Charybdotoxin (ChTX), a venom protein, suppresses Ca2+-activated K+ (K+(Ca)) currents in the glomus cell of neonatal rat carotid body. If it works similarly for cat carotid body chemoreceptors, charybdotoxin is expected to stimulate the chemosensory discharge during normoxia, and particularly hypoxia and hypercapnia. We studied the effects of charybdotoxin (20-40 nM) in vitro (perfused/superfused) on the cat carotid chemosensory discharge, and simultaneously tissue PO2 (PtiO2), as a measure of positive control. ChTX (20 nM) only increased PtiO2 and decreased carotid chemosensory discharge during hypoxia, indicating vasodilation. We conclude that K+(Ca) channels do not appear to play a significant role in chemotransduction in the cat carotid body.  相似文献

PURPOSE: To review the current literature and generate recommendations on the role of newer technology in the management of the unanticipated difficult airway. METHODS: A literature search using key words and filters of English language and English abstracted publications from 1990-96 contained in the Medline, Current Contents and Biological Abstracts databases was carried out. The literature was reviewed and condensed and a series of evidence-based recommendations were evolved. CONCLUSIONS: The unanticipated difficult airway occurs with a low but consistent incidence in anaesthesia practice. Difficult direct laryngoscopy occurs in 1.5-8.5% of general anaesthetics and difficult intubation occurs with a similar incidence. Failed intubation occurs in 0.13-0.3% general anaesthetics. Current techniques for predicting difficulty with laryngoscopy and intubation are sensitive, non-specific and have a low positive predictive value. Assessment techniques which utilize multiple characteristics to derive a risk factor tend to be more accurate predictors. Devices such as the laryngeal mask, lighted stylet and rigid fibreoptic laryngoscopes, in the setting of unanticipated difficult airway, are effective in establishing a patient airway, may reduce morbidity and are occasionally lifesaving. Evidence supports their use in this setting as either alternatives to facemask and bag ventilation, when it is inadequate to support oxygenation, or to the direct laryngoscope, when tracheal intubation has failed. Specifically, the laryngeal mask and Combitube have proved to be effective in establishing and maintaining a patent airway in "cannot ventilate" situations. The lighted stylet and Bullard (rigid) fibreoptic scope are effective in many instances where the direct laryngoscope has failed to facilitate tracheal intubation. The data also support integration of these devices into strategies to manage difficult airway as the new standard of care. Training programmes should ensure graduate physicians are trained in the use of these alternatives. Continuing medical education courses should allow physicians in practice the opportunity to train with these alternative devices.  相似文献

A new design concept for diffusion barriers in high‐density memory capacitors is suggested, and both RuTiN (RTN) and RuTiO (RTO) films are proposed as sacrificial oxygen diffusion barriers. The newly developed RTN and RTO barriers show a much lower sheet resistance than various other barriers, including binary and ternary nitrides (reported by others), up to 800 °C, without a large increase in the resistance. For both the Pt/RTN/TiSix/n++poly‐plug/n+ channel layer/Si and the Pt/RTO/RTN/TiSix/n++poly‐plug/n+ channel layer/Si contact structures, contact resistance—the most important electrical parameter for the diffusion barrier in the bottom electrode structure of capacitors—was found to be as low as 5 kohm, even after annealing up to 750 °C. When the RTN film was inserted as a glue layer between the bottom Pt electrode layer and the TiN barrier film in the chemical vapor deposited (Ba,Sr)TiO3 (CVD–BST) simple stack‐type structure, the RTN glue layer was observed to be thermally stable to temperatures 150 °C higher than that to which the TiN glue layer is stable. Moreover, the capacitance of the physical vapor deposited (PVD)–BST simple stack‐type structure adopted TiN glue layer initially degraded after annealing at 500 °C, and, thereafter, completely failed. In the case of the RTN and RTO/RTN glue layers, however, the capacitance continuously increased up to 550 °C. Thus, the new RTN and RTO films, which act as diffusion barriers to oxygen, are very promising materials for achieving high‐density capacitors.  相似文献

This study proposes a subsystem consisting of an analog buffer and a single‐ended input to a fully differential ΔΣ modulator to obtain low‐power consumption for portable electrocardiogram applications. With the proposed subsystem, the need for an inverting amplifier is avoided, and low‐power consumption is achieved. The ΔΣ modulator with a second order, 1 bit, and cascade of integrators feedforward structure consumes a low power, in which an inverting and a non‐inverting path implement a single‐ended input to fully‐differential signals. A double sampling technique is proposed for a digital‐to‐analog converter feedback circuit to reduce the effect of the reference voltage, reduce the amplifier requirements, and obtain low‐power consumption. Input‐bias and output‐bias transistors working in the weak‐inversion region are implemented to obtain an extremely large swing for the analog buffer. At a supply voltage of 1.2 V, signal bandwidth of 250 Hz, and sampling frequency of 128 kHz, the measurement results show that the modulator with a buffer achieves a 77 dB peak signal‐to‐noise‐distortion ratio, an effective‐number‐of‐bits of 12.5 bits, an 83 dB dynamic range, and a figure‐of‐merit of 156 dB. Sphingolipids are crucial for the life of the cell. In land‐dwelling mammals, they are equally important outside the cell—in the extracellular space of the skin barrier—because they prevent loss of water. Although a large body of research has elucidated many of the functions of sphingolipids, their extensive structural diversity remains intriguing. A new class of sphingolipids based on 6‐hydroxylated sphingosine has recently been identified in human skin. Abnormal levels of these 6‐hydroxylated ceramides have repeatedly been observed in atopic dermatitis; however, neither the biosynthesis nor the roles of these unique ceramide subclasses have been established in the human body. In this Minireview, we summarize the current knowledge of 6‐hydroxyceramides, including their discovery, structure, stereochemistry, occurrence in healthy and diseased human epidermis, and synthetic approaches to 6‐hydroxysphingosine and related ceramides.  相似文献

