The results of specific percolation tests are expressed in terms of field saturated hydraulic conductivity (Kfs) of the soil. The specific tests comprise the Irish SR 6 and the UK BS 6297 standard tests and the inversed auger hole and square hole tests employed for the design of land drainage. Percolation times from these tests are converted to Kfs values using unit gradient theory and the Elrick and Reynolds (Soil Sci. 142(5) (1986) 308) model which takes into account gravitational, pressure head and matric potential gradients. Kfs is then expressed as the inverse of the percolation rate times a constant, in this way the percolation rate can be directly related to Kfs of the soil. A plot of Kfs against percolation rate for the Irish SR 6 and the UK BS 6297 standard tests is asymptotic at Kfs values less than 0.2 m/d and greater than 0.8 m/d. This behaviour creates difficulty in setting limits for percolation rates in standards. Curves are provided which enable Kfs values to be read off from percolation tests without the restrictions of head range currently enforced, for example in the Irish SR 6 and BS 6297 standards. Experimental measurements of percolation rates and Kfs were carried out on two sands in the laboratory and in the field on two soils. Kfs of these four materials was also measured using a tension infiltrometer and the Guelph permeameter. The saturated hydraulic conductivities (Ks) of the sands were also estimated in a falling head laboratory apparatus and by the Hazen formula. There was good agreement between the different tests for Kfs on each material. Because percolation time continued to increase significantly in consecutive tests in the same test hole while Kfs became constant, the latter is a better measure of the suitability of soils for percolation.  相似文献

We review recent work that shows that, for several kinds of devices built by several different manufacturers, MOS and bipolar device radiation response can change significantly with pre-irradiation elevated temperature stress and/or with aging time after device fabrication or packaging. The primary effects that have been observed are related to changes in interface-trap charge buildup, but some effects are also noted for charge trapped in the oxide. On the basis of a wide array of experimental data and first-principles calculations, these changes are attributed to the motion and reactions of hydrogenous species, e.g., protons and water. Similar reactions with hydrogen can also affect the reliability of MOS devices and integrated circuits, as illustrated here for the case of negative bias-temperature instability for high-κ alternative gate dielectrics. Heating the device accelerates some kinds of aging effects, but reverses the effects of others. These observations are consistent with a dual role for hydrogen in improving or degrading device performance, depending on its particular incorporation in the device and subsequent motion and reactions. These results emphasize that measurements used to provide estimates of MOS radiation response and long-term reliability can vary significantly with time. In some cases, typical margins built into radiation response and reliability tests may be inadequate to account for aging-related changes in device performance.  相似文献

The present experiment examined whether ondansetron, co-administered with continuous cocaine, would block the down regulation of accumbens 5-HT3 receptors. Rats were exposed to a 14-day pretreatment regimen that involved the continuous infusion of 40 mg kg(-1) day(-1) cocaine or 0.9% saline via a subcutaneously implanted osmotic minipump. In addition to the continuous cocaine or saline administration, all subjects received daily subcutaneous (s.c.) injections of either vehicle or 0.1 mg kg(-1) ondansetron for the entire 14-day pretreatment regimen. The rats were then withdrawn from this pretreatment regimen for seven days, and slices from the nucleus accumbens obtained. The slices were perfused with 25 mM K+ in the absence and presence of 0, 12.5, 25, or 50 microM m-Chlorophenyl-biguanide HCl (mCPBG). The efflux samples were assayed for dopamine content by high pressure liquid chromatography (HPLC) with electrochemical detection. Continuous cocaine administration significantly attenuated the ability of mCPBG to facilitate K+-induce dopamine overflow compared to saline control rats. In addition, the rats that received ondansetron and cocaine during the 14-day pretreatment period, the ability of mCPBG to enhance K+ stimulated dopamine release was not significantly different from the saline control subjects. For all groups except the cocaine alone group, the effects of mCPBG on K+ stimulated dopamine release were Ca2+ dependent, suggesting that these effects are receptor mediated. These results suggest that continuous cocaine administration functionally down-regulates 5-HT3 receptors in the nucleus accumbens, and that this down-regulation can be blocked by chronic ondansetron administration. Hence, a functional down regulation of accumbens 5-HT3 receptors represents a significant contribution to the tolerance induced by continuous cocaine administration.  相似文献

Abstract To ensure prompt response by real-time air monitors to an accidental release of toxic aerosols in a workplace, safety professionals should understand airflow patterns. This understanding can be achieved with validated computational fluid dynamics (CFD) computer simulations, or with experimental techniques, such as measurements with smoke, neutrally buoyant markers, trace gases, or trace aerosol particles. As a supplementary technique to quantify airflows, the use of a state-of-the art, three-dimensional sonic anemometer was explored. This instrument allows for the precise measurements of the air-velocity vector components in the range of a few centimeters per second, which is common in many indoor work environments. Measurements of air velocities and directions at selected locations were made for the purpose of providing data for characterizing fundamental aspects of indoor air movement in two ventilated rooms and for comparison to CFD model predictions. One room was a mockup of a plutonium workroom, and the other was an actual functioning plutonium workroom. In the mockup room, air-velocity vector components were measured at 19 locations at three heights (60, 120 and 180 cm) with average velocities varying from 1.4 cm s?1 to 9.7 cm s?1. There were complex flow patterns observed with turbulence intensities from 39% up to 108%. In the plutonium workroom, measurements were made at the breathing-zone height, recording average velocities ranging from 9.9 cm s?1 to 35.5 cm s?1 with turbulence intensities from 33% to 108%.  相似文献

