This study evaluated the effectiveness of a large-scale job stress reduction program implemented in the Dutch domiciliary care sector. The employees of 81 organizations were interviewed twice (only nurses in executive jobs; total sample size exceeded 26,000). Organizations that implemented many interventions were expected to be more successful in reducing job stress than were other organizations. It was found that (a) levels of job stress decreased during the observed interval; (b) organizations with many suboptimal scores on selected work characteristics took, on average, more measures to reduce job stress than others; (c) organizations usually implemented a wide variety of measures; and (d) work-directed (but not other) interventions were linked to job stress reduction. Solid films of colloidal quantum dots show promise in the manufacture of photodetectors and solar cells. These devices require high yields of photogenerated charges and high carrier mobilities, which are difficult to achieve in quantum-dot films owing to a strong electron-hole interaction and quantum confinement. Here, we show that the quantum yield of photogenerated charges in strongly coupled PbSe quantum-dot films is unity over a large temperature range. At high photoexcitation density, a transition takes place from hopping between localized states to band-like transport. These strongly coupled quantum-dot films have electrical properties that approach those of crystalline bulk semiconductors, while retaining the size tunability and cheap processing properties of colloidal quantum dots.  相似文献

Application of in-situ microtomography to characterization of power law creep and creep damage in structural materials is presented. It is shown first that the successively reconstructed volumes are adequately monitoring the macroscopic sample shape and that microtomography is an optimal tool to characterize inhomogeneous specimen deformation. Based on a two-step image correlation technique the evolution of single voids is revealed and the basis of a pioneering approach to creep damage studies is presented. The method allows the unequivocal separation of three concurrent damage mechanisms: nucleation, growth, and coalescence of voids. The results indicate that growth rate of voids with equivalent diameters in the range of 2–5 mm is of about one order of magnitude higher than the prediction of continuum solid mechanics. Analysis of void coalescence points out the presence of two stable growth regimes related to coalescence between primary and secondary voids, respectively.  相似文献

Oil is ubiquitous in aquatic sediments and may affect partitioning and bioavailability of hydrophobic organic chemicals (HOCs). In contrast to other sedimentary hydrophobic carbon phases (natural organic matter, soot-like materials), oil residues have hardly received any attention as far as it concerns effects on HOC sorption. This paper describes experimental work dealing with such effects of oil on polycyclic aromatic hydrocarbon (PAH) sorption to sediments. Three different oils were spiked to a marine sediment in concentrations between 0 and 100 g/kg. Sediment-water distribution coefficients (Kd) for six deuterated PAHs were then determined either directly after spiking the oil or after a semi-natural weathering process in the lab (lasting for more than 2 yr). Resulting Kd values demonstrated sorption-reducing (competitive) effects at relatively low oil concentrations and sorption-enhancing effects at high oil concentrations. The latter effects only occurred above a certain threshold [i.e., ca. 15% (w/w) of oil on a sedimentary organic carbon basis] marking the oil concentration at which the hydrocarbon mixture presumably starts forming separate phases. Assuming a two-domain (organic carbon + oil) distribution model, oil-water distribution coefficients (K(oil)) for PAHs were estimated. For fresh oils, log K(oil) values appeared to be very similar for different types of oils, proportional to log K(OW) values and indistinguishable from log K(OC) values. For weathered oils, K(oil) values were also rather independent of the type of oil, but the affinity of low molecular weight PAHs for weathered oil residues appeared to be extremely high, even higher than values reported for most types of soot. Because affinities of high molecular weight PAHs for oils had not changed upon weathering, sorption of all PAHs studied (comprising a log K(OW) range of 4.6-6.9) to the weathered oil residues appeared to be more or less constant (averaged log K(oil) = 7.0 +/- 0.24). These results demonstrate that it is crucial to take the presence of oil and its weathering state into account when assessing the actual fate of PAHs in aquatic environments.  相似文献

Soils from former manufactured gas plant (MGP) sites are often heavily contaminated with polycyclic aromatic hydrocarbons (PAHs). Current risk assessment methods that rely on total PAH concentrations likely overstate adverse effects of such soils since bioavailability is ignored. In this study, solid-phase microextraction (SPME) was applied to estimate bioavailable PAH concentrations and toxicity in earthworms exposed to 15 MGP soils. In addition, PAH sorption to all soils (K0o values) was determined. The results showed a several orders of magnitude variation in Koc values, demonstrating that generic organic carbon-normalized sorption coefficients will typically be overconservative at MGP sites. SPME-predicted bioaccumulation generally was within a factor of 10 of measured bioaccumulation (in earthworm bioassays), in contrast to current risk assessment model estimates that overpredicted bioaccumulation 10-10 000 times. Furthermore, on the basis of estimated total body residues of narcotic PAHs, SPME correctly predicted worm mortality observed during bioassays in the majority of cases. For MGP sites where current risk assessment procedures indicate concerns, SPME thus provides a useful tool for performing a refined, site-specific assessment.  相似文献

