The fatty acid composition and biosynthesis of fatty acids were studied during early embryogenesis of the toadBufo arenarum Hensel. The ova and stages up to the 6 1/2 day embryo have similar fatty acid compositions, with ca. 70% unsaturated acids. The eggs and embryo were permeable to acetate and impermeable to palmitic, linoleic, and eicosa-8,11,14-trienoic acid. Labeled acetate was incorporated by the eggs into the saturated acids-lauric, myristic, palmitic, stearic, arachidic, and behenic-and into the unsaturated acids-myristoleic, palmitoleic, oleic, and eicosaenoic acids. During segmentation and gastrulation, de novo biosynthesis of fatty acids increased, desaturation to myristoleic, palmitoleic, and oleic acids was enhanced; and fatty acids were esterified to triglycerides, phosphatidyl choline, and phosphatidyl ethanolamine. The feeding embryo (11 days) changed the pattern of incorporation to less incorporation into triglycerides.  相似文献

Kondo lattices are systems with unusual electronic properties that stem from strong electron correlation, typically studied in intermetallic 3D compounds containing lanthanides or actinides. Lowering the dimensionality of the system enhances the role of electron correlations providing a new tuning knob for the search of novel properties in strongly correlated quantum matter. The realization of a 2D Kondo lattice by stacking a single-layer Mott insulator on a metallic surface is reported. The temperature of the system is steadily lowered and by using high-resolution scanning tunneling spectroscopy, the phase transition leading to the Kondo lattice is followed. Above 27 K the interaction between the Mott insulator and the metal is negligible and both keep their original electronic properties intact. Below 27 K the Kondo screening of the localized electrons in the Mott insulator begins and below 11 K the formation of a coherent quantum electronic state extended to the entire sample, i.e., the Kondo lattice, takes place. By means of density functional theory, the electronic properties of the system and its evolution with temperature are explained. The findings contribute to the exploration of unconventional states in 2D correlated materials.  相似文献

A tubular micromotor with spatially resolved compartments is presented toward efficient site-specific cargo delivery, with a back-end zinc (Zn) propellant engine segment and an upfront cargo-loaded gelatin segment further protected by a pH-responsive cap. The multicompartment micromotors display strong gastric-powered propulsion with tunable lifetime depending on the Zn segment length. Such propulsion significantly enhances the motor distribution and retention in the gastric tissues, by pushing and impinging the front-end cargo segment onto the stomach wall. Once the micromotor penetrates the gastric mucosa (pH ≥ 6.0), its pH-responsive cap dissolves, promoting the autonomous localized cargo release. The fabrication process, physicochemical properties, and propulsion behavior are systematically tested and discussed. Using a mouse model, the multicompartment motors, loaded with a model cargo, demonstrate a homogeneous cargo distribution along with approximately four-fold enhanced retention in the gastric lining compared to monocompartment motors, while showing no apparent toxicity. Therapeutic payloads can also be loaded into the pH-responsive cap, in addition to the gelatin-based compartment, leading to concurrent delivery and sequential release of dual cargos toward combinatorial therapy. Overall, this multicompartment micromotor system provides unique features and advantages that will further advance the development of synthetic micromotors for active transport and localized delivery of biomedical cargos.  相似文献

The potential benefits of using human resources efficiently in the service sector constitute an incentive for decision makers in this industry to intelligently manage the work shifts of their employees, especially those dealing directly with customers. In the long term, they should attempt to find the right balance between employing as few labor resources as possible and keeping a high level of service. In the short run (e.g., 1 week), however, contracted staff levels cannot be adjusted, and management efforts thus focus on the efficient assignment of shifts and activities to each employee. This article proposes a mixed integer program model that solves the short-term multi-skilled workforce tour scheduling problem, enabling decision makers to simultaneously design workers’ shifts and days off, assign activities to shifts and assign those to employees so as to maximize and balance coverage of a firm’s demand for on-duty staff across multiple activities. Our model is simple enough to be solved with a commercial MIP solver calibrated by default without recurring to complex methodologies, such as extended reformulations and exact and/or heuristic column generation subroutines. A wide computational testing over 1000 randomly generated instances suggests that the model’s solution times are compatible with daily use and that multi-skilling is a significant source of labor flexibility to improve coverage of labor requirements, in particular when such requirements are negatively correlated and part-time workers are a scarce resource.  相似文献

The Hedgehog (Hh) pathway is essential for the embryonic development and homeostatic maintenance of many adult tissues and organs. It has also been associated with some functions of the innate and adaptive immune system. However, its involvement in the immune response has not been well determined. Here we study the role of Hh signalling in the modulation of the immune response by using the Ptch-1-LacZ+/− mouse model (hereinafter referred to as ptch+/−), in which the hemizygous inactivation of Patched-1, the Hh receptor gene, causes the constitutive activation of Hh response genes. The in vitro TCR stimulation of spleen and lymph node (LN) T cells showed increased levels of Th2 cytokines (IL-4 and IL-10) in ptch+/−cells compared to control cells from wild-type (wt) littermates, suggesting that the Th2 phenotype is favoured by Hh pathway activation. In addition, CD4+ cells secreted less IL-17, and the establishment of the Th1 phenotype was impaired in ptch+/− mice. Consistently, in response to an inflammatory challenge by the induction of experimental autoimmune encephalomyelitis (EAE), ptch+/− mice showed milder clinical scores and more minor spinal cord damage than wt mice. These results demonstrate a role for the Hh/ptch pathway in immune response modulation and highlight the usefulness of the ptch+/− mouse model for the study of T-cell-mediated diseases and for the search for new therapeutic strategies in inflammatory diseases.  相似文献

