Naturally selected T-cell receptors (TCRs) are characterised by low binding affinities, typically in the range 1-100 microM. Crystal structures of syngeneic TCRs bound to peptide major histocompatibility complex (pMHC) antigens exhibit a conserved mode of binding characterised by a distinct diagonal binding geometry, with poor shape complementarity (SC) between receptor and ligand. Here, we report the structures of three in vitro affinity enhanced TCRs that recognise the pMHC tumour epitope NY-ESO(157-165) (SLLMWITQC). These crystal structures reveal that the docking mode for the high affinity TCRs is identical to that reported for the parental wild-type TCR, with only subtle changes in the mutated complementarity determining regions (CDRs) that form contacts with pMHC; both CDR2 and CDR3 mutations act synergistically to improve the overall affinity. Comparison of free and bound TCR structures for both wild-type and a CDR3 mutant reveal an induced fit mechanism arising from restructuring of CDR3 loops which allows better peptide binding. Overall, an increased interface area, improved SC and additional H-bonding interactions are observed, accounting for the increase in affinity. Most notably, there is a marked increase in the SC for the central methionine and tryptophan peptide motif over the native TCR.  相似文献

Sewage sludge ash (SSA) is a waste material obtained from the incineration of wastewater sludge. The physical, chemical and mineralogical characteristics of SSA, and the evaluation of its use in cement-based materials, are presented in this paper. Results show that SSA is composed of irregular grains having a high specific surface area and thus leading to a significantly high water demand. A fraction of the ash is rapidly soluble (sulfates, aluminum and silica) and can create new-formed products in presence of lime. SSA induces short delays of cement hydration, probably due to both minor elements of the ash and dilution effect. Compressive strengths of mortars containing 25% and 50% of SSA are always lower than those of reference mortars but it is shown that SSA has a long-term positive effect which might be related to a slight pozzolanic activity. The amount of elements leached from SSA mortars is slightly higher than from the reference mortar without residue but it remains of the same order of magnitude. An extensive literature review was performed in order to compare the residue used in this study with others included in the same category. This analysis highlighted the principal characteristics that must be taken into account to use SSA correctly in cement-based materials.  相似文献

The cell structure and properties of branched and linear polypropylene (PP) foams containing organically modified nanoclay and maleic anhydride grafted polypropylene (PPMA) have been thoroughly investigated. X-ray diffraction (XRD) and melt rheometry were used to identify the structure and linear viscoelastic properties of the nanocomposites, as well as the effectiveness of two different compatibilizers. These nanocomposites were used in injection molding to investigate their foamability and the influence of experimental conditions such as chemical foaming agent concentration, shot size, back pressure, injection speed, as well as melt temperature and different injection methods on the resulting cell structure of the foams. Quite different results were obtained with the linear and the branched PP. While the foamability of the branched PP was intrinsically good, that of the linear one could largely be improved by modifying its rheological properties and favoring nucleation through the addition of nanoclay. The effect of cell structure on the dynamic mechanical properties of the foams was also investigated using dynamic mechanical analysis (DMA). POLYM. ENG. The spreading phenomena of particles during thermal spraying are quite difficult to observe given the kinetics of the process. In this work, the splat formation of glass and alumina is theoretically compared, showing that glass transition and low-thermal conductivity yield a higher ratio between cooling and flattening times, which strongly modifies their spreading behavior. Wipe tests show that splash—splat transition temperature can be modified by the glass composition and its subsequent hydrodynamic properties. The detection of peculiar remaining objects, such as fibers and wavelets shows the possibility of “freezing” some phenomena that are totally unobservable with crystalline oxides, except with high-velocity observations.  相似文献

Two n‐3 polyunsaturated fatty acid (PUFA)‐rich diets differing in their chemical and physical forms were given to rats during 2 wk. Liposomes [phospholipids (PL) organized in bilayer structures] made from a natural marine lipid extract or a mixture of fats containing fish oil [similar fatty acids esterified in triacylglycerols (TAG)] were used. The influence of n‐3 PUFA dietary sources on plasma parameters, i.e. TAG, cholesterol and PL concentrations, was investigated. A similar hypotriglyceridemic effect of n‐3 PUFA from liposomes or fish oil was observed. In contrast, feeding rats with liposomes led to different PL and cholesterol patterns. In the plasma of rats fed liposomes, total cholesterol amounts were positively correlated with PL levels. Liposome and fish oil feedings caused a marked increase in the amounts of n‐3 PUFA, which occurred mainly at the expense of n‐6 PUFA. However, the enrichment in n‐3 PUFA in the different plasma lipid classes differed substantially when ingested in the form of fish oil or liposomes. These results were interpreted in terms of different lipid bioavailability and metabolic fate during the digestive steps and in the liver, with the dietary source.  相似文献