In this study, transmission electron microscopy (TEM) and cryo-scanning electron microscopy (cryo-SEM) were evaluated for their ability to detect lipid bodies in microalgae. To do so, Phaeodactylum tricornutum and Nannochloropsis oculata cells were harvested in both the mid-exponential and early stationary growth phase. Two different cryo-SEM cutting methods were compared: cryo-planing and freeze-fracturing. The results showed that, despite the longer preparation time, TEM visualisation preceded by cryo-immobilisation allows a clear detection of lipid bodies and is preferable to cryo-SEM. Using freeze-fracturing, lipid bodies were rarely detected. This was only feasible if crystalline layers in the internal structure, most likely related to sterol esters or di-saturated triacylglycerols, were revealed. Furthermore, lipid bodies could not be detected using cryo-planing. Cryo-SEM is also not the preferred technique to recognise other organelles besides lipid bodies, yet it did reveal chloroplasts in both species and filament-containing organelles in cryo-planed Nannochloropsis oculata samples.  相似文献

Since the discovery of a flavin-dependent thymidylate synthase (ThyX or FDTS) that is absent in humans but crucial for DNA biosynthesis in a diverse group of pathogens, the enzyme has been pursued for the development of new antibacterial agents against Mycobacterium tuberculosis, the causative agent of the widespread infectious disease tuberculosis (TB). In response to a growing need for more effective anti-TB drugs, we have built upon our previous screening efforts and report herein an optimization campaign of a novel series of inhibitors with a unique inhibition profile. The inhibitors display competitive inhibition toward the methylene tetrahydrofolate cofactor of ThyX, enabling us to generate a model of the compounds bound to their target, thus offering insight into their structure–activity relationships.  相似文献

In this paper, an optimization method is presented for instructions to evacuate by car the population of a region threatened by a hazard. By giving the optimized instructions to the evacuees, traffic conditions and, therefore, the evacuation efficiency are optimized. The instructions, containing a departure time, a safe destination, and a route, are created using an optimization method consisting of two phases: the generation of the search space and the algorithm AES+ evacuation, a version of ant colony optimization. The main contributions of the proposed optimization method are the unique approach to generate the search space in which network degeneration is taken into consideration, the possibility to optimize instructions under the assumption of both full and partial compliance of the evacuees with the instructions, and the flexibility in the sense that the user of the method can define his or her own objective function and choose a suitable traffic simulation model. The paper contains a comprehensive case study. The case study shows that the effectiveness of the optimized instructions is more than doubled when compared with the effectiveness of instructions set up by straightforward rules (like evacuating to the nearest destination using the shortest route). Further, the case study shows that the number of arrivals under optimized, but possibly sub-optimal instructions is equal to at least 90% of the theoretical upper bound on this number of arrivals.  相似文献

Obtaining a quantitative understanding of the transmission dynamics of influenza A is important for predicting healthcare demand and assessing the likely impact of intervention measures. The pandemic of 2009 provides an ideal platform for developing integrative analyses as it has been studied intensively, and a wealth of data sources is available. Here, we analyse two complementary datasets in a disease transmission framework: cross-sectional serological surveys providing data on infection attack rates, and hospitalization data that convey information on the timing and duration of the pandemic. We estimate key epidemic determinants such as infection and hospitalization rates, and the impact of a school holiday. In contrast to previous approaches, our novel modelling of serological data with mixture distributions provides a probabilistic classification of individual samples (susceptible, immune and infected), propagating classification uncertainties to the transmission model and enabling serological classifications to be informed by hospitalization data. The analyses show that high levels of immunity among persons 20 years and older provide a consistent explanation of the skewed attack rates observed during the pandemic and yield precise estimates of the probability of hospitalization per infection (1–4 years: 0.00096 (95%CrI: 0.00078–0.0012); 5–19 years: 0.00036 (0.00031–0.0044); 20–64 years: 0.0015 (0.00091–0.0020); 65+ years: 0.0084 (0.0028–0.016)). The analyses suggest that in The Netherlands, the school holiday period reduced the number of infectious contacts between 5- and 9-year-old children substantially (estimated reduction: 54%; 95%CrI: 29–82%), thereby delaying the unfolding of the pandemic in The Netherlands by approximately a week.  相似文献
