BACKGROUND: To prevent ischemic complications during coronary bypass grafting on the beating heart, a nonocclusive distal anastomosis technique is needed. One recently developed nonocclusive technique requires apposition of the intima of the graft to the adventitia of the recipient artery, in contrast to current surgical practice, which dictates apposition of both intimas. METHODS: To compare the sole effect of intima-adventitia apposition (n = 18) versus traditional intima-intima apposition (n = 18), we investigated radiolabeled platelet deposition and histomorphologic aspects of vascular wall healing quantitatively in a porcine carotid artery bypass graft model. Both groups were evaluated at 2 hours, 2 days, or 4 weeks. RESULTS: Within the first 2 hours, 3 of 6 pigs with intima-adventitia apposition exhibited cyclic flow reductions as a result of massive mural thrombosis. After intima-adventitia apposition, the number of deposited platelets was significantly higher compared with intima-intima apposition, 147.1 +/- 73.0 x 10(6) and 4.6 +/- 1.0 x 10(6) platelets/cm2 (mean +/- standard error of the mean), respectively (p = 0.03). At 2 days, the suture line was covered with small mural thrombi, whereas no thrombi were found after intima-intima apposition. At 4 weeks, intimal hyperplasia at heel and toe was not significantly different from that with intima-intima apposition. CONCLUSIONS: Despite thrombotic phenomena in the early phase, intima-adventitia apposition yielded a patent anastomosis with a small intimal hyperplasia response.  相似文献

Single‐stranded model oligodeoxyribonucleotides, each containing a single protonatable base—cytosine, adenine, guanine, or 5‐methylcytosine—centrally located in a background of non‐protonatable thymine residues, were acid‐titrated in aqueous solution, with UV monitoring. The basicity of the central base was shown to depend on the type of the central base and its nearest neighbours and to rise with increasing oligonucleotide length and decreasing ionic strength of the solution. More complex model oligonucleotides, each containing a centrally located 5‐methylcytosine base, were comparatively evaluated in single‐stranded and double‐stranded form, by UV spectroscopy and high‐field NMR. The N3 protonation of the 5‐methylcytosine moiety in the double‐stranded case occurred at much lower pH, at which the duplex was already experiencing general dissociation, than in the single‐stranded case. The central guanine:5‐methylcytosine base pair remained intact up to this point, possibly due to an unusual alternative protonation on O2 of the 5‐methylcytosine moiety, already taking place at neutral or weakly basic pH, as indicated by UV spectroscopy, thus suggesting that 5‐methylcytosine sites in double‐stranded DNA might be protonated to a significant extent under physiological conditions.  相似文献

The inactivation of one X chromosome in females is normally random with regard to which X is inactivated. However, exclusive or almost-exclusive inactivation of one X may be observed in association with some X-autosomal rearrangements, mutations of the XIST gene, certain X-linked diseases, and MZ twinning. In the present study, a methylation difference near a polymorphism in the X-linked androgen-receptor gene was used to investigate the possibility that nonrandom X inactivation is increases in fetuses and newborns that are associated with confined placental mosaicism (CPM) involving an autosomal trisomy. Extreme skewing was observed in 7 (58%) of 12 cases with a meiotic origin of the trisomy, but in none of 10 cases examined with a somatic origin of the trisomy, and in only 1 (4%) of 27 control adult females. In addition, an extremely skewed X-inactivation pattern was observed in 3 of 10 informative cases of female uniparental disomy (UPD) of chromosome 15. This may reflect the fact that a proportion of UPD cases arise by "rescue" of a chromosomally abnormal conceptus and are therefore associated with CPM. A skewed pattern of X inactivation in CPM cases is hypothesized to result from a reduction in the size of the early-embryonic cell pool, because of either poor early growth or subsequent selection against the trisomic cells. Since approximately 2% of pregnancies detected by chorionic villus sampling are associated with CPM, this is likely a significant contributor to both skewed X inactivation observed in the newborn population and the expression of recessive X-linked diseases in females.  相似文献

