The synthesis of new nucleoside derivatives, ara-uridine-2'-carbamates, and their incorporation into synthetic DNA oligomers is described. The modification directs ligands into the major groove of duplex DNA and somewhat destabilizes the duplexes of modified oligonucleotides with complementary DNA or RNA. In the case of pyrenemethyl carbamate modification in DNA-DNA duplexes, the destabilization is considerably reduced. The pyrenemethyl derivative also shows remarkable spectral properties: a "reversed" absorbance change for pyrene at 350 nm in the course of denaturation of the DNA duplex, as compared to the change seen in the nucleotide absorbance at 260 nm. This derivatization also causes pronounced sequence-dependent excimer formation in the major groove.  相似文献

A great advantage of the white-light interferometry is that it can be used for profile objects with a rough surface. A speckle pattern that arises in the image plane allows one to observethe interference; however, this pattern is also the source of the measurement uncertainty. We derive the theoretical limits of the longitudinal uncertainty by virtue of the first-order statistics of thespeckle pattern. It is shown that this uncertainty depends on the surface roughness of the measured object only; it does not depend on the setup parameters.  相似文献

We have constructed a broadband ultrafast time-resolved infrared (TRIR) spectrometer and incorporated it into our existing time-resolved spectroscopy apparatus, thus creating a single instrument capable of performing the complementary techniques of femto-/picosecond time-resolved resonance Raman (TR3), fluorescence, and UV/visible/infrared transient absorption spectroscopy. The TRIR spectrometer employs broadband (150 fs, approximately 150 cm(-1) FWHM) mid-infrared probe and reference pulses (generated by difference frequency mixing of near-infrared pulses in type I AgGaS2), which are dispersed over two 64-element linear infrared array detectors (HgCdTe). These are coupled via custom-built data acquisition electronics to a personal computer for data processing. This data acquisition system performs signal handling on a shot-by-shot basis at the 1 kHz repetition rate of the pulsed laser system. The combination of real-time signal processing and the ability to normalize each probe and reference pulse has enabled us to achieve a high sensitivity on the order of deltaOD approximately 10(-4) - 10(-5) with 1 min of acquisition time. We present preliminary picosecond TRIR studies using this spectrometer and also demonstrate how a combination of TRIR and TR3 spectroscopy can provide key information for the full elucidation of a photochemical process.  相似文献

The effect of production parameters for electron-beam deposition of two-layer metal - ceramic coatings in one production cycle on the structure and phase composition of the outer ceramic layer based on zirconium oxide stabilized with yttrium oxide is studied. The in-diffusion of platinum into a low-doped n-type float zone silicon guided and enhanced by radiation damage produced by co-implantation of helium ions was investigated. The implantation of 1 MeV platinum ions at different doses ranging from 5 × 1011 to 5 × 1012 cm−2 was used to produce a finite source for platinum diffusion. Single and multiple energy implantation of helium ions with energies 7, 9, 11 and 13 MeV introducing different profiles of radiation defects were applied to enhance and shape the diffusion of platinum atoms performed by 20 min annealing at 725 °C in vacuum. The distribution of in-diffused platinum was studied by monitoring the acceptor level of substitutional platinum (EC − ET = 0.23 eV) by deep level transient spectroscopy. Results show that the helium co-implantation significantly enhances platinum diffusion and allows its control up to the depths of hundreds of micrometers. The resulting Pts distribution is given by the profile of radiation damage produced by helium ions while the amount of in-diffused Pts can be controlled by the dose of platinum implantation. It is also shown that an extra annealing at 685 °C performed prior to helium implantation substantially increases the amount of in-diffused platinum.  相似文献

Selection of Optimal Stopping Time for Nonlinear Diffusion Filtering   总被引:7,自引:3,他引:7  
We develop a novel time-selection strategy for iterative image restoration techniques: the stopping time is chosen so that the correlation of signal and noise in the filtered image is minimized. The new method is applicable to any images where the noise to be removed is uncorrelated with the signal, under the assumptions that the filter used is suitable for the given type of data, and that neither the additive noise nor the filtering procedure alter the average gray value; no other knowledge (e.g. the noise variance, training data etc.) is needed.We analyse the theoretical properties of the method, then test the performance of our time estimation procedure experimentally, and demonstrate that it yields near-optimal results for a wide range of noise levels and for various filtering methods.  相似文献

Gaston is a peer-to-peer large-scale file system designed to provide a fault-tolerant and highly available file service for a virtually unlimited number of users. Data management in Gaston disseminates and stores replicas of files on multiple machines to achieve the requested level of data availability and uses a dynamic tree-topology structure to connect replication schema members. We present generic algorithms for replication schema creation and maintenance according to file user requirements and autonomous constraints that are set on individual nodes. We also show specific data object structure as well as mechanisms for secure and efficient update propagation among replicas with data consistency control. Finally, we introduce a scalable and efficient technique improving fault-tolerance of the tree-topology structure connecting replicas.  相似文献

Multimaterial, microelectromechanical systems-based cantilever probes were developed for high-frequency magnetic field imaging. The basic configuration of the probe consists of a cantilever beam fabricated using surface micromachining and bulk micromachining techniques with dielectric silicon nitride and silicon oxide materials on a silicon wafer. A gold patterned metallization at the tip of the cantilever provides a source of eddy current heating due to the perpendicular component of the high-frequency magnetic field. This thermally absorbed power is converted to mechanical deflection by a multimaterial trilayer cantilever system. The deflection is measured with a beam-bounce optical technique employed in atomic force microscopy systems. We discuss the modeling, design, fabrication, and characterization of these field imaging probes  相似文献

The continuous-wave high power laser emission of Nd:GdVO/sub 4/ at the fundamental wavelength of 1.06 /spl mu/m and its 531-nm second harmonic obtained by intracavity frequency doubling with an LBO nonlinear crystal is investigated under pumping by diode laser at 808 nm (on the /sup 4/I/sub 9/2//spl rarr//sup 4/F/sub 5/2/ transition) and 879 nm (on the /sup 4/I/sub 9/2//spl rarr//sup 4/F/sub 3/2/ transition). It is shown that, in spite of a lower absorption at 879 nm, the infrared emission is comparable under these two wavelengths of pump. The green emission performances were, however, improved by the 879 nm pump: 5.1 W at 531 nm with M/sup 2/=1.46 and 0.31 overall optical-to-optical efficiency was obtained from a 3-mm-thick 1-at.% Nd:GdVO/sub 4/ laser medium and a 10-mm-long LBO nonlinear crystal in a Z-type cavity for 16.5 W pump power. In similar conditions, the maximum green power for the 808 nm pump was 4.4 W, with 0.26 overall optical-to-optical efficiency and M/sup 2/=3.40 beam quality; at this pump wavelength the green emission shows evident saturation for pump power in excess of 9.9 W. This behavior is connected with the enhanced heat generation under 809-nm pumping, as evidenced by the increased thermal lensing of the fundamental emission. A careful alignment of the laser enables emission almost free of chaotic intensity fluctuations.  相似文献
