The drugs based on platinum metals represent one of the oldest, but also one of the most effective groups of chemotherapeutic agents. Thanks to many clinical studies it is known that resistance of tumor cells to drugs is a frequent cause of chemotherapy failure. With regard to platinum based drugs, multidrug resistance can also be connected with increased expression of low-molecular weight protein metallothionein (MT). This study aimed at investigating the interactions of MT with cisplatin or carboplatin, using the adsorptive transfer technique coupled with differential pulse voltammetry Brdicka reaction (AdTS DPV Brdicka reaction), and a comparison of in vitro results with results obtained in vivo. The results obtained from the in vitro study show a strong affinity between platinum based drugs and MT. Further, we analyzed extracts of neuroblastoma cell lines treated with cisplatin or carboplatin. It is clear that neuroblastoma UKF-NB-4 cisplatin-resistant and cisplatin-sensitive cell lines unlikely respond to the presence of the platinum-based cytostatics cisplatin and carboplatin. Finally, we determined the level of MT in samples from rabbits treated with carboplatin and patients with retinoblastoma treated with the same drug.  相似文献

ABSTRACT:  The objective was to examine the effects of potassium lactate (KL) and raw-meat blend on the shelf-life of fresh-pork-sausage patties in lighted and unlighted display. Four fresh-pork-sausage blends (70% prerigor meat, 30% fresh-pork trim; 80%/20%; 90%/10%; and 100%/0%) were formulated ( n = 3) with 0% and 3% KL, and patties were displayed for 10 d at 4 °C in lighted display or for 7 d in unlighted display followed by 3 d in lighted display. Patties with KL had lower aerobic plate counts (APC), more stable color (greater a* and chroma, less visual discoloration), and less lipid oxidation ( P < 0.05) during display than did patties without KL. Patties in the light for 10 d were more discolored and had more lipid oxidation ( P < 0.05) than those in the dark for 7 d followed by the light for 3 d. Patties with KL in the light for 10 d had similar color ( P > 0.05) to patties without KL that were in the dark for the first 7 d. Adding KL resulted in similar color stability ( P > 0.05) among patties made from 80, 90, and 100% prerigor meat and greater color stability ( P < 0.05) than similarly formulated patties without KL. By using 3% KL, fresh-pork-sausage patty color stability was maximized, APC and lipid oxidation were minimized, and formulation costs can be decreased by using less-expensive fresh-pork trim for up to 20% of the blend. Adding KL also minimized the effects of display lighting on myoglobin and lipid oxidation.  相似文献

In this paper, we draw upon digital 2D and 3D modelling techniques to analyse two sixteenth-century visual records of the city of Istanbul at that time: the Prospect of Constantinople by Melchior Lorichs (or Lorck) held at the University of Leiden, and the view of the Hippodrome and Hagia Sophia in the Freshfield Folio held at Trinity College, Cambridge. We propose that, through digitally-based analysis of these sources that matches aspect ratios and viewpoints, it is possible to extract new evidence for the topography and structures of the city during this period.  相似文献

The static fatigue of SiC-based fiber bundles and single fibers has been examined in previous papers, with emphasis placed on the analysis of the stress–rupture time data, and on the modelling of delayed failure from slow crack growth. The present paper investigates the oxidation of the fibers during static fatigue, at temperatures in the intermediate temperature range (500°–800°C). Two oxidation-induced phenomena have been evidenced: the formation of a thin silica film at the surface of fibers and the delayed failure of fiber bundles and single filaments. The stress–rupture time data are interpreted with respect to the chemical and structural characteristics of fibers, and to the oxide film growth rate. The structural analysis of the fibers was carried out using scanning electron microscopy and Auger electron spectroscopy. Delayed failure was found to result from slow crack propagation from surface defects, as a result of the consumption of the free carbon at grain boundaries and the local stresses induced by the SiC→SiO2 transformation at the crack tip. The respective contributions of these phenomena to static fatigue are discussed.  相似文献

In this study, the effects of cloud inhomogeneity on microwave rain rate retrievals are investigated. A single-channel (85 GHz) empirically based algorithm using a neural network approach is presented. The objective is to correct the beam-filling error (BFE), that might occur because of the inherent variability within coarse microwave pixels, with subpixel information. To this aim, we used the Tropical Rainfall Measuring Mission passive microwave, thermal infrared and radar data. The integration of spatial information into the retrieval algorithm enables us to partially overcome the BFE. We use two parameters which characterize the horizontal cloud inhomogeneity within the microwave radiometer field of view, and we add them to simulated brightness temperatures as inputs of the neural network algorithm. The first one is the cloud fraction derived from infrared measurement, and the second corresponds to the fraction of the rainy area derived from radar measurements. The output rain rates were validated using the Precipitation Radar data. It was found that adding cloud fraction of microwave pixels, can lead to more accurate retrievals. Instantaneous precipitation estimates demonstrated correlations of /spl sim/0.6-0.7 and /spl sim/0.7-0.8 with radar-derived rain rates, for ocean and land retrievals respectively. In spite of the problem inherent in deriving the cloud (or rain) fraction, the initial validation results presented in this study are reasonably encouraging and show the advantage of utilizing the information from different sensors in order to optimize the retrieval of rainfall.  相似文献

Fault-tolerance analysis reveals possible system behavior under the influence of faults. Such analysis is essential for satellites where faults might be caused by space radiation and autonomous recovery is needed. In this paper we present a statistical simulation approach for fault-tolerance analysis of satellite On-Board Computers (OBCs) that are based on Commercial Off-The-Shelf (COTS) components. Since the logic level of COTS electronics is unknown to satellite designers, a new higher-level fault-tolerance analysis is required. We propose such technique that relies on OBC modeling and fault modeling, based on the modeling principle of Single-Event Upsets (SEUs). For the first time we can compare the efficiency of fault-tolerance techniques implemented in software and Field-Programmable Gate Array (FPGA). In addition, our approach enables to analyze system fault-tolerance at early development stages. In a case study the approach is applied to an OBC with a Microsemi SmartFusion SoC, that executes a satellite attitude control algorithm. The gained statistical simulation results enabled 50% reduction in the hardware overhead of the implemented memory scrubbing technique without loss in fault-tolerance. Our method revealed critical fault-tolerance drawbacks of the initial system design that could have lead to satellite mission failure.  相似文献

The synthesis of phytochelatins is catalyzed by γ-Glu-Cys dipeptidyl transpeptidase called phytochelatin synthase (PCS). Aim of this study was to suggest a new tool for determination of phytochelatin synthase activity in the tobacco BY-2 cells treated with different concentrations of the Cd(II). After the optimization steps, an experiment on BY-2 cells exposed to different concentrations of Cd(NO3)2 for 3 days was performed. At the end of the experiment, cells were harvested and homogenized. Reduced glutathione and cadmium (II) ions were added to the cell suspension supernatant. These mixtures were incubated at 35 °C for 30 min and analysed using high performance liquid chromatography coupled with electrochemical detector (HPLC-ED). The results revealed that PCS activity rises markedly with increasing concentration of cadmium (II) ions. The lowest concentration of the toxic metal ions caused almost three fold increase in PCS activity as compared to control samples. The activity of PCS (270 fkat) in treated cells was more than seven times higher in comparison to control ones. Km for PCS was estimated as 2.3 mM.  相似文献
