Plasmalogens (1-O-alk-1′-enyl-2-acyl-sn-glycero-3-phosphocholines and -phosphoethanolamines) are important constituents of spermatozoa membranes and possess significant antioxidative properties. This particularly holds as plasmalogens from spermatozoa also possess a very high content of highly unsaturated fatty acyl residues (especially 22:6). The organic spermatozoa extracts of two different ruminants (cattle and roe deer) were analyzed for their contents of characteristic choline plasmalogen oxidation products by matrix-assisted laser desorption and ionization time-of-flight (MALDI-TOF) mass spectrometry. It will be shown that 1-hydroxy-2-docosahexaenoyl-sn-glycero-3-phosphocholine (LPC 22:6) and formyl-LPC 22:6 are reliable measures of lipid oxidation of spermatozoa and allow, accordingly, conclusions about the storage conditions. All data on spermatozoa were also confirmed by the investigation of the oxidation behavior of selected reference compounds. It will be shown that, equally if plasmalogens or diacyl PC species are used, oxidation takes place primarily at the double bond next to the glycerol backbone. These data were additionally confirmed by recording the corresponding post source decay (PSD) fragment ion spectra.  相似文献

Mitochondrial biogenesis is a highly controlled process that depends on diverse signalling pathways responding to cellular and environmental signals. AMP-activated protein kinase (AMPK) is a critical metabolic enzyme that acts at a central control point in cellular energy homeostasis. Numerous studies have revealed the crucial roles of AMPK in the regulation of mitochondrial biogenesis; however, molecular mechanisms underlying this process are still largely unknown. Previously, we have shown that, in cellular slime mould Dictyostelium discoideum, the overexpression of the catalytic α subunit of AMPK led to enhanced mitochondrial biogenesis, which was accompanied by reduced cell growth and aberrant development. Here, we applied mass spectrometry-based proteomics of Dictyostelium mitochondria to determine the impact of chronically active AMPKα on the phosphorylation state and abundance of mitochondrial proteins and to identify potential protein targets leading to the biogenesis of mitochondria. Our results demonstrate that enhanced mitochondrial biogenesis is associated with variations in the phosphorylation levels and abundance of proteins related to energy metabolism, protein synthesis, transport, inner membrane biogenesis, and cellular signalling. The observed changes are accompanied by elevated mitochondrial respiratory activity in the AMPK overexpression strain. Our work is the first study reporting on the global phosphoproteome profiling of D. discoideum mitochondria and its changes as a response to constitutively active AMPK. We also propose an interplay between the AMPK and mTORC1 signalling pathways in controlling the cellular growth and biogenesis of mitochondria in Dictyostelium as a model organism.  相似文献

Two series of polyesters containing phenoxaphosphine rings and several halogens were synthesized by low-temperature solution polycondensation in the presence of triethylamine. One series, containing halogens only in the bisphenol moiety, was obtained from 2,8-dichloroformyl-10-phenylphenoxaphosphine-10-oxide and chlorinated bisphenol. In the second series, halogens belonged both to bisphenol and diacid dichlorides. For the last series, 2,8-dichloroformyl-10-(4-bromophenyl)phenoxaphosphine-10-oxide was prepared as a new monomer. All resulting polyesters were characterized by elemental analysis, reduced viscosity, IR and 1H NMR spectroscopy, and thermogravimetric analysis. These polymers began to lose weight at about 400°C in air being seen as self-extinguishing ones.  相似文献

Using a D-optimal design of experiments, the influences of feedstock powder and plasma gases on deposition efficiency, gas tightness, and the electrochemical behavior of vacuum plasma-sprayed yttria-stabilized zirconia for solid oxide fuel cell electrolytes were examined. In-flight particle temperature and velocity, measured by online particle diagnostics, were correlated with plasma and deposit properties. Electrochemical testing of cells was performed to determine the influence of gas tightness and microstructure of electrolyte deposit on cell behavior. This study examined correlations between the differential absorbance at 272nm (deltaA272) and the formation of disinfection by-products (DBPs) in chlorinated water from the Tolt River, a water source for Seattle, WA. The DBPs investigated included chloroform (CHCl3), dichlorobromomethane (CHCl2Br), mono-, di- and trichloroacetic acids (MCAA, DCAA, and TCAA, respectively), chloral hydrate (CH), dichloroacetonitrile (DCAN) and 1,1,1-trichloropropanone (TCP). Whereas the kinetics of DBP formation are complex and are non-linear, the same DBP data represented as a function of deltaA272 are quite simple. Absorbance decreases when the water is chlorinated, i.e., deltaA272 is always negative. The DBP vs. -deltaA272 correlations can almost always be quantified by linear equations, at least above some threshold value of -deltaA272, with R2 values > 0.95. The only DBP that did not follow this trend was CH, for which an exponential relationship better described the data. TCP and DCAN were unstable at pH 7 and 8, but at pH 6 linear correlations between their concentrations and -deltaA272 were as strong as those for the more stable DBPs. The threshold -deltaA272 value is approximately the same for many of the DBPs studied, supporting the hypothesis that individual DBPs are released following the formation of a common intermediate, or at least a small group of similar intermediates. The DBP vs. -deltaA272 correlations may have practical value since they provide an alternative approach for monitoring the formation of individual DBP species on-line, but the generality of the relationships needs to be further examined.  相似文献

