We describe a rapid, sensitive and semi-quantitative plate assay for monitoring pheromone activity in the fission yeast Schizosaccharomyces pombe. It is based on the observation that meiosis requires stimulation by pheromone and exploits diploid strains that will only sporulate after addition of exogenous pheromone. The tester strains are heterozygous for mating type, are non-switching, and are mutated in one of the early subfunctions (either mat1-Mc or mat1-Pc), so that meiosis is only induced after exposure to exogenous pheromone (M-factor or P-factor, respectively). Pheromone activity is assessed as an iodine-positive halo of sporulation surrounding the pheromone source, and the width of the halo is related to the amount of pheromone being produced. The assay is sufficiently sensitive to monitor the low amount of M-factor produced by an M mam1 strain, and its sensitivity towards P-factor is greatly increased by using a hyper-sensitive tester strain lacking the Sxa2 protease that is believed to degrade this pheromone. The laboratory tests showed that a dual-phase steel can be produced on the basis of the chemical composition given here, providing the coiling temperature is below the martensite start temperature. It is then possible to dispense with an additional air cooling period on the runout table, which is mandatory with production concepts known up to now. The best mechanical properties are attained at a coiling temperature of 200°C. The final rolling temperature should be around Ar3 to avoid a heavy ferrite deformation without softening and, on the other hand, to reduce the yield stress increase caused by aging. Nitrogen aging can be suppressed by the addition of small amounts of titanium to bind nitrogen, forming a dispersion of titanium nitrides. Detailed chemical analysis of wintertime PM?? collected at a rural village site in Germany showed the presence of a series of compounds that correlated very well with levoglucosan, a known biomass burning tracer compound. Nitrated aromatic compounds with molecular formula C?H?NO? (M(w) 169) correlated particularly well with levoglucosan, indicating that they originated from biomass burning as well. These compounds were identified as a series of methyl-nitrocatechol isomers (4-methyl-5-nitrocatechol, 3-methyl-5-nitrocatechol, and 3-methyl-6-nitrocatechol) based on the comparison of their chromatographic and mass spectrometric behaviors to those from reference compounds.Aerosol chamber experiments suggest that m-cresol, which is emitted from biomass burning at significant levels, is a precursor for the detected methyl-nitrocatechols. The total concentrations of these compounds in the wintertime PM??were as high as 29 ng m?3, indicating the secondary organic aerosol (SOA) originating from the oxidation of biomass burning VOCs contributed non-negligible amounts to the regional organic aerosol loading. 相似文献

Seven species of teleost fishes comprising major prey of northwest Atlantic harbor seals were analyzed for polybrominated diphenyl ethers (PBDEs). PBDE concentrations in whole fish samples (n = 87) were compared with those measured previously in harbor seal blubber to evaluate the transfer of PBDEs from prey to predator. Hexabromocyclododecane (HBCD) concentrations were measured in three fish species to provide an initial estimation of HBCD contamination in this ecosystem. HBCD was detected in 87% of the fish samples at concentrations ranging from 2.4 to 38.1 ng/g, lw (overall mean 17.2 ± 10.2 ng/g, lw). ∑PBDE concentrations in fish ranged from 17.9 to 94 ng/g, lw (overall mean 62 ± 34 ng/g, lw). ∑PBDE concentrations in the harbor seals were two orders of magnitude higher than levels in the fish. Biomagnification factors (BMFs) from fish to seals averaged from 17 to 76, indicating that tetra- to hexa-BDEs are highly biomagnified in this marine food web. BDE-47 was the dominant congener in all samples, suggesting exposure to the penta-BDE mixture. The presence of higher brominated congeners including BDE-209 at measurable levels in fish and seal tissue, along with the very high biomagnification of BDE-153, as well as − 155, and − 154, suggests recent exposure to the octa- and deca-BDE formulations in this US coastal marine food web, as well as the additional contribution of BDE-209 debromination in fish to the loading of persistent PBDEs in the seals. Fast analysis of bioaerosols in clean room environments is necessary in order to prevent contamination of pharmaceutical products, minimize machine downtimes, or both. The detection and identification of microbes will be carried out in several steps: After impaction of the aerosol on a surface, the particles are presorted with glancing light illumination and fluorescence imaging in order to distinguish between abiotic and biotic particles. Since only the biotic particles are of interest, the analysis time can be minimized due to reduction of the data set. Nanoarchitecturing of carbon nanospheres onto the surface of activated carbon (AC) gives birth to a new composite carbon material that features a hierarchical structure with macro‐ and nanometer dimensions of the respective carbon components and exhibits a remarkably enhanced adsorption capability for heavy‐metal ions ( and Fe3+) from aqueous solution as compared to AC. Thus, we first propose that nanoarchitecturing of AC can be utilized not only as a flexible method for the synthesis of novel, hybrid, nanostructured composite carbon materials but also as a new and “green‐route” strategy for functionalization of the surface of AC in an effective manner. Perfluorooctane sulfonate (PFOS) and other perfluoroalkyl compounds (PFCs) were measured in liver samples from 29 marine tucuxi dolphins from Rio de Janeiro state (RJ), Brazil. PFC measurement combined liquid chromatography and mass spectrometry, using a CapLC system connected to a Quadrupole-LIT mass spectrometer. PFOS was the only PFC detected and it was so in all samples. PFOS concentrations (ng x g(-1) dw) of dolphins (n=23) from the highly contaminated Guanabara Bay (in RJ) varied between 43 and 2431 as well as between 76 and 427 from areas of RJ other than the quoted bay (n=6). Concentrations of three fetuses and one neonate varied between 664 and 1590. Fetus/mother ratios were calculated in two situations (2.75 and 2.62). It seems that mother-to-calf transference plays important role for relationships between PFOS and age. When a one-year-old male calf presenting 2431 ng x g(-1) dw was excluded from the test, significant correlations were observed between PFOS concentrations and both age and total length. Despite the placental transference, PFOS concentrations were not significantly lower in females than in males. PFOS levels in marine tucuxi dolphins from Guanabara Bay are among the highest detected to date in cetaceans, and this may represent a threat to the small population concerned. 相似文献

Electromechanical interactions can be described by so-called constitutive equations.Constitutive equations describing the coupling of multibody dynamics with Kirchhoffs theory of electrical networks as a quasi stationaryapproximation of Maxwells theory define discrete electromechanicalsystems, i.e. systems with a finite degree of freedom.Then, based on the principle of virtual work, motion equations can be obtained as Lagranges equations in explicit form due to a unified approach.
