The morphology of poly(phenylene sulfide), PPS, obtained as processed film has been studied using wide angle X-ray scattering (WAXS), scanning electron microscopy (SEM), and differential scanning calorimetry (DSC). A unique morphology has been identified in the original film. The spherulites are very small in size and possess a cylindrical symmetry with their lamellae oriented on the edge. In these processed PPS films, the a-axis of the lattice is preferentially aligned perpendicular to the film plane, while the b and c axes are predominantly in the film plane. In the PPS isothermally crystallized from the melted, original film, there is no such preferential orientation. Thermal analysis of the original and melt crystal-lized PPS shows that while the degree of crystallinity is about the same, the nature of the amorphous phase in the two materials is different. In the original film, we did not observe a heat capacity increment at a glass transition temperature by DSC, indicating that all of the amorphous phase belongs to the category of rigid amorphous phase. In the melt crystallized PPS, a distinct glass transition was seen, though only a portion of the amorphous phase becomes mobile at Tg. The differences in orientation and mobility of the amorphous phase in the original film compared to melt crystallized PPS are explained by the different thermal processing procedures used for the two materials.  相似文献

White spruce (Picea glauca) emits monoterpenes that function as defensive signals and weapons after herbivore attack. We assessed the effects of drought and methyl jasmonate (MeJA) treatment, used as a proxy for herbivory, on monoterpenes and other isoprenoids in P. glauca. The emission of monoterpenes was significantly increased after MeJA treatment compared to the control, but drought suppressed the MeJA-induced increase. The composition of the emitted blend was altered strongly by stress, with drought increasing the proportion of oxygenated compounds and MeJA increasing the proportion of induced compounds such as linalool and (E)-β-ocimene. In contrast, no treatment had any significant effect on the levels of stored monoterpenes and diterpenes. Among other MEP pathway-derived isoprenoids, MeJA treatment decreased chlorophyll levels by 40%, but had no effect on carotenoids, while drought stress had no impact on either of these pigment classes. Of the three described spruce genes encoding 1-deoxy-D-xylulose-5-phosphate synthase (DXS) catalyzing the first step of the MEP pathway, the expression of only one, DXS2B, was affected by our treatments, being increased by MeJA and decreased by drought. These findings show the sensitivity of monoterpene emission to biotic and abiotic stress regimes, and the mediation of the response by DXS genes.  相似文献

In this study, we addressed the question of whether an omic approach could genuinely be useful for biomarker profile analysis across different field sites with different physicochemical characteristics. We collected earthworms (Lumbricus rubellus) from seven sites with very different levels of metal contamination and prevailing soil type and analyzed tissue extracts by 1H nuclear magnetic resonance spectroscopy. Pattern recognition analysis of the data showed that both site- and contaminant-specific effects on the metabolic profiles could be discerned. Zinc was identified as the probable major contaminant causing a metabolic change in the earthworms. Individual sites could be resolved on the basis of NMR spectral profiles by principal component analysis; these site differences may also have been caused by additional abiotic factors such as soil pH. Despite an inevitable degree of confounding between site and contaminant concentrations, it was possible to identify metabolites which were correlated with zinc across all different sites. This study therefore acts as a proof of principle for the use of NMR-based metabolic profiling as a diagnostic tool for ecotoxicological research in polluted field soils.  相似文献

Pathogen infection stimulates the fatty acid (FA) metabolism and the production of pro-inflammatory derivatives of FA. Barramundi, Lates calcarifer, was fed on a diet rich in preformed long-chain (?C20) polyunsaturated fatty acids (LC-PUFA) from fish oil (FO), to compare with diets containing high levels of C18 precursors for LC-PUFA – stearidonic (SDA) and γ-linolenic acid (GLA) – from Echium plantagineum (EO), or rapeseed oil (RO) rich in α-linolenic acid (ALA), but a poor source of LC-PUFA and their precursors. After 6 weeks, when growth rates were similar amongst the dietary treatments, a sub-lethal dose of Streptococcus iniae was administered to half of the fish, while the other half were maintained unchallenged and were pair-fed with the infected fish. Under a disease challenge situation, the tissue FA depots depleted at 3 days post-infection (DPI) and were then restored to their previous concentrations at 7 DPI. During the infection period, EO fish had a higher content of n3 and n6 PUFA in their tissues, higher n3:n6 PUFA ratio and reduced levels of the eicosanoids, TXB2 and 6-keto-PGF, in their plasma compared with RO fish. Fish fed on FO and EO had a longer lasting and enduring response in their FA and eicosanoid concentrations, following a week of bacterial infection, compared with those fed on RO. EO, containing SDA and GLA and with a comparatively higher n3:n6 PUFA ratio, proved more effective than RO in compensating for immunity stress.  相似文献

