Geotrichum candidum is a ubiquitous filamentous yeast-like fungus commonly isolated from soil, air, water, milk, silage, plant tissues, digestive tract in humans and other mammals. This species is widely used as adjunct culture in the maturation of cheese. The genus Geotrichum is composed of 18 species. A recent taxonomic revision concluded that the old Galactomyces geotrichum/G. candidum complex contained four separate species of which Galactomyces candidus sp. nov./G. candidum. M13 primer can be used for identifying species of the Geotrichum genus. Used in combination, RAPD-PCR and RAM-PCR permit strains to be differentiated. The species can be unambiguous differentiated from the two species most frequently described in human pathology: Geotrichum clavatum (reclassified Saprochaete clavata) and Geotrichum capitatum (reclassified Magnusiomyces capitatus/Saprochaete capitata). Sources of exposure are food ingestion--cheese consumption playing a major role--inhalation and contact. A bibliographic survey was conducted to assess corresponding hazards and risks. G. candidum infections (mainly pulmonary or bronchopulmonary, but also cutaneous, oral, disseminates) are very rare: fewer than 100 cases reported between 1842 and 2006. Moreover, cases were not all confirmed by repeated isolations and demonstration of the fungus' presence in tissues, a prerequisite to establish a true diagnosis of geotrichosis. Immunocompromised population was recently shown as a target for opportunistic infection. The most effective treatments include either azole drogs as ketonazole, iconazole and clotrimazole, or polyene antibiotics as amphotericin B, nystatin and pimaricin, or voriconazole-amphotericin B association. Less than 1 case/year of disease was possibly caused by G. candidum and it never included dairy products or foodborne infection. The risk of developing an infection due to G. candidum in connection with its technological use and consumption of dairy products is virtually nil. Purpose
This study evaluated the effects of weathering (1008 h) and thickness (1 and 3.5 mm) on the superficial microhardness of the ocular button, using five colorless acrylic resins, and colorless buttons specific for ocular prosthesis.Materials and methods
One-hundred-and-twenty specimens were made and distributed allocated to 12 groups. Microhardness analysis was carried out with a microdurometer (Shimadzu, Japan) before and after weathering for 1008 h. Data were then analyzed statistically by ANOVA and the Tukey Test at 1% significance.Results
The data demonstrated that the ocular button and the QC 20 resin presented, respectively, the highest (22.9) and lowest (18.31) values of surface microhardness (Knoop), independently of the variable evaluated.Conclusion

Effects of tactile feedback on movement accuracy and speed were studied. Younger and older participants performed three tasks (1, select and drag word; 2, menu navigation; 3, select and drag cell) using commercial software and a mouse with or without tactile feedback. Task time and error number were recorded. Tasks were divided according to presence or absence of tactile feedback, and participants were divided into subgroups (high, average, low) based on Task 1 performance. Overall, older participants took longer (p < .0001) and made more errors (p < .001) than younger participants. There was an effect of feedback by task in younger participants for all six outcomes (p < 0.02). At the task level, with feedback, younger participants reduced performance time (13%) and errors (24%) on Task 1. Low- and average-performance younger participants benefited most from feedback for Task 1. Older low-performance participants also benefited from feedback for Task 1. For Task 3, older participants tended to take more time and make more errors with feedback. Tactile feedback may enhance performance when feedback is event related. Older people may not integrate sensation as well as younger individuals to enhance performance. Geotrichum candidum is an ascomycetous anamorph yeast-like fungus found in various habitats. It is a component of the natural flora of milk and is used as a maturing agent for both soft and hard cheeses. This microorganism displays phenotypic variability and may act as an opportunist pathogen, causing geotrichosis. Cytological analysis of G. candidum strain ATCC 204307 showed this strain to have eight chromosomes. We prepared chromosomal DNA from 13 strains of G. candidum differing in habitat and morphotype. We used pulsed field gel electrophoresis (PFGE) in two sets of conditions to determine the size of the chromosomal DNA molecules. The strains investigated had five to eight chromosomes, 0.6 to 4.5 Mb in size. We estimated genome size in these 13 strains to be between 11 and 19 Mb. Pulsed-field gel electrophoresis profiles showed a high degree of polymorphism, indicating considerable variability between strains. Genome size and the presence of large chromosomes appeared to be correlated with morphotype. Strains with a mold-like or intermediate morphotype tended to have larger genomes than strains with a yeast-like morphotype did. 相似文献

