This paper discusses the flexibility concept in the light of the production-systems capabilities needed to cope with changes occurring at different time horizons. Two basic flexibility components emerge: set-up (in time units) and variety (operations and transfer). The paper further describes a timed Petri-nets approach to machine-flexibility evaluation (set-up and operations variety). A timed Petrinets decomposition technique is used to derive measures of flexible machines' performance under changes that a flexible system has to cope with, such as failures, changeover interruptions, fluctuations in demand, and re-routing.  相似文献

In this work, the possibility of finding newly formed Maillard reaction products produced as a result of the subcritical water extraction (SWE) conditions is explored. Simplified powdered glycation model systems were prepared mixing amino acid (Lys, Arg or Ala) and glucose in a molar ratio 1:4. Samples constituted by glucose or amino acids alone were also prepared as controls. SWE was carried out at room temperature, 100 °C and 200 °C and 100 bar of pressure for 20 min. Different assays were performed in order to determine the extent of glycation by analyzing the decrease of free amino groups and/or the formation of Maillard reaction products (early, advanced and end products). Namely, formation of early colorless Maillard reaction products, Amadori compounds, was detected by ESI-MS; advanced glycation end products (AGEs) were detected by measuring the fluorescence intensity (λex = 360 nm, λem = 460 nm) while end brown products were detected by reading the absorbance at 360 and 420 nm. Besides, the antioxidant capacity of the aqueous extracts was determined by using ABTS and ORACFL assays. Results obtained indicated the occurrence of the Maillard reaction under our specific extraction conditions. Early, advanced and end products were detected in the samples. Caramelization of sugar also occurred. As expected, the extent of the non-enzymatic browning depended on the intensity of the thermal treatment. Additionally, data on antioxidant activity suggested the formation of neoantioxidants. These compounds were predominantly formed at 200 °C. In conclusion, this report demonstrates the formation of antioxidant compounds in simplified glycation model systems under SWE conditions.  相似文献

The active role of alumina, pentalithium aluminate (Li5AlO4, Li-aluminate), and pentasodium aluminate (Na5AlO4, Na-aluminate) as the surface protection coatings produced via atomic layer deposition on Li and Mn-rich NCM cathode materials 0.33Li2MnO3·0.67LiNi0.4Co0.2Mn0.4O2 is discussed. A notable improvement in the electrochemical behavior of the coated cathodes has been found while tested in Li-coin cells at 30 °C. Though all the coated cathodes demonstrate enhanced electrochemical cycling and rate performances, Na-aluminate coated cathodes exhibit exemplary behavior. Prolonged cycling and rate capability testing demonstrate that after more than 400 cycles at 1 C rate, the uncoated cathode delivers only 63 mAh g−1, while those with alumina, Li-aluminate, and Na-aluminate coatings exhibit approximately two times higher specific capacities. The coated cathodes display steady average discharge potential and lower evolution of the voltage hysteresis during prolonged cycling compared to the uncoated cathode. Importantly, Na-aluminate coated cathode shows a lowering in gases (O2, CO2, H2, etc.) evolution. Post-cycling analysis of the electrodes demonstrates higher morphological integrity of the coated cathode materials and lower transition metals dissolution from them. The coatings mitigate undesirable side reactions between the electrodes and the electrolyte solution in the cells.  相似文献

β-Conglycinin is a functional glycoprotein and one of the most important soybean allergens. The aim of the present research was to investigate the role of the N-glycans moieties of β-conglycinin on its in vitro immunoreactivity. The soy allergen was obtained by isoelectric precipitation from commercial soy protein isolate and was enzymatically deglycosylated by PNGase F (Peptide N-Glycosidase F EC 3.5.1.52). In order to optimize deglycosylation conditions different reaction times and allergen concentrations were tested. The extent of deglycosylation was estimated by SDS–PAGE, CZE, RP-HPLC, and MALDI-TOF MS analyses, which provided information related to changes in protein structure. The antigenicity of both native β-conglycinin and its deglycosylated form was evaluated by western-blotting and indirect ELISA employing polyclonal rabbit anti-soybean sera and horseradish peroxidase-labeled goat anti-rabbit IgG while the in vitro allergenicity was assessed by means of indirect competitive inhibition ELISA employing human sera (IgE) of soy allergics. β-Conglycinin was effectively deglycosylated by PNGase F. Data on immunological tests suggested that glycosyl moieties forming this glycoprotein might be involved in its immunoreactivity.  相似文献

