A red Rioja wine was aged in barrels made of Spanish oak wood for 21 months. The evolutions of colour percentage intensity, families of phenolic compounds and low molecular weight phenolic compounds were studied in these wines and compared with those of the same wine aged in barrels made of French and American oak. The analysis of chromatic parameters and total anthocyanins indicates that the wines aged in Spanish and French oak wood barrels have similar chromatic characteristics, but are significantly different to those of wines aged in barrels made of American oak wood, indicating a different degree of modification of the colour. The ageing process also had an important influence on the low molecular weight polyphenols composition of wine. The evolution of these components allowed the production of wines with different characteristics, in relation to the type of wood used in barrel making process. On the other hand, Spanish oak wood can be considered suitable for barrel production for quality wines, since a wine aged in barrels made of Spanish oak wood showed similar and intermediate characteristics to those of the same wine aged in French and American oak woods. 相似文献

A new screening method for the detection and identification of GMO, based on the use of multiplex PCR followed by microarray, has been developed and is presented. The technology is based on the identification of quite ubiquitous GMO genetic target elements first amplified by PCR, followed by direct hybridisation of the amplicons on a predefined microarray (DualChip® GMO, Eppendorf, Germany). The validation was performed within the framework of a European project (Co-Extra, contract no 007158) and in collaboration with 12 laboratories specialised in GMO detection. The present study reports the strategy and the results of an ISO complying validation of the method carried out through an inter-laboratory study. Sets of blind samples were provided consisting of DNA reference materials covering all the elements detectable by specific probes present on the array. The GMO concentrations varied from 1% down to 0.045%. In addition, a mixture of two GMO events (0.1% RRS diluted in 100% TOPAS19/2) was incorporated in the study to test the robustness of the assay in extreme conditions. Data were processed according to ISO 5725 standard. The method was evaluated with predefined performance criteria with respect to the EC CRL method acceptance criteria. The overall method performance met the acceptance criteria; in particular, the results showed that the method is suitable for the detection of the different target elements at 0.1% concentration of GMO with a 95% accuracy rate. This collaborative trial showed that the method can be considered as fit for the purpose of screening with respect to its intra- and inter-laboratory accuracy. The results demonstrated the validity of combining multiplex PCR with array detection as provided by the DualChip® GMO (Eppendorf, Germany) for the screening of GMO. The present study aimed to demonstrate the effectiveness of the 5s rDNA gene for the identification of commercially-valuable species of cephalopod belonging to the families Loliginidae and Ommastrephidae. Our results demonstrate distinct banding patterns in each of the six species sampled (Loligo surinamensis; Loligo sanpaulensis; Lolliguncula brevis; Sepiotheuthis sepioidea; Ornithoteuthis antillarum; Illex argentinus), as well as diagnostic traits at the genus and probably family levels. The results emphasize the efficiency of the 5s rDNA marker as a low-cost and rapid forensic technique, which not only permits the identification of species, but also differentiation of members of the Loliginidae and Ommastrephidae. 相似文献

Nowadays it is essential to test new preservation and decontamination procedures using naturally occurring chemicals against important pathogenic bacteria in meat. We tested the antimicrobial effect of rosemary extracts and the bacteriocin nisin against Campylobacter jejuni at a low storage temperature (8 °C) with or without short-term pre-freezing. The antimicrobial effect of rosemary extract was four times greater in laboratory media than in chicken meat juice. Furthermore, low temperature storage conditions prolonged the survival of C. jejuni in chicken meat juice. Nisin, with an approximately 1.0 log reduction was neither effective alone nor in combination with the extract. Pre-freezing with plant extract addition proved to be effective treatment by more than 3.0 log reduction in 48 h. The results in chicken meat food model again showed the synergistic effect of freezing and plant extract antimicrobial activity. As the combination of pre-freezing and plant extract treatment reduced the cell number by more than 2.0 log reduction, studies should be conducted to further evaluate this promising treatment for Campylobacter reduction in the poultry meat supply. 相似文献

