At 55 to 70 degrees C, thermal inactivation D-values for Escherichia coli O157:H7, Salmonella, and Listeria monocytogenes were 19.05 to 0.038, 43.10 to 0.096, and 33.11 to 0.12 min, respectively, in ground turkey and 21.55 to 0.055, 37.04 to 0.066, and 36.90 to 0.063 min, respectively, in ground beef. The z-values were 5.73, 5.54, and 6.13 degrees C, respectively, in ground turkey and 5.43, 5.74, and 6.01 degrees C, respectively, in ground beef. In both ground turkey and beef, significant (P < 0.05) differences were found in the D-values between E. coli O157:H7 and Salmonella or between E. coli O157:H7 and L. monocytogenes. At 65 to 70 degrees C, D-values for E. coli O157:H7, Salmonella, and L. monocytogenes were also significantly (P < 0.05) different between turkey and beef. The obtained D- and z-values were used in predicting process lethality of the pathogens in ground turkey and beef patties cooked in an air impingement oven and confirmed by inoculation studies for a 7-log (CFU/g) reduction of E. coli O157:H7, Salmonella, and L. monocytogenes. 相似文献

ABSTRACT: Thermal inactivation D (decimal reduction time at a certain heating temperature) values of Escherichia coli O157:H7 at 55 °C to 70 °C were 21.36 to 0.031 min in raw franks and 24.91 to 0.038 min in fully cooked franks. Although statistically significant differences were found on the D values of E. coli O157:H7 between raw and fully cooked franks, the z value of E. coli O157:H7 in raw franks (5.07 °C) and fully cooked franks (5.08 °C) was not significantly different. The obtained D and z values were used to validate the process lethality of the pathogen in the raw franks that were processed according to a multistage cooking/cooling schedule or in the fully cooked franks that were pasteurized in-packages via steam. In this study, the calculated process lethality for both the cooking (process lethality = 254 min) and post-cook pasteurization (process lethality = 39 min) processes was far greater than the processing time that was needed for achieving a 7D reduction of E. coli O157:H7 during cooking and post-cook pasteurization of the franks. 相似文献

Absorption of niacin from the rumen was tested in vivo using a buffer solution that contained either nicotinic acid or nicotinamide. The experimental design was a 3 X 3 Latin square with three midlactation, ruminally cannulated Holstein cows. Orthogonal comparisons were control versus niacin and nicotinic acid versus nicotinamide. Nicotinamide was more rapidly absorbed from the rumen than nicotinic acid; this may have been caused by differences in the dissociation constants of the compounds. The effects that differential absorption may have on ruminal fermentation and animal metabolism require further study. 相似文献

This two-phase trial involved 83 Holstein heifers. The rearing phase consisted of two diets (alfalfa silage plus corn grain for ad libitum intake vs. corn silage plus urea for ad libitum intake) and two breeding age groups (13 vs. 16 mo). The lactation phase compared the above treatments plus two lactation feeding systems: concentrate fed individually to production versus a TMR. The heifers were assigned randomly to the rearing phase at 7 wk of age and fed their respective diets until 14 d prepartum. They were placed on preassigned lactation diets 14 d prepartum and remained on the lactation phase for 550 consecutive d. Daily gains and height at the withers were similar between forage groups during the first half of the rearing phase; in the second half, the heifers fed alfalfa silage were taller at the withers. Those fed corn silage consumed less DM and CP throughout the rearing phase and gained more BW than the heifers fed alfalfa in the latter half. There were no differences in daily gain or DMI between the breeding age groups. In the lactation phase, the group fed alfalfa for ad libitum intake consumed more DM and gave more milk during the first 90 d of lactation than heifers fed corn silage. However, after 90 d the difference in cumulative milk production was not significant. There were no differences in milk production. FCM, or DMI between breeding age groups. The cows fed concentrate consumed more DM and gave more milk during the first 90 d of lactation. By 550 d, there were no differences. The objective of this study was to determine the effect of temperature on whole milk density measured at four different temperatures: 5, 10, 15, and 20 °C. A total of ninety-three individual milk samples were collected from morning milking of thirty-two Holstein Friesian dairy cows, of national average genetic merit, once every two weeks over a period of 4 weeks and were assessed by Fourier transform infrared spectroscopy for milk composition analysis. Density of the milk was evaluated using two different analytical methods: a portable density meter DMA35 and a standard desktop model DMA4500M (Anton Paar GmbH, UK). Milk density was analysed with a linear mixed model with the fixed effects of sampling period, temperature and analysis method; triple interaction of sampling period x analysis method x temperature; and the random effect of cow to account for repeated measures. The effect of temperature on milk density (ρ) was also evaluated including temperature (t) as covariate with linear and quadratic effects within each analytic method. The regression equation describing the curvature and density–temperature relationship for the DMA35 instrument was ρ = 1.0338−0.00017T−0.0000122T2 (R2 = 0.64), while it was ρ = 1.0334 + 0.000057T−0.00001T2 (R2 = 0.61) for DMA4500 instrument. The mean density determined with DMA4500 at 5 °C was 1.0334 g cm−3, with corresponding figures of 1.0330, 1.0320 and 1.0305 g cm−3 at 10, 15 and 20 °C, respectively. The milk density values obtained in this study at specific temperatures will help to address any bias in weight–volume calculations and thus may also improve the financial and operational control for the dairy processors in Ireland and internationally. 相似文献