Monosaccharide composition was determined in apolipoprotein B-48 (apoB) of chylomicrons of rat mesenteric lymph. Chylomicrons were separated into three fractions based on density. Triglyceride and apolipoprotein content were determined in each. ApoB was isolated and quantified using precipitation with isopropanol. Chylomicrons were collected in lymph under normal conditions, and with Poloxalene 2930 when chylomicron secretion was inhibited. Most of the triglyceride was carried in the least dense fraction, while the highest apoB content was in the most dense fraction under normal conditions. Mannose and galactosamine contents of apoB were similar in all fractions while contents of both glucosamine and galactose were highest in the least dense fraction. When chylomicron secretion was inhibited by Poloxalene, the amount of triglyceride recovered in the least dense fraction was significantly reduced. Despite the inhibition of lipid transport in the least dense fraction of chylomicrons by Poloxalene, there was little change in apoB recoveries and in the relative content of various monosaccharides in the apoB from each of the three fractions as compared to results obtained during lipid absorption under normal conditions. In conclusion, carbohydrate composition of apoB of chylomicrons is heterogeneous and varies with chylomicron density.  相似文献

The CD94/NKG2 receptors expressed by subpopulations of NK cells and T cells have been implicated as receptors for a broad range of both classical and nonclassical HLA class I molecules. To examine the ligand specificity of CD94/NKG2 proteins, a soluble heterodimeric form of the receptor was produced and used in direct binding studies with cells expressing defined HLA class I/peptide complexes. We confirm that CD94/NKG2A specifically interacts with HLA-E and demonstrate that this interaction is dependent on the association of HLA-E with peptide. Moreover, no interaction between CD94/NKG2A and classical HLA class I molecules was observed, as assayed by direct binding of the soluble receptor or by functional assays using CD94/NKG2A+ NK cells. The role of the peptide associated with HLA-E in the interaction between HLA-E and CD94/NKG2A was also assessed. All class I leader sequence peptides tested bound to HLA-E and were recognized by CD94/NKG2A. However, amino acid variations in class I leader sequences affected the stability of HLA-E. Additionally, not all HLA-E/peptide complexes examined were recognized by CD94/NKG2A. Thus CD94/NKG2A recognition of HLA-E is controlled by peptide at two levels; first, peptide must stabilize HLA-E and promote cell surface expression, and second, the HLA-E/peptide complex must form the ligand for CD94/NKG2A.  相似文献

A tissue culture bilayer system that mimics some aspects of early alveolar infection by Mycobacterium tuberculosis was developed. This model incorporates human lung epithelial type II pneumocyte (A549) (upper chamber) and endothelial cell (lower chamber) layers separated by a microporous membrane. This construction makes it possible to observe and quantify the passage of bacteria through the two layers, to observe the interaction of the bacteria with the various cell types, and to examine the basic mechanisms of immune cell recruitment to the site of infection. After 10(7) organisms were added to the upper chamber we microscopically observed large numbers of bacteria attached to and within the pneumocytes and we determined by viable-cell counting that a small percentage of the inoculum (0.02 to 0.43%) passed through the bilayer into the lower chamber. When peripheral blood mononuclear cells were added to the lower chamber, microscopic examination indicated a migration of the mononuclear cells through the bilayer to the apical surface, where they were seen associated with the mycobacteria on the pneumocytes. The added complexity of the bilayer system offers an opportunity to define more precisely the roles of the various lung cell types in the pathogenesis of early tuberculosis.  相似文献

Tightly constrained determinative methods can be used to characterise the silica minerals (opal-A, opal-CT, opal-C, quartz, moganite) and physical properties of silica sinters. Optimal X-ray powder diffraction operating parameters indicate silica lattice order/disorder using untreated, dry, <106 μm powders scanned at 0.6° 2θ/min with a step size of 0.01° from 10–40° 2θ and an internal Si standard. Simultaneous differential thermal and thermogravimetric analysis of 15.0±0.1 mg sinter samples of <106 μm grain size, at a heating rate of 20°C/min in dry air, identify thermal events associated with dehydration, organic combustion, and changes of state. Where abundant organic matter is present, nitrogen is the preferred atmosphere for thermal analysis. Thermogravimetric-determined water contents of sinters differ from Penfield determinations reflecting the differing nature of the two techniques. Laser Raman microprobe techniques can be used to explore the mineralogy of particular sinter morphologies and habits down to 10 μm diameter. The nature of the silica species present can assist in characterising individual sinter deposits and, combined with textural, density and/or porosity determinations, can lead to a better understanding of the hydrology and paleohydrology of a geothermal prospect.  相似文献
