We have built and begun testing a small low energy negative ion cyclotron for direct detection of 14C. At present, the cyclotron is operated in a high resolution mode at the 31st harmonic, with 1–2 kV on the dees. The high harmonic and a minimum number of turns of approximately 100, should give a fwhm mass resolution of about 130000 — sufficient to suppress the background from molecular ions such as 13CH?. Background such as scattered ions of 12C? and 13C? should be totally suppressed by the cyclotron acceleration process. (At the 88″ cyclotron at LBL we found that ions only 1% off-resonance are suppressed by more than a factor of 1017.) A miniature Cs sputter source located at the center of the cyclotron is expected to provide more than 1 μA of negative carbon ions. Negative ions are used in order to eliminate the interference from 14N. Unlike high energy cyclotrons, focussing is obtained solely from the axial components of the accelerating electric field. The magnetic field is kept flat to within 1 part in 104 in order to maintain exact isochronism throughout the several thousand accelerating rf cycles. The low final energy of 40 keV eliminates any danger from radiation or need for shielding, and the final orbit radius of only 10.5 cm, reduce the size and cost of the machine to that of conventional mass spectrometers.  相似文献

The composition and volume of ultrafiltrate produced by hollow fiber ultrafiltration of cottage cheese whey with the Bio-Rad Bio-Fiber 50 Miniplant were studied and fitted to models. Temperature, pH, and protein concentration of the feed cheese whey, the flow rate of the feed cheese whey through the Miniplant, and the pressure differential across the membranes were the independent variables in the model fitting. Feed whey temperature and pressure differential across the membranes were the most significant variables affecting the volume of ultrafiltrate produced. We demonstrate a new attractive approach for ubiquitous quantitative chemical or biological sensing when analog signals are acquired from conventional optical disk drives, and these signals are used for quantitative detection of optical changes of sensing films deposited on conventional CD and DVD optical disks. Our developed analytical model of the operation of this Lab-on-DVD system describes the optical response of sensing films deposited onto the read surface of optical disks by taking into account the practical aspects of system performance that include possible reagent leaching effects, water sampling (delivering) efficiency, and possible changes of the film morphology after water removal. By applying a screen-printing process, we demonstrated a laboratory-scale automated production of sensing films with an average thickness of approximately 10 microm and a thickness relative standard deviation of <3% across multiple films. Finally, we developed a system for delivery of water-sample volumes to sensing films on the disk that utilized a multifunctional jewel case assembly.  相似文献

A planar heterojunction bipolar transistor (HBT) with an AlGaAs emitter layer epitaxially grown onto a selectively defined grown base layer, where the base is grown with the collector as part of the original epi, is discussed. The transistors fabricated with this process exhibit good gain and output characteristics. Transistors with 7×7 μm2 emitters have exhibited a DC current gain of 10 to 1000 for base doping from 1×1019 to 8×1017 cm3, respectively, and Early voltages ⩾100 V. The propagation delay of 19-stage ring oscillators was 87 ps/gate. The transistor-fabrication process was designed to be manufacturable, and the planar nature of the transistor surface should permit large-scale integration with good yields  相似文献

Almond hulls and citrus pulp have been fed to dairy cows with variable responses for milk production, but no information exists on their effect on enteric methane emissions. This experiment examined the effects of dietary supplementation with either almond hulls or ensiled citrus pulp on the milk yield, milk composition, and enteric methane emissions of dairy cows. Thirty-two Holstein dairy cows in mid lactation were offered 1 of 3 diets over a 28-d experiment. Twelve cows received a control (CON) diet, 10 cows a diet containing almond hulls (ALH), and 10 cows a diet containing ensiled citrus pulp (CIT). All cows were offered 6.0 kg of dry matter (DM)/d of crushed corn, 2.0 kg of DM/d of cold-pressed canola, and 0.2 kg of DM/d of a mineral mix. In addition, cows fed the CON diet were offered 14.5 kg of DM/d of alfalfa cubes; cows fed the ALH diet were offered 10.5 kg of DM/d of alfalfa cubes and 4.0 kg of DM/d of almond hulls; and cows on the CIT diet were offered 11.5 kg of DM/d of alfalfa cubes and 3.0 kg of DM/d of ensiled citrus pulp. Milk yield was measured daily and milk composition was measured on 4 d of each week. Individual cow methane emissions were measured by a sulfur hexafluoride tracer technique on d 24 to 28 of the experiment. The mean milk yield of cows fed the CON diet (27.4 kg/d) was greater than the mean milk yield of cows fed the ALH diet (24.6 kg/cow per day), whereas the mean milk yield of cows fed the CIT diet (26.2 kg/cow per day) was not different from the mean milk yield from cows fed the other 2 diets. Dietary treatment did not influence the concentrations of milk fat, protein, and lactose or fat yields, but the mean protein yield from cows fed the CON diet (0.87 kg/d) was greater than that from cows fed the ALH diet (0.78 kg/d) but not different to those fed the CIT diet (0.85 kg/d). In general, we found no differences in the proportion of individual fatty acids in milk. The mean pH of ruminal fluid from cows offered the CON diet was not different to the pH in the ruminal fluids of cows offered the ALH or the CIT diets. The mean methane emissions (g/d) and yields (g/kg of DM intake) were not influenced by dietary treatment. These findings indicate that, although almond hulls and ensiled citrus pulp can be used as a low-cost feed supplement, almond hulls did negatively affect milk production and neither inhibited enteric methane emissions.  相似文献