Identifying the sources and impacts of organic and inorganic contaminants at the watershed scale is a complex challenge because of the multitude of processes occurring in time and space. Investigation of geochemical transformations requires a systematic evaluation of hydrologic, landscape, and anthropogenic factors. The 1160 km2 Boulder Creek Watershed in the Colorado Front Range encompasses a gradient of geology, ecotypes, climate, and urbanization. Streamflow originates primarily as snowmelt and shows substantial annual variation. Water samples were collected along a 70-km transect during spring-runoff and base-flow conditions, and analyzed for major elements, trace elements, bulk organics, organic wastewater contaminants (OWCs), and pesticides. Major-element and trace-element concentrations were low in the headwaters, increased through the urban corridor, and had a step increase downstream from the first major wastewater treatment plant (WWTP). Boron, gadolinium, and lithium were useful inorganic tracers of anthropogenic inputs. Effluent from the WWTP accounted for as much as 75% of the flow in Boulder Creek and was the largest chemical input. Under both hydrological conditions, OWCs and pesticides were detected in Boulder Creek downstream from the WWTP outfall as well as in the headwater region, and loads of anthropogenic-derived contaminants increased as basin population density increased. ABSTRACT Polyphenol oxidase (PPO) was isolated from Bramley's Seedling apples with 75.7‐fold purification and 26.5% recovery by ammonium sulfate precipitation, phenyl sepharose chromatography, ion exchange chromatography, and hydroxyapatite chromatography. Molecular weight was estimated to be about 45 kDa by sodium dodecyl sulfate‐polyacrylamide gel electrophoresis (SDS PAGE). Optimum PPO activity was at pH 6.5 and greater than 50% activity was retained during storage for 72 h at pH 5.5 to 6.5. Optimum temperature for activity was 30 °C and the enzyme had residual activity of greater than 50% during storage for 72 h at 20 °C to 30 °C and for 24 h at 40 °C to 50 °C. Of the substrates tested, activity was greatest with 4‐methylcatechol followed by catechol, pyrogallol, and (?)epicatechin. Arabinoxylans (AX) were extracted from Sonalika variety of wheat (whole wheat flour and wheat bran) with barium hydroxide and sodium hydroxide and purified by a combination of alcohol precipitation and glucoamylase digestion. Structural features of purified AX were elucidated by methylation analysis, 13C NMR, FT‐IR, periodate oxidation and optical rotation measurements. The AX showed a backbone of xylose residues with β(1–4) linkages and were branched mainly through O‐3 of xylose residues. Completely branched xylosyl residues were also present. A model warehouse with carefully regulated environmental conditions was used to study the behavior of adult Indian meal moths during commodity infestation. Sequences of moth distributions in the warehouse, calculated by spatial analysis, clearly showed the coordinated movements of moth populations from eclosion to death. Many facets of adult behavior in a warehouse were coordinated with the photoperiod. Adult moths emerged at the end of a photophase and fairly rapidly moved to the walls and, to a lesser extent, to the undersides of the commodity pallets. Most females are mated in the first 24 h after emergence, largely during the scotophase. Air circulation within the warehouse probably compromised pheromone-directed guidance of males to females, although pheromones may still have a major role in regulating other aspects of male mating behavior. When mating subsided, many of the males flew upwind to the air circulating unit, but only during the photophases. The females moved to the return air side of the warehouse and then migrated towards the same end as the males, but only during the scotophases. They moved from pallet to pallet largely by walking on the warehouse floor and laid eggs in or on the cups of commodity. Direct oviposition on the commodity during the infestation period was lower than expected, probably because commodity odors were dissipated by air circulation and did not provide appropriate orientation.
