The periplasmic aldehyde oxidoreductase PaoABC from Escherichia coli is a molybdenum enzyme involved in detoxification of aldehydes in the cell. It is an example of an αβγ heterotrimeric enzyme of the xanthine oxidase family of enzymes which does not dimerize via its molybdenum cofactor binding domain. In order to structurally characterize PaoABC, X-ray crystallography and small angle X-ray scattering (SAXS) have been carried out. The protein crystallizes in the presence of 20% (w/v) polyethylene glycol 3350 using the hanging-drop vapour diffusion method. Although crystals were initially twinned, several experiments were done to overcome twinning and lowering the crystallization temperature (293 K to 277 K) was the solution to the problem. The non-twinned crystals used to solve the structure diffract X-rays to beyond 1.80 Å and belong to the C2 space group, with cell parameters a = 109.42 Å, b = 78.08 Å, c = 151.77 Å, β = 99.77°, and one molecule in the asymmetric unit. A molecular replacement solution was found for each subunit separately, using several proteins as search models. SAXS data of PaoABC were also collected showing that, in solution, the protein is also an αβγ heterotrimer. 相似文献

In this paper, we analyse a systematically designed and easily tunable backstepping-based boundary control concept for a gantry crane with heavy chain and payload. The corresponding closed-loop system is formulated as an abstract evolution equation in an appropriate Hilbert space. Non-restrictive conditions for the controller coefficients are derived, under which the solutions are described by a C0-semi-group of contractions, and are asymptotically stable. Moreover, by applying Huang's theorem we can finally even show that under these conditions the controller renders the closed-loop system exponentially stable. 相似文献

The crushing behaviour of sinter from blast furnace operations is known to depend on its temperature. In order to characterize it, hot sinter was subject to single particle tests in a very basic drop weight tester. Additionally, a laboratory jaw crusher was sacrificed to work the same feed material. As the results regarding net energy input were not consistent, two common rocks were tested in comparison with both the drop weight and the jaw crusher. Although some discrepancies remained, the results with the rocks can basically be considered as consistent. This investigation illustrates some of the difficulties that can arise when established test methods are applied on material with deviant properties. 相似文献

This article deals with the differences in the long-term durability and thermal stability under load of terrace deckings from various materials. The tested materials were deckings made of wood, thermally modified timber (TMT), and wood–polymer–composites (WPC). For the determination of the test temperatures for component testing according to EN 310, the surface temperatures of the decks during a normal hot summer day were measured. A cyclic test according to EN 321 was applied to all decking materials. Afterwards the component testing was repeated. All wooden samples reveal considerable cracks, some were twisted, and few were even broken. In particular regarding the optical appearance, wood decks show advantages against the TMT decks. Some WPC decks show very fine cracks on the face, which were additionally analyzed by means of X-ray computed tomography (CT). Except for the WPC deck with higher wood content, no WPC deck revealed significant changes after the cyclic test. The CT analysis was also suitable to find cracks inside the materials and illustrate them. Thus, the whole damage inside a sample could be characterized by calculating a kind of error pattern. No considerable cracks or failures could be observed on the WPC decks. 相似文献

Microassembly of piezoceramic fibers in micro cavities at the surface of sheet metal is a novel approach for high volume production of smart adaptronic structural metal parts. In this paper a technology, manufacturing processes and characterization results of metal sheets with an active piezo-metal substructure based on directly integrated piezoceramic fibers are described. The processes include micro-milling of cavities in sheet metal, precision grinding of piezoceramic fibers, PECVD insulating layers with high dielectric strength, microassembly and force-locked joining by forming. In experiments, measurements of the surface roughness and geometric parameters of the piezoceramic fibers and micro cavities were performed. Further, the electrical properties of the insulation coatings were measured. The sensor function of the piezo-metal substructure was proven by a mechanical excitation resulting in a proportional measured sensor signal. 相似文献

Squalene–hopene cyclases (SHCs) catalyze the polycyclization of squalene into a mixture of hopene and hopanol. Recently, amino-acid residues lining the catalytic cavity of the SHC from Alicyclobacillus acidocaldarius were replaced by small and large hydrophobic amino acids. The alteration of leucine 607 to phenylalanine resulted in increased enzymatic activity towards the formation of an intermolecular farnesyl–farnesyl ether product from farnesol. Furthermore, the addition of small-chain alcohols acting as nucleophiles led to the formation of non-natural ether-linked terpenoids and, thus, to significant alteration of the product pattern relative to that obtained with the wild type. It is proposed that the mutation of leucine at position 607 may facilitate premature quenching of the intermediate by small alcohol nucleophiles. This mutagenesis-based study opens the field for further intermolecular bond-forming reactions and the generation of non-natural products. 相似文献

Fish larvae may intercept their own wake during sharp turns, which might affect their escape performance. We analysed C-starts of larval zebrafish (Danio rerio, Hamilton, 1822) using a computational fluid dynamics approach that simulates free swimming (swimming trajectory is determined by fluid forces) by coupling hydrodynamics and body dynamics. The simulations show that fish may intercept their own wake when they turn by 100–180°. During stage 1 of a C-start, the fish generates a strong jet at the tail that is shed into the wake. During stage 2, the fish intercepts this wake. Counterfactual simulations showed that wake interception increased the lateral force on the fish and reduced the fish''s turning angle by more than 5°. Wake interception caused no significant acceleration tangential to the trajectory of the fish and did not affect total power output. While experimental and simulation evidence suggests that fish larvae can either undershoot or intercept but not overshoot their wake, our simulations show that larger fish might be able to avoid intercepting their wake by either under- or overshooting. As intercepting its own wake modifies the fish''s escape trajectory, fish should account for this effect when planning their escape route. 相似文献

A reusable catalyst with dual active center for chemical water oxidation is synthesized for the first time by immobilization of iridium oxide nanoparticles (IrO2 NPs) on monodisperse-porous manganese oxide microspheres acting both catalytic active center and support. Individual catalytic activity of manganese oxide microspheres is explained by multiple oxidation states of manganese which are capable of forming oxidative oxygen species. Monodisperse-porous microspheres in the form of Mn5O8, MnO2 and Mn2O3 are used for synthesis of different catalysts and the highest activity in water oxidation is observed with the catalyst synthesized using Mn5O8 microspheres. The catalytic activity is correlated with the total Mn(II) and Mn(III) percentage of manganese oxide type selected for composite catalyst. The oxygen evolution up to 244 μmol is achieved in 30 min with the catalyst synthesized using Mn5O8 microspheres. Maximum TON and TOF numbers are obtained as 298 and 557 h?1 with excellent reusability. 相似文献
