In order to utilise sardinelle (Sardinellaaurita) protein by-products, which is normally discarded as industrial waste in the process of fish manufacturing, heads and viscera proteins were hydrolysed by different proteases to obtain antioxidative peptides. All hydrolysates showed different degrees of hydrolysis and varying degrees of antioxidant activities. Hydrolysate generated with crude enzyme extract from sardine (Sardinapilchardus) displayed high antioxidant activity, and the higher DPPH radical-scavenging activity (87 ± 2.1% at 2 mg/ml) was obtained with a degree of hydrolysis of 6%. This hydrolysate was fractionated by size exclusion chromatography on a Sephadex G-25 into eight major fractions (P1–P8). Fraction P4, which exhibited the highest DPPH scavenging activity, was then fractionated by reversed-phase high performance liquid chromatography (RP-HPLC). Seven antioxidant peptides were isolated. The molecular masses and amino acids sequences of the purified peptides were determined using ESI-MS and ESI-MS/MS, respectively. Their structures were identified as Leu-His-Tyr, Leu-Ala-Arg-Leu, Gly-Gly-Glu, Gly-Ala-His, Gly-Ala-Trp-Ala, Pro-His-Tyr-Leu and Gly-Ala-Leu-Ala-Ala-His. The first peptide displayed the highest DPPH radical-scavenging activity (63 ± 1.57%; at 150 μg/ml) among these peptides. 相似文献

Rapid measurements of milk properties and discrimination of milk origin are necessary techniques for quality control of milk products. The present study was undertaken to evaluate the potential of using front face fluorescence spectroscopy (FFFS) and synchronous fluorescence spectroscopy (SFS) for monitoring the quality of forty-five ewe’s milk samples originating from different feeding systems. Physico-chemical analyses and fluorescence spectra were conducted on samples during lactation periods (the first 11 weeks). The principal component analysis (PCA) separately applied to the physico-chemical and fluorescence spectral data showed only small discrimination between milk samples based on lactation periods and diet compositions. Similar results were obtained by separately applying factorial discriminant analysis (FDA) on each technique. In a second step, concatenation technique were applied to FFF spectra acquired after excitation set at 250, 290, 380 nm and emission set at 410 nm. Results obtained showed a good discrimination among milk samples with regard to feeding systems given to the ewes throughout the lactation periods. The angiotensin I-converting enzyme (ACE) inhibitory activities of protein hydrolysates prepared from heads and viscera of sardinelle (Sardinella aurita) by treatment with various proteases were investigated. Protein hydrolysates were obtained by treatment with Alcalase®, chymotrypsin, crude enzyme preparations from Bacillus licheniformis NH1 and Aspergillus clavatus ES1, and crude enzyme extract from sardine (Sardina pilchardus) viscera. All hydrolysates exhibited inhibitory activity towards ACE. The alkaline protease extract from the viscera of sardine produced hydrolysate with the highest ACE inhibitory activity (63.2 ± 1.5% at 2 mg/ml). Further, the degrees of hydrolysis and the inhibitory activities of ACE increased with increasing proteolysis time. The protein hydrolysate generated with alkaline proteases from the viscera of sardine was then fractionated by size exclusion chromatography on a Sephadex G-25 into eight major fractions (P1–P8). Biological functions of all fractions were assayed, and P4 was found to display a high ACE inhibitory activity. The IC50 values for ACE inhibitory activities of sardinelle by-products protein hydrolysates and fraction P4 were 1.2 ± 0.09 and 0.81 ± 0.013 mg/ml, respectively. Further, P4 showed resistance to in vitro digestion by gastrointestinal proteases. The amino acid analysis by GC/MS showed that P4 was rich in phenylalanine, arginine, glycine, leucine, methionine, histidine and tyrosine. The added-value of sardinelle by-products may be improved by enzymatic treatment with visceral serine proteases from sardine. 相似文献

In this paper, a robust integral sliding mode control problem for networked control systems with multiple data packet losses is investigated. The data packet losses occur in both sensor-to-controller and controller-to-actuator channels in a random way and they are modelled by independent Bernoulli binary distributed white sequences. The considered class of system is also subject to matched uncertainties and external disturbances presence. A specific sliding mode surface which take into consideration the stochastic nature of the system is proposed to generate a stable sliding mode dynamics for closed loop system. An integral sliding mode controller is designed to ensure the reachability of the specified sliding mode domain. Finally, simulation results, carried out on a linearized model of cart pendulum system, illustrate the effectiveness of the proposed approach. 相似文献

3D video is composed out of two or more, temporally synchronized, 2D video streams acquired at different camera poses and accompanied by geometrical information. In a mixed resolution 3D video stream, a subset of views is coded at reduced resolution. It has been shown in the literature that subjective quality of mixed resolution 3D video is close to that of full resolution 3D video. In order to improve the coding gain in mixed resolution coding scenario we present a new depth encoding method called view upsampling optimization. A novel depth distortion metric based on the performance of the depth-based super resolution is also presented. Finally, to improve the quality of the decoded video an improved depth-based super resolution method that uses view synthesis quality mapping is used for upsampling of low resolution views. The simulations, performed with the recently standardized MVC+D encoder, show that the proposed solution combined with the state of the art view synthesis distortion outperforms the anchor MVC+D coding scheme by 14.5 % of dBR on average for the total coded bitrate and by 17 % of dBR on average for the synthesized views. 相似文献