For use in gas-fired thermophotovoltaic systems, a selective emitter made from Yb2O3 foam ceramic has been developed. This foam ceramic is mechanically stable, and FTIR spectroscopy showed that 10% of the radiation power emitted by the foam can be converted by Si photocells. The thermal and thermal-shock stability of Yb2O3 foam ceramic was analyzed. The foam passed 200 heating/cooling cycles without major damage. Tubes were manufactured from this material and tested in a thermophotovoltaic demonstration system. An electrical power of 86 W was achieved at a thermal power of 16 kW. Using a simulation model, the potential efficiency of a thermophotovoltaic system based on our technology applied for the conversion of concentrated solar radiation was estimated. 相似文献

The genetic programming (GP) paradigm, which applies the Darwinian principle of evolution to hierarchical computer programs,
has been applied with breakthrough success in various scientific and engineering applications. However, one of the main drawbacks
of GP has been the often large amount of computational effort required to solve complex problems. Much disparate research
has been conducted over the past 25 years to devise innovative methods to improve the efficiency and performance of GP. This
paper attempts to provide a comprehensive overview of this work related to Canonical Genetic Programming based on parse trees
and originally championed by Koza (Genetic programming: on the programming of computers by means of natural selection. MIT,
Cambridge, 1992). Existing approaches that address various techniques for performance improvement are identified and discussed

The feasibility of performing SI-traceable carbon isotope amount ratio measurements following conversion of carbon into CF4 was studied. A procedure for the direct fluorination of carbon with elemental fluorine was developed, and the conversion step was checked for losses, blank contributions, and the absence of systematic isotope effects. Gas chromatography was used to identify and quantify the gaseous fluorination products and to isolate CF4 from byproducts. After fluorination of graphite carbon, CF4 and perfluoroalkanes with up to six carbon atoms were observed as reaction products. Within an uncertainty of 10%, the graphite carbon was fully recovered in the gaseous carbon fluorides, with the main product being CF4 (80-90%) and C2F6 as the major byproduct. The fluorination and GC procedures were found to introduce an alteration not bigger than 0.03 +/- 0.04/1000 on the isotopic composition of CF4. Carbon blank contributions introduced during the fluorination procedure were below 0.5% relative to a typical sample of 4 mg of carbon. For two of the materials investigated, the carbon isotope ratios measured on a differential mass spectrometer were reproducible within a standard deviation of approximately 0.1/1000 for several individual fluorinations. For these materials, the developed fluorination procedure is a straightforward process, which can be used as a foundation to establish SI-traceable measurements of carbon isotope amount ratios. However, for the third graphite material the formation of byproducts (C2F6-C6F14) was found to induce significant isotopic fractionation. 相似文献

The performance of an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) based on a monoclonal antibody (mAb) for ochratoxin A (OTA) detection was evaluated in a comparative study with high-performance liquid chromatography (HPLC) analysis using 68 freshly harvested coffee samples from the North of Paraná State, Brazil. The anti-OTA mAb showed high specificity and low cross-reactivity with OTA analogues (OTB and OTalpha), but cross-reacted with OTC. This ic-ELISA showed a detection limit of 3.75 ngg-1 sample, when compared to 0.80 ngg-1 by HPLC, with an ic-ELISA/HPLC correlation coefficient of 0.90. As regards OTA analysis of these coffee samples, natural contamination was detected in 10 samples (14.7%) by both methods, where the ic-ELISA values (range 3.9-7.3 ngg-1) were 1.1 to 1.6-fold higher than HPLC data (2.7-4.7 ngg-1). Five samples (7.4%) were OTA positive (range 0.84-1.30 ngg-1) only by HPLC assay, probably due to the higher detection limit reached by ic-ELISA. OTA was undetectable in 53 samples (77.9%) by both methods, while all positive samples (range 0.84-7.30 ngg-1) showed OTA levels lower than 8 ngg-1 (maximum limit recommended by the European Union). The matrix interference of green coffee was minimized by dilution of sample extracts before carrying out the ELISA assay. This mAb-based ic-ELISA can be effectively applied for OTA screening in coffee, because it is simple, sensitive and sample preparation is easy. 相似文献