The preparation of cheap nitrogen-enriched materials with large adsorptive capacities and selectivity towards volatile organic compounds remains a challenge. Ammoxidation has been used to prepare nitrogen-enriched activated carbons using a demineralised Polish lignite. The lignite samples were demineralised by two different methods before nitrogen-enrichment by ammoxidation and physical activation in steam. The surface chemistry was investigated by elemental analysis, Boehm titration, infrared and XPS spectroscopies and adsorptive properties by a linear solvation energy relationship approach. Results show a quasi-total demineralisation and a higher reactivity towards nitrogen for the demineralised samples. The BET surface is also higher than for the non-demineralised lignite. Active carbons previously ammoxidated and demineralised are more interesting in terms of selective removal of gaseous pollutants.  相似文献

Tensile properties and crack propagation properties, especially critical strain energy release rate in mode I, GIC, have been used to investigate fracture properties of elastomers and their relationships with microstructure. These investigations were mainly based on a series of comparisons: first, the behaviour of polychloroprene rubber (CR), undergoing stress hardening due to strain induced crystallization (SIC) and oxidative crosslinking (OCL) was compared with that of chlorinated polyethylene (CPE), which undergoes SIC but not OCL, and with a polyurethane based on hydroxyl terminated polybutadiene (PU) which undergoes OCL but not SIC. Comparisons were also made on CR between fracture behaviour at ambient temperature, where SIC occurs and at 100 °C where there is no SIC. Finally, oxidative crosslinking was used to vary in a continuous way the crosslink density in CR and PU, in order to evaluate the role of crosslinking in fracture behaviour.  相似文献

A novel RuII(arene) theranostic complex is presented. It is based on a 1,4,7,10‐tetraazacyclododecane‐1,4,7,10‐tetraacetic acid macrocycle bearing a triarylphosphine and can be tracked in vivo by using the γ emission of 153Sm atoms. Notably, the heteroditopic ligand can be selectively metalated with ruthenium at the phosphorus atom despite the presence of other functionalities that are prone to metal coordination. Subsequent labeling with radionuclides such as 153Sm can then be performed easily. The resulting heterobimetallic complex exhibits favorable solubility and stability properties in biologically relevant media. It also shows in vitro cytotoxicity in line with that expected for this type of metallodrug, and is nontoxic to the organism as a whole. As a proof of concept, initial studies in healthy mice were performed to obtain information about the uptake, biodistribution, and excretion of the radiolabeled complex.  相似文献

Inorganic fullerene-like molybdenum disulfide (IF-MoS2) nanoparticles are known to exhibit great friction and wear-reducing abilities in severe boundary lubrication regimes, when added to a base oil alone. Their use in fully formulated lubricants was investigated in this study, and the tribological benefits attributed to the IF-MoS2 nanoparticles were found to be lost in the presence of dispersants. Various experimental techniques were used on three reference oils (base oil containing only IF-MoS2, only dispersants and both IF-MoS2 and dispersants) in order to understand the effect of succinimide-based dispersants on the three phases needed for effective nanoparticle-based lubrication, namely (1) the passing of the nanoparticles through the contact (2) the exfoliation of the IF-MoS2 inside the contact and (3) the adhesion of the released MoS2 platelets on the friction surfaces. The dispersants were shown to improve the dispersion of the nanoparticles in the oil by reducing their agglomeration, but prevented the adhesion of a low-friction MoS2 tribofilm on the steel surfaces. In-situ contact visualization revealed that the well-dispersed nanoparticles passed through the contact and exfoliated nanoparticles were observed after tribological testing. These results imply that nanoparticle dispersion itself does not seem to be an issue concerning nanoparticle effectiveness, even though the reduced agglomerate size and inertia may have affected nanoparticle flow near the contact, as well as entrapment and exfoliation conditions inside the contact. The use of succinimide-based dispersants may, however, have affected the tribochemistry of the contact, by an excessive adsorption on the steel surfaces and/or by encapsulating the released MoS2 platelets, preventing tribofilm adhesion. A balance was finally found between nanoparticle dispersion and friction reduction, but for very low dispersant concentrations and after a running-in period. The role of succinimide-based dispersants and their effect on nanoparticle lubrication were discussed in the light of these results.  相似文献

When coatings are strongly bonded to their substrates it is often difficult to measure the adhesion values. The proposed method, which is suggested naming “silver print test”, consists in covering the central part of the samples with a thin layer of silver paint, before coating. The process used for testing this new method was the Air plasma spraying (APS), and the materials used were alumina coatings on C35 steel substrates, previously pre-oxidized in CO2. The silver painted area was composed of small grains that did not oxidize but that significantly sintered during the APS process. The silver layer reduced the surface where the coating was linked to the substrate, which allowed its debonding, using the classical adhesion test ASTM C633-13, while the direct use of this test (without silver painting) led to ruptures inside the glue used in this test. The numerical modelling, based on the finite element method with the ABAQUS software, provided results in good agreement with the experimental measurements. This concordance validated the used method and allowed accessing to the values of adherence when the experimental test ASTM C633-13 failed, because of ruptures in the glue. After standardization, the “silver print test” might be used for other kinds of deposition methods, such as PVD, CVD, PECVD.  相似文献