Pichia anomala (strain K) is an efficient biocontrol agent against post-harvest diseases affecting apples. To study the role of strain K genes in biocontrol activity, it is useful to identify selectable markers on which to base a gene disruption strategy. The Pichia anomala TRP1 gene (PaTRP1) was isolated by complementation of the multi-auxotrophic S. cerevisiae strain FY-1679-18b. DNA sequence analysis revealed the presence of a 699 bp ORF encoding a 233 amino acid protein showing the typical conserved structure of proteins of the phosphoribosyl anthranilate isomerase (PRAI) family. Codon analysis revealed a high number of unused codons. Downstream from PaTRP1 was found the 3' extremity of a gene highly similar to the IPP1 gene (coding for the inorganic pyrophosphatase). 相似文献

A test meal (300 mg casein, 600 mg sucrose, 100 mg corn oil, tracer dose of 9.103H oleic acid) was given to fasting adult rats with intestinal lymph fistulas. One group received an acute oral dose of ethanol
(3.2 g/kg body weight) simultaneously with the test meal. Controls received 2.5 ml of water instead of ethanol. Ingestion
of ethanol temporarily delayed the removal of lipid radioactivity from the stomachs. More than 25% of radioactivity fed remained
8 hr after feeding whereas with control rats less than 10% of lipid radioactivity fed remained 6 hr after feeding. In controls
and ethanoltreated rats, the amounts of exogenous lipids in the intestinal lumen and mucosa were low and similar enough. Quantities
of endogenous and exogenous lipids found in the lymph collected during 24 hr after feeding were similar in the two groups,
but the fat absorption peak was found after 6 hr in alcoholic rats and before 6 hr in controls. This delay was probably due
to the retention of lipids in the stomach. More of the exogenous lipid was always transported by small particles moving in
the region of α1 globulins in cellulose acetate electrophoresis than by larger particles remaining at the origin. This proportion was enhanced
in the ethanoltreated animals. The larger fat particles were richer in endogenous fatty acids in alcohol-treated rats than
in controls.

Two footstep strategies associated with expert workers and two associated with novice workers were studied for lifting and lowering a 12 kg box. Kinematic variables, L5/S1 resultant moments and body postures of eight healthy male subjects were used to analyse the effects of the footstep strategies. A tridimensional dynamic rigid body model was used to estimate L5/S1 resultant moment, using inertial forces to estimate external forces; these procedures were validated with force platforms. Results showed that expert strategies either reduced the path of the subject's centre of gravity or reduced the asymmetry of posture at deposit. The footstep strategies showed no consistent effect on L5/S1 resultant moment. The use of different footstep strategies might be aimed at different goals, which may depend on the context. On the one hand, the reduction in the path of the subject's centre of gravity might represent a mean to reduce the effort required and avoid fatigue, which might be appropriate in some circumstances; on the other hand, the reduction of asymmetry of posture at deposit might be more appropriate in other circumstances. Footstep strategies will deserve further attention since this study showed that they can play an important role in reducing the risks of injury.
Relevance to industry
Manual materials handling is a major known source of industrial injury. Biomechanical studies of handling strategies really used by workers should help understand the loadings encountered in real handling tasks, which may lead to the identification of safer handling strategies. 相似文献

It is known that several factors can affect the clinical success and durability of fixed partial dentures. Therefore, the aim of the present study was to present a literature review about the longevity, clinical success, and quality, as well as, patients' and dental surgeons' satisfaction of ceramic fixed dentures. High rate of patients' satisfaction has been observed in relation to the esthetic of ceramic crowns. In addition, the literature has shown that dental ceramics can be used in several clinical situations with high success rate and longevity. Milk fat (MF) and rapeseed oil (RO) blends were analyzed by differential scanning calorimetry (DSC). It was shown that peak and onset temperatures can be used to determine the percentage of each fat in the blend and that the relative enthalpy of one peak assigned to low‐melting triacylglycerols (TAG) can also be used to determine the percentage of RO in the blend. A linear relation was also established between MF content of the blend and its dropping point (DP), indicating that DP can be linearly related with the above DSC data. A blend of MF/RO 70 : 30 (wt/wt) was then chosen as a model system for enzymatic interesterification (EIE). The applicability of DSC analyses to EIE products was checked and a correct correlation could be established between DSC values and the interesterification degree and DP. Among the data from the DSC profiles, the peak associated with low‐melting TAG was the best indicator of the reaction course. In the same way, a high‐melting MF stearin fraction was interesterified with RO. In that case, onset temperatures and peak “a” were better reaction indicators than for the interesterified MF/RO blend. We therefore suggest that values from DSC endotherms could be used to monitor EIE of fat blends. 相似文献
