while at constant J, the capacity or loading was 4.5 times higher for the UF as compared to the MF membrane. At high beer yeast concentrations (5.1-6.8% dry wt), although flux improvements were not observed between the linear and helical modules for UF, the UF fluxes were 72% higher than that obtained with MF. Also, for MF, with the same high beer yeast concentrations, the helical module exhibited 30-90% higher fluxes than that obtained with the linear module. At constant flux (117-137 L m-2 h-1) and intermediate baker's yeast concentrations (0.65-2.7% dry wt), 10-20 times the capacity was obtained for the helical over the linear module. Yeast cells were the dominant foulant. For constant UF flux (70 L m-2 h-1) experiments at high beer yeast concentrations ((4.3-7.7) x 10(7) cells/mL or 5.1-6.8% dry wt), the capacity (loading) for the helical module was 10 times that of the linear module. Again, the yeast cells were the dominant foulant. A new mass-transfer correlation for ultrafiltration of dextran T500 solutions for laminar flow in a helical hollow fiber module was obtained, viz. The modified fluorescence method was used to determine the accumulation of norfloxacin by Mycobacterium aurum A+ and Mycobacterium smegmatis mc(2)155. By using an exogenous norfloxacin concentration of 10 microg/ml, a steady-state concentration (SSC) of 160 to 180 ng of norfloxacin/mg of cells was obtained for M. aurum, and an SSC of 120 to 140 ng of norfloxacin/mg of cells obtained for M. smegmatis. For both species of mycobacteria, the SSC was achieved within 5 min. The silicon oil method was investigated and gave higher SSCs than the modified fluorescence method. Further studies on the mechanism of norfloxacin accumulation by M. aurum were performed. An increase in the pH of the wash buffer from 7.0 to 9.0 did not significantly affect the final SSC obtained. Accumulation was nonsaturated over a norfloxacin concentration range of 0 to 100 microg/ml, and the proton motive force inhibitor 2,4-dinitrophenol (1 and 2 mM), whether it was added before or after norfloxacin was added, had no effect on the final SSC obtained. 2,4-Dinitrophenol also had no effect on norfloxacin accumulation by M. smegmatis. Furthermore, norfloxacin accumulation by M. aurum was unaffected by the presence of either Tween 80 or subinhibitory concentrations of ethambutol in the growth medium. Therefore, it is proposed that norfloxacin accumulation by mycobacteria occurs by simple, energy-independent diffusion.  相似文献

The importance of the antioxidant enzyme superoxide dismutase (CuZnSOD) in the metabolic switch from normotrophic to methylotrophic conditions was studied in the facultative methylotrophic yeast Candida boidinii. Copper adaptation was performed to qualify C. boidinii as a suitable cellular system to study the effect of induction of CuZnSOD, and other biochemical components along the copper detoxification system, on methanol adaptation. Copper adaptation results in the induction of CuZnSOD peroxidase activity as well as of glutathione. The effects at the metabolic level of exposure to both copper and methanol were also studied: the results suggest that the effect on antioxidant enzyme levels as a function of the change of trophic condition are predominant with respect to the effects of copper administration. Thus, the methanol-dependent induction of such enzymes is likely to provide a sufficient protection for the cells against toxic effects depending on copper administration. Administration of copper under methylotrophic conditions decreases the growth rate in spite of the high levels of antioxidant enzymes that are elicited by copper treatment. The adaptation to methanol metabolism was studied alsoafter methanol-independent induction of CuZnSOD, glutathione and catalase levels, obtained by exposure to high copper concentrations in glucose-containing medium. The metabolic changes induced by copper are persistent over several re-inoculations in normo-cupric glucose medium, thus allowing the study of the glucose-to-methanol switch on cells exhibiting high levels of antioxidant enzyme activities. Under such conditions the lag time observed during the transition from normotrophic to methylotrophic conditions is strongly reduced.  相似文献