The aim of this work is to provide Small and Medium Enterprises (SMEs) with adapted design methods for manufacturing systems. In a total quality strategy, the design activity consists of searching for the means able to satisfy all users' needs from company customers to floor plant workers. That includes usual costs, delays and quality targets but also employees' involvement. Due to the variety of data sources and to the continuous changes of goals, the information is imperfect. The main concerns are uncertainty, inaccuracy and vagueness of both objectives and solutions. The fuzzy set theory provides a quite convenient framework to represent all those imperfections using one mathematical objet. Fuzzy logic is used to aggregate specifications and to deploy solutions. Possibility theory finally allows a graded evaluation of expected performances. The obtained result is graded, providing the engineer with some degrees of freedom for the realization of the manufacturing system. The valuation of the design quality is an undeniable progress towards the continuous improvement of the whole system performance. Finally, such a criterion may also be used for an empirical search for a convenient solution, as for the automatic computation of an optimal result.  相似文献

This work addresses an integrated treatment to produce drinking water involving the ion exchange (IEX) and nanofiltration processes. In the first stage, the experimental procedure consisted of evaluating the IEX equilibrium and the dynamics of fixed-bed saturation/regenerations, for the system Cl?/SO42, using an anionic resin, Purolite A850, in the form sulphate. A mathematical model which considers axial dispersion for the liquid phase and linear driving force for intraparticle mass transfer predicted well the breakthrough curves of Cl? and SO42?. In the second stage, the selective separation of Cl?, Na+ and SO42? ions through a flat membrane nanofiltration TS80 was assessed.  相似文献

To explore the role of protein kinase A (PKA) in the late phase of long-term potentiation (L-LTP) and memory, we generated transgenic mice that express R(AB), an inhibitory form of the regulatory subunit of PKA, only in the hippocampus and other forebrain regions by using the promoter from the gene encoding Ca2+/ calmodulin protein kinase IIalpha. In these R(AB) transgenic mice, hippocampal PKA activity was reduced, and L-LTP was significantly decreased in area CA1, without affecting basal synaptic transmission or the early phase of LTP. Moreover, the L-LTP deficit was paralleled by behavioral deficits in spatial memory and in long-term but not short-term memory for contextual fear conditioning. These deficits in long-term memory were similar to those produced by protein synthesis inhibition. Thus, PKA plays a critical role in the consolidation of long-term memory.  相似文献

ABSTRACT

Monodisperse platinum nanoparticles were synthesized by microemulsion technique. Feasibility of formation of microemulsion with chosen proportions of components is a common problem with this method. Here, prior to preparation of microemulsion for synthesis purpose, systematic study was carried to check the suitability of selected microemulsion system by establishing phase diagram for ternary system at constant temperature on triangular coordinates. Temperature dependency of microemulsion was studied by preparing phase diagram of temperature against mass fraction of aqueous phase in ternary mixture. Both these triangular coordinate diagram at constant temperature and temperature dependency plots helped to choose proper concentrations of components in the system. Effects of molar concentration of water-to-surfactant ratio, platinum salt concentration, continuous oil phase concentration, and presence of cosurfactant with surfactant on particle size were investigated for selected system of components. Synthesized platinum nanoparticles were characterized by dynamic light scattering and transmission electron microscope. Light backscattering profiles obtained by Turbiscan were used to judge the stability of microemulsions. Further, partial weightage of affecting parameters on size of synthesized particle were studied by Taguchi orthogonal array method.  相似文献
