In this study the levels of aflatoxins (AF B1, B2, G1 and G2) and ochratoxin A (OTA) were monitored in several food products imported in Italy with a high contamination risk. A total of 345 samples were collected from the Maritime Authority of Salerno Customs Port during the period from January 2008 to December 2009 and analyzed by immunoaffinity chromatography as clean-up, high performance liquid chromatography with fluorescence detection for quantification and tandem mass spectrometry for confirmation. The analytical methods were validated on different food matrices and meet the performance criteria set by EC Regulation No. 401/2006 for mycotoxin analysis. The results obtained in this survey showed that 7% of the total samples contained detectable levels of AFs and OTA, and 1.2% had AFs concentrations exceeding the maximum limits set by EU regulation. OTA was the most prevalent mycotoxin, with an incidence of 17.6% of samples analyzed for OTA. The highest detected levels were 23.70 μg kg?1 of OTA in a green coffee sample and 70.69 μg kg?1 of AFs in an apricot kernels sample. Among the food products analyzed, hazelnuts paste and dried vine fruits were the commodities mainly contaminated with AFs and OTA, respectively. 相似文献

Purpose
To assess the compliance of Daily Disposable Contact Lenses (DDCLs) wearers with replacing lenses at a manufacturer-recommended replacement frequency. To evaluate the ability of two different Health Behavioural Theories (HBT), The Health Belief Model (HBM) and The Theory of Planned Behaviour (TPB), in predicting compliance.Method
A multi-centre survey was conducted using a questionnaire completed anonymously by contact lens wearers during the purchase of DDCLs.Results
Three hundred and fifty-four questionnaires were returned. The survey comprised 58.5% females and 41.5% males (mean age 34 ± 12 years). Twenty-three percent of respondents were non-compliant with manufacturer-recommended replacement frequency (re-using DDCLs at least once). The main reason for re-using DDCLs was “to save money” (35%). Predictions of compliance behaviour (past behaviour or future intentions) on the basis of the two HBT was investigated through logistic regression analysis: both TPB factors (subjective norms and perceived behavioural control) were significant (p < 0.01); HBM was less predictive with only the severity (past behaviour and future intentions) and perceived benefit (only for past behaviour) as significant factors (p < 0.05).Conclusions
Non-compliance with DDCLs replacement is widespread, affecting 1 out of 4 Italian wearers. Results from the TPB model show that the involvement of persons socially close to the wearers (subjective norms) and the improvement of the procedure of behavioural control of daily replacement (behavioural control) are of paramount importance in improving compliance. With reference to the HBM, it is important to warn DDCLs wearers of the severity of a contact-lens-related eye infection, and to underline the possibility of its prevention. 相似文献123.

Micro injection moulding process represents a key technology for realizing micro components and micro devices used in several fields: IT components, biomedical and medical products, automotive industry, telecommunication area and aerospace. The development of new micro parts is highly dependent on manufacturing systems that can reliably and economically produce micro components in large quantities. In this work, the authors investigate the process parameters on the overall quality of a miniaturised dog-bone-shaped specimen in order to determine the process constraints. The factors affecting parts aspects and mass are studied by experimentation designed using DoE methodology and then discussed. Two polymer materials (polyoxymethylene and liquid crystal polymer), particularly suitable for injection moulding applications due to their flowability and stability, are tested and evaluated in relation to the process replication capability. It has been found that the holding pressure and holding time for POM and holding pressure and injection velocity for LCP have the highest influence on achieving high part mass. Differently, melt temperature has the highest influence on minimising the process variability for both tested polymers. A further investigation has been carried out on the relationship between the holding pressure and the part mass and dimensions demonstrating the existence of a linear correlation between specimens mass and dimensions. 相似文献

Schistosoma mansoni HDAC8 is a reliable target to fight schistosomiasis, and several inhibitors have been reported in the literature up to now. Nevertheless, only a few displayed selectivity over the human deacetylases and some exhibited very low or no activity against parasite larvae and/or adult worms. We report here the in vitro enzyme and biological activity of a small library of HDAC inhibitors from our lab, in many cases exhibiting submicromolar/nanomolar potency against smHDAC8 and diverse degrees of selectivity over hHDAC1 and/or hHDAC6. Such compounds were tested against schistosomula, and a selection of them against the adult forms of S. mansoni, to detect their effect on viability. Some of them showed the highest viability reduction for the larval stage with IC50 values around 1 μM and/or displayed ∼40–50 % activity in adult worms at 10 μM, joined to moderate to no toxicity in human fibroblast MRC-5 cells. 相似文献