Deficits in performance of both spatial and visual tasks are common following tissue loss in the right temporal lobe. Since spatial and visual attributes are frequently confounded in experimental tasks, we have studied patients following unilateral temporal lobectomy, in an attempt to determine which aspect mediates the observed deficits. Spatial and visual memory performance was compared in normal controls (n = 16), left temporal (LTL; n = 19) and right temporal (RTL; n = 19) lobectomy patients, by presentation of eight abstract designs in a spatial array for subsequent recall and recognition of the designs (visual memory) and recall of their spatial position (spatial memory). By varying the retention intervals for each group, all three groups were matched on both recall and recognition of the designs at sub-ceiling levels. In contrast, recall of the position of the designs (spatial memory), tested at equivalent delays to those of the visual memory tests, revealed a deficit in the RTL patients compared to both controls and LTL patients (p < 0.05). Magnetic resonance imaging (MRI) was used to quantify the extent of resection of the hippocampus and parahippocampal regions in the two patient groups and showed a significant correlation between hippocampal and parahippocampal removal and spatial memory in the RTL group only. These data support the notion of a disproportionately large involvement of the right hippocampus and adjacent regions in spatial memory.  相似文献

We prepared a series of oligosaccharides from king crab cartilage chondroitin sulfate K after exhaustive digestion with testicular hyaluronidase, and determined the structures of four tetrasaccharides and a pentasaccharide by fast atom bombardment mass spectrometry, high performance liquid chromatography analysis of chondroitinase AC-II digests, and 500-MHz 1H NMR spectroscopy. The tetrasaccharides shared the common core structure GlcAbeta1-3GalNAcbeta1-4GlcAbeta1-3GalNAc with various sulfation profiles. One structure was GlcAbeta1-3GalNAc(4S)beta1-4GlcAbeta1-3GalNAc(4S), whereas three of them have the following hitherto unreported structures including a novel glucuronate 3-O-sulfate: GlcA(3S)beta1-3GalNAc(4S)beta1-4GlcAbeta1-3GalNAc(4S), GlcAbeta1-3GalNAc(4S)beta1-4GlcA(3S)beta1-3GalNAc(4S), and GlcA(3S)beta1-3GalNAc(4S)beta1-4GlcA(3S)beta1-3GalNAc(4S), where 3S or 4S represents 3-O- or 4-O-sulfate, respectively. The structure of the pentasaccharide was determined as GlcA(3S)beta1-3GalNAc(4S)beta1-4GlcA(3S)beta1- 3GalNAc(4S)beta1-4GlcA. Chondroitinase ABC digestion of the tetrasaccharides with GlcA(3S) at the internal position destroyed the disaccharide unit containing GlcA(3S) derived from the reducing side and resulted in only the disaccharide unit from the non-reducing side. In contrast, these tetrasaccharides remained totally resistant to chondroitinase AC-II. The results indicated that it is necessary to reevaluate the disaccharide composition of chondroitin sulfate poly- or oligosaccharides purified from various biological sources, since they were usually determined after chondroitinase ABC digestion. It is probable that the structures containing GlcA(3S) would not have been detected.  相似文献

We calculated the electrostatic force between a planar interface, such as a planar-supported lipid bilayer membrane, and the tip of a stylus on which another lipid bilayer or some other biomacromolecular system might be deposited. We considered styli with rounded tips as well as conical tips. To take into account the effect of dynamical hydrogen-bonded structures in the aqueous phase, we used a theory of nonlocal electrostatics. We used the Derjaguin approximation and identified the systems for which its use is valid. We pointed out where our approach differs from previous calculations and to what extent the latter are inadequate. We found that 1) the nonlocal interactions have significant effects over distances of 10-15 A from the polar zone and that, at the surface of this zone, the effect on the calculated force can be some orders of magnitude; 2) the lipid dipoles and charges are located a distance L from the hydrophobic layer in the aqueous medium and this can have consequences that may not be appreciated if it is ignored; 3) dipoles, located in the aqueous region, can give rise to forces even though the polar layer is unchanged, and if this is ignored the interpretation of force data can be erroneous if an attempt is made to rationalize an observed force with a knowledge of an uncharged surface; 4) the shape of the stylus tip can be very important, and a failure to take this into account can result in incorrect conclusions, a point made by other workers; and 5) when L is nonzero, the presence of charges and dipoles can yield a force that can be nonmonotonic as a function of ionic concentration.  相似文献