Hyaluronic acid (HA) is a Glycosaminoglycan made of disaccharide units containing N-acetyl-D-glucosamine and glucuronic acid. Its molecular mass can reach 10 MDa and its physiological properties depend on its polymeric property, polyelectrolyte feature and viscous nature. HA is a ubiquitous compound found in almost all biological tissues and fluids. So far, HA grades are produced by biotechnology processes, while in the human organism it is a major component of the extracellular matrix (ECM) in brain tissue, synovial fluid, vitreous humor, cartilage and skin. Indeed, HA is capable of forming hydrogels, polymer crosslinked networks that are very hygroscopic. Based on these considerations, we propose an overview of HA-based scaffolds developed for brain cancer treatment, central and peripheral nervous systems, discuss their relevance and identify the most successful developed systems.  相似文献

Liver nuclear incorporation of stearic (18∶0), linoleic (18∶2n−6), and arachidonic (20∶4n−6) acids was studied by incubation in vitro of the [1-14C] fatty acids with nuclei, with or without the cytosol fraction at different times. The [1-14C] fatty acids were incorporated into the nuclei as free fatty acids in the following order: 18∶0>20∶4n−6≫18∶2n−6, and esterified into nuclear lipids by an acyl-CoA pathway. All [1-14C] fatty acids were esterified mainly to phospholipids and triacylglycerols and in a minor proportion to diacylglycerols. Only [1-14C] 18∶2n−6-CoA was incorporated into cholesterol esters. The incorporation was not modified by cytosol addition. The incorporation of 20∶4n−6 into nuclear phosphatidylcholine (PC) pools was also studied by incubation of liver nuclei in vitro with [1-14C]20∶4n−6-CoA, and nuclear labeled PC molecular species were determined. From the 15 PC nuclear molecular species determined, five were labeled with [1-14C]20∶4n−6-CoA: 18∶0–20∶4, 16∶0–20∶4, 18∶1–20∶4, 18∶2–20∶4, and 20∶4–20∶4. The highest specific radioactivity was found in 20∶4–20∶4 PC, which is a minor species. In conclusion, liver cell nuclei possess the necessary enzymes to incorporate exogenous saturated and unsaturated fatty acids into lipids by an acyl-CoA pathway, showing specificity for each fatty acid. Liver cell nuclei also utilize exogenous 20∶4n−6-CoA to synthesize the major molecular species of PC with 20∶4n−6 at the sn-2 position. However, the most actively synthesized is 20∶4–20∶4 PC, which is a quantitatively minor component. The labeling pattern of 20∶4–20∶4 PC would indicate that this molecular species is synthesized mainly by the de novo pathway.  相似文献

Oil-derived middle distillates (straight-run gas oil and mixture with light cycle oil and coker gas oil) for Ultra-Low Sulfur Diesel (ULSD) production by HyDroTreating (HDT) were pretreated by selective Nitrogen Organic Compounds (NOC) adsorption. Highly crystalline Metal-Organic Framework (MOF) MIL-101(Cr) prepared with propylene oxide (proton scavenger) as textural improver was used to that end. MOF was characterized by N2 physisorption,X-ray diffraction,thermal analysis,infrared,Raman and UV-vis spectroscopies,and electron microscopy (SEM and HR-TEM). NOC removal was carried out at room temperature and atmospheric pressure,the adsorbent being easily regenerable under mild conditions. Extruded MOF efficiently removed NOC from real feedstocks to concentrations ~ 80 ppm which allowed ULSD production at much milder conditions to those used during pristine feedstocks HDT. Operating temperature could be significantly diminished (from 350 to 330 °C,at 56 kg cm−2(5.77 MPa),LHSV = 1.5 h−1,H2/oil = 2500 ft3bbl−1(445 m3m−3)) which could notably prolong cycle life of NiMo/Al2O3 formulation used.  相似文献

A comprehensive kinetic model for the photooxidation of oxalic acid (C2O4H4) in a fluidized bed (FB) of a TiO2/quartz sand composite, in a UV-fully illuminated photoreactor, was developed and correlated with experimental results. A proper solution for the mass balance of the reacting system was achieved by combining the complete radiation field profile and the most recent and updated pathway for the adsorption and photodecomposition of oxalic acid onto TiO2 (Mendive et al., 2007), which was employed to determine the kinetic expression and model parameters.  相似文献

The transmission eigenvalue problem arises in scattering theory. The main difficulty in its analysis is the fact that, depending on the chosen formulation, it leads either to a quadratic eigenvalue problem or to a non-classical mixed problem. In this paper we prove the convergence of a mixed finite element approximation. This approach, which is close to the Ciarlet–Raviart discretization of biharmonic problems, is based on Lagrange finite elements and is one of the less expensive methods in terms of the amount of degrees of freedom. The convergence analysis is based on classical abstract spectral approximation result and the theory of mixed finite element methods for solving the stream function–vorticity formulation of the Stokes problem. Numerical experiments are reported in order to assess the efficiency of the method.  相似文献