Complexity reduction and kaizen events to balance manual assembly lines: an application in the field
Notwithstanding the existence of a broad research base on assembly line balancing (ALB), companies do not use the mathematical approaches developed in the literature to configure assembly lines. This article aims to fill the gap between research and application by presenting and testing in a real industrial context a methodology based on complexity reduction and kaizen events. First, the methodology supports reducing the complexity that affects real-life assembly systems in terms of the variety of, e.g. finished products, materials and parts. Next, the methodology proposes the conduction of kaizen events by using lean manufacturing tools, such as process analysis, time observation, waste identification, workstation standard documents and yamazumi charts. The methodology is successfully applied to a case study that describes its use in the confectionery process for a major chocolatier company along with the results of the application. The main contribution of this paper consists in presenting a method to manage the line balancing activity within everyday industrial realities, helping practitioners to improve and maintain the performance over time. 相似文献

Materials Requirement Planning (MRP) technique is widely employed by most manufacturing companies, even though field applications point out some weaknesses, including ignored production capacity constraints and fixed lead-times. These weaknesses often lead to infeasible production schedules, which trigger fluctuating workloads over time, significant adjustment effort and eventually unpredictably long lead times. This paper introduces a capacity-oriented MRP procedure that combines the traditional MRP procedure with an approach based on linear programming: in this way, requirement of lead times pre-determined a priori outside the MRP procedure is overcome. The new procedure is then applied to a real-life company and results highlight that feasible plans of orders are generated without requiring lead-times as input and without relevant computational burden. 相似文献

Dietary consumption of fruits and vegetables is related to a risk reduction in a series of leading human diseases, probably due to the plants’ antioxidant content. Plant-derived nanovesicles (PDNVs) have been recently receiving great attention regarding their natural ability to deliver several active biomolecules and antioxidants. To investigate the presence of active antioxidants in fruits, we preliminarily analyzed the differences between nanovesicles from either organic or conventional agriculture-derived fruits, at equal volumes, showing a higher yield of nanovesicles with a smaller size from organic agriculture-derived fruits as compared to conventional ones. PDNVs from organic agriculture also showed a higher antioxidant level compared to nanovesicles from conventional agriculture. Using the PDNVs from fruit mixes, we found comparable levels of Total Antioxidant Capacity, Ascorbic Acid, Catalase, Glutathione and Superoxide Dismutase 1. Finally, we exposed the nanovesicle mixes to either chemical or physical lytic treatments, with no evidence of effects on the number, size and antioxidant capacity of the treated nanovesicles, thus showing a marked resistance of PDNVs to external stimuli and a high capability to preserve their content. Our study provides for the first time a series of data supporting the use of plant-derived nanovesicles in human beings’ daily supplementation, for both prevention and treatment of human diseases. 相似文献

Mtfr1/Chppr is a nuclear gene coding for a mitochondrial protein capable of inducing fission of this organelle in a sequence-specific manner. Here we show that in mice, Mtfr1/Chppr is ubiquitously expressed and displays the highest level of expression in pubertal and adult testes and in particular in spermatids and Leydig cells. To investigate Mtfr1 function in vivo, we analyzed homozygous mice null for this gene obtained through a gene trap approach. We show that these mice fail to express Mtfr1 and that in their testes several genes coding for enzymes involved in the defense against oxidative stress are downregulated. Among these, we studied in particular glutathione peroxidase 3 and show its expression in selected testis cell types. Furthermore, we demonstrate oxidative DNA damage specifically in testes of Mtfr1-deficient mice likely resulting from a reduced antioxidant activity. As a whole, these data suggest that Mtfr1 protects the male gonads against oxidative stress. 相似文献

Tracing the appearance and evolution of virus variants is essential in the management of the COVID-19 pandemic. Here, we focus on SARS-CoV-2 spread in Italian patients by using viral sequences deposited in public databases and a tracing procedure which is used to monitor the evolution of the pandemic and detect the spreading, within the infected population of emergent sub-clades with a potential positive selection. Analyses of a collection of monthly samples focused on Italy highlighted the appearance and evolution of all the main viral sub-trees emerging at the end of the first year of the pandemic. It also identified additional expanding subpopulations which spread during the second year (i.e., 2021). Three-dimensional (3D) modelling of the main amino acid changes in mutated viral proteins, including ORF1ab (nsp3, nsp4, 2’-o-ribose methyltransferase, nsp6, helicase, nsp12 [RdRp]), N, ORF3a, ORF8, and spike proteins, shows the potential of the analysed structural variations to result in epistatic modulation and positive/negative selection pressure. These analyzes will be of importance to the early identification of emerging clades, which can develop into new “variants of concern” (i.e., VOC). These analyses and settings will also help SARS-CoV-2 coronet genomic centers in other countries to trace emerging worldwide variants. 相似文献