An in situ corrosion study of austenitic stainless steel 08CH18N10T in high temperature water was performed. The material under study is used in the construction of steam generator of PWR (pressurized water reactor) nuclear power stations and is similar to AISI 321 stainless steel.In situ 300-h tests were performed under autoclave conditions at 280 °C and 8 MPa and consisted of impedance measurements, polarization measurements and electrochemical noise measurements. The experiments were performed in deionised water with the pH adjusted to 9.5, in the presence/absence of chlorides. An additional modification of corrosivity was achieved by changing oxygen concentration.A detailed analysis of the impedance data is presented identifying in the impedance spectra contributions of oxide, corrosion reaction, double layer and diffusion process. A good agreement was found between corrosion data from electrochemical impedance spectroscopy (EIS) and that from electrochemical noise (EN) measurements. It was confirmed that the oxide response cannot be attributed to the overall oxide layer but only to the part corresponding to the space charge layer, thus indicating the semi-conductive character of the oxide.  相似文献

Light cycle oil (LCO) and waste sunflower cooking oil (WSO) were co‐processed with the aim of obtaining more environmentally friendly fuels. Partial hydrogenation of naphthalene was also investigated as a model reaction. Commercial NiW/SiO2‐Al2O3, as a reference catalyst, and NiW/(pseudoboehmite + SBA‐15), as a new research catalyst, were tested. Liquid products were analyzed by simulated distillation, elemental analysis, and FTIR spectroscopy. Elemental analysis indicated higher efficiency of the research catalyst in hydrodesulfurization, hydrodenitrogenation, and hydrodeoxygenation of pure LCO and mixed feedstock containing WSO. Reactions with pure WSO resulted in less sulfur leaching into the product and a lower degree of deoxygenation compared with the commercial catalyst.  相似文献

Short fiber reinforcement is a suitable way to improve the tribological properties of elastomers. However, rubbers products are often exposed to highly dynamic mechanical loadings. Hereby it is crucial to study the change in dynamic behavior due to the addition of short fibers. Therefore, these properties were investigated in terms of dynamic mechanical thermal analysis, heat build‐up (HBU), and fatigue crack growth resistance under cyclic loadings for two different rubber compounds. A peroxide cured ethylene–propylene–diene rubber (EPDM) and a sulfur cured natural rubber (NR) compound were chosen and reinforced with two types of short aramid fibers. It was found that the short fibers could contribute to the improvement in the crack growth resistance, HBU, and the dynamic mechanical behavior of the composites depending on the testing conditions. Hybrid magnetic drug nanocarriers are prepared via a self-assembly process of poly(methacrylic acid)-graft-poly(ethyleneglycol methacrylate) (p(MAA-g-EGMA)) on growing iron oxide nanocrystallites. The nanocarriers successfully merge together bio-repellent properties, pronounced magnetic response, and high loading capacity for the potent anticancer drug doxorubicin (adriamicin), in a manner not observed before in such hybrid colloids. High magnetic responses are accomplished by engineering the size of the magnetic nanocrystallites (~13.5 nm) following an aqueous single-ferrous precursor route, and through adjustment of the number of cores in each colloidal assembly. Complementing conventional magnetometry, the magnetic response of the nanocarriers is evaluated by magnetophoretic experiments providing insight into their internal organization and on their response to magnetic manipulation. The structural organization of the graft-copolymer, locked on the surface of the nanocrystallites, is further probed by small-angle neutron scattering on single-core colloids. Analysis showed that the MAA segments selectively populate the area around the magnetic nanocrystallites, while the poly(ethylene glycol)-grafted chains are arranged as protrusions, pointing towards the aqueous environment. These nanocarriers are screened at various pHs and in highly salted media by light scattering and electrokinetic measurements. According to the results, their stability is dramatically enhanced, as compared to uncoated nanocrystallites, owing to the presence of the external protective PEG canopy. The nanocarriers are also endowed with bio-repellent properties, as evidenced by stability assays using human blood plasma as the medium.  相似文献
