The inhibition of protein-protein interactions (PPIs) is an effective approach for therapy. Owing to their large binding surface areas to target proteins, macrocyclic peptides are suitable molecules for PPI inhibition. In this study, we developed single-chain tandem macrocyclic peptides (STaMPtides) that inhibits the vascular endothelial growth factor (VEGF) receptor 2 (VEGFR2). They were artificially designed to comprise two different VEGFR2-binding macrocyclic peptides linked in tandem by peptide linkers and secreted by Corynebacterium glutamicum. Most potent VEGFR2-inhibitory STaMPtides with length-optimized linkers exhibited >1000 times stronger inhibitory activity than their parental monomeric peptides, possibly due to the avidity effect of heterodimerization. Our approach of using STaMPtides for PPI inhibition may be used to inhibit other extracellular factors, such as growth factors and cytokines. 相似文献

Understanding the interfacial interactions plays a key role in controlling mechanical and physical properties of polymer/clay nanocomposites (PCNs). In this work, the surface interactions between constituents of experimentally prepared PCNs which are the pristine linear low density polyethylene (PE) chains, PE compatibilizers, montmorillonite clay surface layer, and surfactants were studied quantitatively by employing molecular dynamics simulation technique. The interaction energy between the polymer and the clay was found to be inversely proportional with the volume of the surfactant which decreases the electrostatic interactions between the compatibilizer and the hydrophilic clay surface. However, the van der Waals (vdW) interactions between alkyl tails of surfactants and the PE chains increase with the tail length of the surfactants. The most attractive interaction was between the surfactant's head group and the clay surface. We showed that there existed fine balance between the electrostatic and vdW type forces on the stability and the enhanced properties of the PE–organoclay nanocomposites. In this study, effect of processing method on microstructure formation and related electrical conductivity and electromagnetic interference shielding effectiveness of carbon nanofiber (CNF) filled thermoplastic polyurethane (TPU) composites, prepared via three different processing techniques; (i) melt compounding (MC) in a twin screw extruder, (ii) simple solution mixing (SM) on a magnetic stirrer, and (iii) solution mixing with sonication (SM-U) were investigated. It was found that the electrical conductivity values of samples decreased in the order of SM > SM-U > MC for a particular amount of CNF. The electromagnetic test results showed that the samples prepared with SM and SM-U methods yielded higher total shielding effectiveness (SET) values than those prepared with MC. SET values of samples including of 20 phr of CNF prepared with MC, SM-U and SM methods were varied in the range of 10–30 dB, 20–60 dB and 20–80 dB, respectively within a frequency range of 1–12 GHz. 相似文献

A novel method of preparing detergent-enzyme complexes that can be employed in organic media was developed utilizing newly synthesized water-soluble nonionic gemini-type detergents, N,N-bis(3-D-gluconamidopropyl)-3-(dialkyl-L-glutamatecarbonyl)propanamides (BIG2CnCA: n = 10,12,14,16,18) and N,N-bis(3-D-lactonamidopropyl)-3-(dialkyl-L-glutamatecarbonyl)propanamides (BIL2CnCA: n = 16,18), and nonionic twin-headed detergents, N,N-bis(3-D-gluconamidopropyl)alkanamides (BIG1Cn: n = 12,14,16,18,delta9). This method simply entails mixing a selected enzyme with an appropriate detergent in an aqueous solution followed by lyophilization, and it offers the advantages of enhanced enzymatic activity in organic solvents and eliminates both enzyme loss and the necessity for an organic solvent in the preparation stage. Using various modified lipases originating from Aspergillus niger (Lipase A), Candida rugosa (Lipase C), Pseudomonas cepacia (Lipase P), and porcine pancreas (PPL), prepared using the novel method and detergents, including conventional synthesized nonionic detergents such as dialkyl N-D-glucona-L-glutamates (2CnGE: n = 12,18delta9) and octanoyl-N-methylglucamide (MEGA-8), enantioselective transesterifications of 6-methyl-5-hepten-2-ol (sulcatol) and 2,2-dimethyl-1,3-dioxolane-4-methanol (solketal) with a vinyl or isopropenyl carboxylate were carried out in an organic solvent. The modified lipase activity was influenced by both the lipases and the structure of the detergents. The value for the hydrophile-lipophile balance (HLB) of the detergent provided a means of correlating the structure and the obtained modified lipase activity. For detergents of the same class with a HLB value of approximately 9 and 12, the highest activity was obtained for Lipase A and Lipase P, and Lipase C and PPL, respectively. Among detergents of the same HLB value tested, the gemini-type detergents possessing the most bulky head and tail were most effective as a modifier for lipases of all types. The preparation and reaction conditions for these novel gemini-type detergent-modified lipases were optimized using BIG2C12CA (HLB = 9.4) by studying the effect of the detergent/lipase ratio and the nature of organic solvents on the complex formation. The high enzymatic activities of the BIG2C12CA-modified lipases were independent of the solubility of the lipases in organic solvents, unlike in the case of 2CnGE-modified lipases prepared using the conventional suspension system. 相似文献

Investigation into the corrosion inhibition of low carbon steel in 0.1-M HCl solution by 1-ethyl-1-methylpyrrolidinium (EMTFB) and the effect of KI addition on the inhibition efficiency was carried out using potentiodynamic polarization (PDP) and electrochemical impedance spectroscopy and surface analysis (scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDAX)) techniques. Results show that EMTFB suppresses low carbon steel dissolution in the corrosive environment. Inhibition efficiency increased with the increase in EMTFB concentration. Addition of iodide ions to EMTFB raises inhibition efficiency from 75 to 98%. PDP results indicate that EMTFB affects majorly anodic reactions while EMTFB + KI act as cathodic-type inhibitor. The adsorption of EMTFB onto low carbon steel surface is by physical adsorption mechanism and follows Langmuir adsorption isotherm model. SEM and EDAX results confirm the adsorption of EMTFB alone and in combination with KI onto the steel surface. 相似文献