The production of metabolites by microalgae is affected by environmental conditions in which they are living. The metabolic responses of two marine microalgae, Nitzschia closterium and Isochrysis zhangjiangensis, to a 3-day UVA-stress and 3-day UVA-recovery treatment were compared, based on their growth, fatty acid profiles and content of total carotenoids. When cultured under photosynthetically active radiation, coupled with UVA treatment, both microalgae underwent a significant increase in their growth during the UVA-recovery period compared to the control. The proportions of polyunsaturated fatty acids, including linoleic acid and eicosapentaenoic acid, as well as total carotenoids, were significantly increased in both microalgae, mainly in the UVA-stress period, but not the UVA-recovery period. The metabolic responses of the two microalgae to UVA treatment were species-dependent and could be utilised to produce microalgal biomass rich in polyunsaturated fatty acids and carotenoids for use as functional food ingredients.  相似文献

Non-thermal food preservation technology is based on the application of high pressures up to 600 MPa. Here we report a resonance Raman (RR) spectroscopic analysis of smoked salmon meat after high pressure processing. High quality spectra, which can be obtained even from packed salmon without spectral interference of the packing foil, allow determining pressure-dependent irreversible changes of the main RR-active components of salmon meat, astaxanthin and myoglobin/haemoglobin. High pressure-treatment causes a decrease of the relative RR intensities of astaxanthin as probed with 514 nm excitation which is in line with a slight attenuation of the originally intense red colour of the salmon meat. 413-nm excited RR spectra indicate a heterogeneous broadening of astaxanthin bands accompanied by the formation of deoxy-myoglobin or deoxy-haemoglobin. The results suggest that pressure-treatment facilitates the oxidative degradation of astaxanthin coupled to the reduction of metmyoglobin (methaemoglobin).  相似文献

A liquid chromatography–tandem mass spectrometry (LC–MS/MS) method was developed and validated for the simultaneous determination of six major ergot alkaloids, ergometrine, ergosine, ergotamine, ergocornine, ergokryptine and ergocristine, as well as their corresponding epimers in food and feed samples. The method involves extraction under alkaline conditions and subsequent clean-up by applying a simple and rapid liquid–liquid partitioning procedure prior to LC–MS/MS analysis. Evaluation of the method revealed good linearity, accuracy and precision. The limits of quantification varied from 0.1 to 1 μg/kg depending on the analyte and matrix. The average extraction and clean-up recoveries in different matrices were between 45 (only for ergometrine in biscuit) and 90%. The uncertainty associated with the analytical method was not higher than 51% and 30%, at concentration levels of 2.5 and 150 μg/kg respectively. Analyte epimerization proved to be minimal during the analytical procedure. The method has been successfully applied to the determination of ergot alkaloids in some Belgian food and feed commodities. Ergot alkaloids were found in 104 out of 122 samples investigated. Ergosine was the most frequently occurring alkaloid, while the highest levels were observed for ergotamine, ergocristine or ergosine, depending on the product type. The total alkaloid content in positive samples varied from 1 to 1145 μg/kg.  相似文献

1 Scope

During food processing, the Maillard reaction (МR) may occur, resulting in the formation of glycated proteins. Glycated proteins are of particular importance in food allergies because glycation may influence interactions with the immune system. This study compared native and extensively glycated milk allergen β‐lactoglobulin (BLG), in their interactions with cells crucially involved in allergy.

2 Methods and results

BLG was glycated in MR and characterized. Native and glycated BLG were tested in experiments of epithelial transport, uptake and degradation by DCs, T‐cell cytokine responses, and basophil cell degranulation using ELISA and flow cytometry. Glycation of BLG induced partial unfolding and reduced its intestinal epithelial transfer over a Caco‐2 monolayer. Uptake of glycated BLG by bone marrow–derived dendritic cells (BMDC) was increased, although both BLG forms entered BMDC via the same mechanism, receptor‐mediated endocytosis. Once inside the BMDC, glycated BLG was degraded faster, which might have led to observed lower cytokine production in BMDC/CD4+ T‐cells coculture. Finally, glycated BLG was less efficient in induction of degranulation of BLG‐specific IgE sensitized basophil cells.

3 Conclusions

This study suggests that glycation of BLG by MR significantly alters its fate in processes involved in immunogenicity and allergenicity, pointing out the importance of food processing in food allergy.  相似文献

Abstract : A process for the micropropagation of Amorphophallus rivierii (elephant yam or konjac) and for the extraction and purification of the glucomannan polysaccharide from fresh konjac plant corms has been developed. This process involves extraction with 2-propanol, which has the additional benefit of extracting carotenoids as a potentially valuable side-product. Starch granules with an unusually high and homogeneous gelatinisation temperature range are normally present in the corms, particularly immature ones, and this greatly reduces the strength of gels formed using the glucomannan. Therefore, the extraction process also involves the selective hydrolysis of the starch, by α- and β-amylases that have been specially selected for an absence of contaminating β-mannanase or β-glucanase activity that would depolymerise the glucomannan and render it nonfunctional. Bacillus lichenformis α-amylase was preferred. Using this process pure glucomannan could be extracted which, when mixed with K -carrageenan, forms a gel almost twice as strong as locust bean gum-K -carrageenan gels of the same concentration.  相似文献