Gut and blood samples from 119 finishing pigs derived from 11 farms were collected during routine slaughter at an abattoir. Sections of formalin-fixed, paraffin-embedded tissues were labeled immunohistochemically using genus-specific, mouse monoclonal antibody against chlamydial lipopolysaccharide; goat polyclonal antiserum against the major outer membrane protein of Chlamydia trachomatis; and mouse monoclonal antibody against the ovine abortion subtype of C. psittaci. Gut samples from 33 of 111 (29.7%) individual pigs stained positive with the genus-specific monoclonal antibody, and of these 30 of 32 (93.7%) also reacted with the C. trachomatis-specific antiserum. Labeled inclusions were restricted to mature enterocytes of the large intestine in 33 of 111 cases. Infection of small intestinal enterocytes was noted in only one of 82 ileal samples. The blood samples were tested for antichlamydial antibodies by enzyme-linked immunosorbent assay (ELISA) and complement fixation test (CFT). With ELISA, 95 of the 115 sera tested (82.6%) yielded positive antichlamydial reactions. With CFT, 34 of the 119 sera tested (28.6%) were unequivocally positive (> or = 1:10, 100% binding), and 10 (7.6%) yielded doubtful positive reactions (1:10, 50-75% binding). Positive ELISA and CFT titers showed poor agreement (kappa = 0.112), whereas the agreement between positive findings by immunohistochemical labeling and CFT was fair (kappa = 0.205).  相似文献

This paper demonstrates the capability of our previously published undoped Double‐Gate (DG) MOSFET explicit and analytical compact model to also forecast the effect of the volume inversion (VI) on the intrinsic capacitances. For that purpose, we present simulation results for these capacitances. We show now that the model presents an accurate dependence on the silicon layer thickness, consistent with two‐dimensional numerical simulations, for both thin and thick silicon films. As opposed to our previous work, here we test the capacitance model for three different film thicknesses and also show that the transition from VI regime to dual gate behaviour is well simulated. We demonstrate in this paper that even if the current drive and transconductance are enhanced in VI regime, our results show that intrinsic capacitances are higher as well, which may limit the high‐speed (delay time) behaviour of DG MOSFETs under VI regime. The good agreement between the numerical simulations and our model shows the high potential of our complete DG MOSFET model. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

The invasive softshell clam (Mya arenaria Linnaeus, 1758) is native to the northwestern region of the Atlantic Ocean. This species has been introduced in the northeast Pacific and along the European coasts, due to intense naval transports and aquaculture, and it is now present in all the European seas. In this paper we describe seven new microsatellite loci for Mya arenaria. The isolated loci are polymorphic with a number of alleles per locus between 6 and 14. The observed and expected heterozygosities ranged from 0.417 to 0.951, and from 0.643 to 0.895, with an average of 0.716 and 0.775, respectively. These microsatellite markers should be useful in analyzing this species' genetic diversity, which could explain various processes of its invasion history.  相似文献

Transparent permeation barrier layers are not only used for food packaging but are also needed to encapsulate flexible electronic devices. Magnetron sputtering allows the deposition of high quality oxide barrier layers with a low water vapor and oxygen permeation. This paper compares different metal oxide layers which are deposited onto a polyethylene terephthalate (PET) film using a reactive dual magnetron sputtering process. The oxides of aluminum, silicon, titanium, zinc and a zinc-tin alloy are compared regarding their permeation barrier, structural and surface properties to determine the relationship between the layer structure and the gas permeation. Thereby, scanning electron microscopy (SEM) and atomic force microscopy (AFM) were used to characterize the morphology and surface structure of the layers and X-ray diffraction analysis (XRD) was used to determine the solid state phase.Cross-section images taken with SEM show a very compact structure for both aluminum oxide and zinc-tin oxide layers. These materials also have the lowest water vapor permeation compared to all other materials. Zinc oxide and titanium oxide layers both exhibit a columnar structure. Zinc oxide is polycrystalline and has a surprising low water vapor and oxygen permeation. In contrast to that the amorphous titanium oxide layers show a high water vapor and oxygen permeation which is not decreasing with an increasing layer thickness above 40 nm.  相似文献