Tumor cell aggregation is critical for cell survival following the loss of extracellular matrix attachment and dissemination. However, the underlying mechanotransduction of clustering solitary tumor cells is poorly understood, especially in non-small cell lung cancers (NSCLC). Here, we examined whether cell surface protrusions played an important role in facilitating the physical contact between floating cells detached from a substrate. We employed poly-2-hydroxyethyl methacrylate-based 3D culture methods to mimic in vivo tumor cell cluster formation. The suprastructural analysis of human NSCLC A549 cell spheroids showed that finger-like protrusions clung together via the actin cytoskeleton. Time-lapse holotomography demonstrated that the finger-like protrusions of free-floating cells in 3D culture displayed exploratory coalescence. Global gene expression analysis demonstrated that the genes in the organic hydroxyl transport were particularly enriched in the A549 cell spheroids. Particularly, the knockdown of the water channel aquaporin 3 gene (AQP3) impaired multicellular aggregate formation in 3D culture through the rearrangement of the actomyosin cytoskeleton. Moreover, the cells with reduced levels of AQP3 decreased their transmigration. Overall, these data indicate that cell detachment-upregulated AQP3 contributes to cell surface protrusions through actomyosin cytoskeleton remodeling, causing the aggressive aggregation of free-floating cells dependent on the property of the substratum and collective metastasis.  相似文献

Numerical simulations were conducted using computational fluid dynamics to evaluate the effect of ventilation conditions on radon (222Rn) reduction performance in a residential building. The results indicate that at the same ventilation rate, a mechanical ventilation system is more effective in reducing indoor radon than a natural ventilation system. For the same ventilation type, the indoor radon concentration decreases as the ventilation rate increases. When the air change per hour (ACH) was 1, the indoor radon concentration was maintained at less than 100 Bq/m3. However, when the ACH was lowered to 0.01, the average indoor radon concentration in several rooms exceeded 148 Bq/m3. The angle of the inflow air was found to affect the indoor air stream and consequently the distribution of the radon concentration. Even when the ACH was 1, the radon concentrations of some areas were higher than 100 Bq/m3 for inflow air angles of 5° and 175°.  相似文献

The surface energies of pristine multi-walled carbon nanotubes (MWCNTs) and MWCNTs functionalized with carboxylic acid (MWCNT-COOH), acyl chloride and ethyl amine were characterized, and the effects of the changes in MWCNT surface energies on the interfacial adhesion and reinforcement of the composites were explored. When the surface energy of pristine MWCNTs was compared to that of functionalized MWCNTs, a decrease in the dispersive surface energy and an increase in the polar surface energy were observed. Interfacial adhesion energies between MWCNTs and various polymers were estimated from surface energy values of MWCNTs and various polymers. Among the MWCNTs, polyethylene, polystyrene and bisphenol-A polycarbonate (PC) had the highest interfacial energy with pristine MWCNTs, while nylon 6,6 and polyacrylamine exhibited the highest interfacial energy with MWCNT-COOH. When tensile properties and adhesion at the interface of PC and nylon 6,6 composites containing MWCNTs were examined, composites having high interfacial adhesion energy exhibited greater adhesion at the interface and reinforcement.  相似文献

In this paper, a general study on improving adaptability of quadruped walking and climbing robot in complex environment is presented. First, a sensing system composed of range and gyroscope sensors in a novel arrangement is developed. By combining the sensing signals and the internal state of the robot, the surface geometry of the environment is sufficiently reconstructed in real-time. Secondly, a planning algorithm for the robot to overcome the reconstructed environment is conducted. Based on the reshaped surface, the planning algorithm not only provides the exact body trajectory and foot positions but also the adaptability of the robot in a specific environment. A method to improve the adaptability of the walking and climbing robot is also introduced. Thanks to the adherent ability of the robot, the center of gravity of the robot is allowed to move outside the support polygon to increase the reach-ability of the next swing leg. Finally, the effectiveness of the proposed approach is verified by the performances of the experiments in complex environments using a quadruped walking and climbing robot named MRWALLSPECT IV.  相似文献