This article is concerned with a nonlinear theory of thermodynamics for elastic materials in which the particles are subjected to classical displacement, temperature, and mass diffusion fields and whose microelements possess microtemperatures and microconcentrations. The equations of the linear theory are also obtained. It is shown that there exists coupling between temperature, chemical potential, microtemperatures, and microconcentrations even for isotropic bodies. With the help of the semigroup theory of linear operators, we prove the well-posedness of the linear anisotropic problem and we study the asymptotic behavior of the solutions. 相似文献

In recent years, the use of fiber‐reinforced polymer composites in civil infrastructure has been promoted as a solution to the deterioration of bridges, buildings, and other structures composed of traditional materials, such as steel, concrete, and wood. Any application of a polymer composite in an outdoor environment invariably involves exposure to moisture. There is also potential for exposure to saline conditions in waterfront or offshore structures, and alkaline environments, as would be encountered by a reinforcing bar in a cementitious material. This study characterizes the sorption and transport of distilled water, salt solution, and a simulated concrete pore solution in free films of vinyl ester, isophthalic polyester (isopolyester) and epoxy resins, all commercially important materials for use in structural composites. Diffusion of all three liquids in each of the three materials was observed to follow a Fickian process. Mass loss was observed for the isopolyester in salt water and concrete pore solution at 60°C, suggesting hydrolysis that was accelerated by the high temperature exposure. Both the rate of uptake, as well as the equilibrium uptake, were greater at 60°C, compared with ambient conditions. Diffusion coefficients calculated from the mass uptake data revealed that, although the epoxy resin had the highest equilibrium uptake, it had the lowest diffusion coefficient. © 1999 John Wiley & Sons, Inc. A highly alkaline trypsin from the intestine of Grey triggerfish (Balistes capriscus), with high activity at low temperature, was purified and characterised. The enzyme was purified to homogeneity using acetone precipitation, Sephadex G-100 gel filtration and Mono Q-Sepharose anion-exchange chromatography, with a 13.9-fold increase in specific activity and 41.3% recovery. The molecular weight of the purified alkaline trypsin was estimated to be 23.2 kDa by sodium dodecyl sulphate–polyacrylamide gel electrophoresis (SDS–PAGE) and size exclusion chromatography. Purified trypsin appeared as a single band on native–PAGE. Interestingly, the enzyme was highly active over a wide range of pH, from 9.0 to 11.5, with an optimum at pH 10.5, using Nα-benzoyl-DL-arginine-p-nitroanilide (BAPNA) as a substrate. The relative activities at pH 9.0, 11.5 and 12.0 were 86.5%, 92.6% and 52.4%, respectively. The enzyme was extremely stable in the pH range 7.0–12.0. In addition, the enzyme had high activity at low and moderate temperatures with an optimum at around 40 °C and had more than 80% of its maximum activity at 20 °C. The purified enzyme was strongly inhibited by soybean trypsin inhibitor (SBTI) and phenylmethylsulphonyl fluoride (PMSF), a serine protease inhibitor. The enzyme showed extreme stability towards oxidising agents, retaining about 87% and 80% of its initial activity after 1 h incubation at 40 °C in the presence of 1% sodium perborate and 1% H2O2, respectively. A two-phase nanocomposite coating that consists of inclusions of silver in a vanadium nitride matrix (VN/Ag) was investigated as a potential adaptive coating with a reduced friction coefficient from 25 to 1000 °C. This nanocomposite structure was selected based on the premise that silver and silver vanadate phases would form on the surface of these coatings, reducing their friction coefficient in the (i) room to mid-range and (ii) mid-range to high temperatures, respectively. Silver and vanadium were expected to react with oxygen at high temperatures and create a lubricious silver vanadate film on the coating. The VN/Ag coatings were deposited using unbalanced magnetron sputtering and their elemental composition was evaluated using X-ray photoelectron spectroscopy. The tribological properties of the materials against Si3N4 balls were investigated at different temperatures. The lowest friction coefficients recorded for samples with identical compositions were 0.35, 0.30, 0.10 and 0.20 at 25, 350, 700 and 1000 °C, respectively. Post-wear testing Raman spectroscopy and X-ray diffraction (XRD) measurements revealed the formation of silver vanadate compounds on the surface of these coatings. In addition, real time Raman spectroscopy and high temperature XRD revealed that silver vanadate, vanadium oxide and elemental silver formed on the surface of these coatings upon heating to 1000 °C. Upon cooling, silver and vanadium oxide were found to combine at about 400 °C, leading predominantly to the formation of silver vanadate phases on the surface of these materials. 相似文献
