The industrially important yeast Candida utilis is widely used in production of food and medical materials, but its host-vector system has not been well developed. We screened for compact and efficient ARSs to construct practically useful vectors. The C. utilis strain AHU3053 was found to be efficiently transformed by the conventional lithium acetate method and was used as the host. The C. utilis IAM4264 genomic library was constructed by inserting the partial Sau3AI digests in pRI51, which has a kanMX gene expressible in C. utilis. By examining 98 C. utilis G418-resistant transformants, five plasmids had the highest ARS activity. By trimming of the inserts, the 1490 and 552 bp fragments with transformation activity of over 10(3)/microg DNA were obtained from ARS3 and ARS4, respectively. Although several sequences identical to S. cerevisiae ARS consensus sequences (ACSs) were found in ARS3 and ARS4, our deletion analysis indicated that these were not essential for the activity. Because the minimal functional ARS fragment was also several-fold larger than that of S. cerevisiae, the C. utilis ARSs have some unique characteristics resembling the Sz. pombe ARSs. Axisymmetric nozzle flows with a free‐jet expansion are simulated considering several substances and several flow conditions, and the thermophysical properties in the nozzle and the free‐jet region are predicted. The present numerical method is based on the preconditioning method developed by Yamamoto and the mathematical models of thermophysical properties of the substances. As numerical examples show, gas flows of carbon dioxide, water, and nitrogen under a subcritical pressure condition are first calculated. Calculated distances to the Mach disk are compared with the experimental results, and also the density distributions are compared among these three substances. Second, carbon dioxide flows while changing the pressure from subcritical to supercritical values are calculated and the effect of pressure on the flow field is investigated. A new hot forming system for high-alloy metals with poor workability is developed. This forming system is a direct combination of a forming machine and continuous electric resistance heating, and is capable of the high-speed forming of metals with flexible and precise control of elevated temperature. The prototype of the new forming system is applied to the forming of high-alloy metals with poor formability, such as a Ti–6Al–4V bar, and the basic characteristics of this forming system are clarified. Using this system, sound, high speed forming of high alloys becomes possible, and the Ti–6Al–4V bar is successfully formed without fracture. 相似文献

This paper is an overview of the high-speed DRAM architecture developments. We discuss developments on density growth, interface technology, memory-core architecture, and DRAM+ASIC technology. We can find the developments of density as 2× growth instead of 4× by each generation. Interface technologies will have a tendency to use the terminated bus structure for higher data rate. Memory-core architecture developments are the trials for actual bandwidth improvements. DRAM+ASIC technologies seem to require universal interface solutions. Most anti-cancer drug screening assays are currently performed in two dimensions, on flat, rigid surfaces. However, there are increasing indications that three-dimensional (3D) platforms provide a more realistic setting to investigate accurate morphology, growth, and sensitivity of tumor cells to chemical factors. Moreover, interstitial flow plays a pivotal role in tumor growth. Here, we present a microfluidic 3D platform to investigate behaviors of tumor cells in flow conditions with anti-migratory compounds. Our results show that interstitial flow and its direction have significant impact on migration and growth of hepatocellular carcinoma cell lines such as HepG2 and HLE. In particular, HepG2/HLE cells tend to migrate against interstitial flow, and their growth increases in interstitial flow conditions regardless of the flow direction. Furthermore, this migratory activity of HepG2 cells is enhanced when they are co-cultured with human umbilical vein endothelial cells. We also found that migration activity of HepG2 cells attenuates under hypoxic conditions. In addition, the effect of Artemisinin, an anti-migratory compound, on HepG2 cells was quantitatively analyzed. The microfluidic 3D platform described here is useful to investigate more accurately the effect of anti-migratory drugs on tumor cells and the critical influence of interstitial flow than 2D culture models. 相似文献

We propose a methodology to optimize radiation protection in radioactive waste disposal after the closure of a disposal facility based on a probabilistic approach. In this methodology, a set of alternative options of the disposal system design with the associated uncertainties estimated through a probabilistic approach are developed. Then, the methodology evaluates the advantages and disadvantages of each option for the optimization of radiation protection, in which it is possible to determine the compliance with a dose constraint of 0.3 mSv/y and to collect information for making a decision on the optimization. In particular, the obtained information on the exposure, which is the mode and width of the dose distribution, can be converted into information on engineering measures and site conditions for the options. This process allows us to discuss which option should be selected as the optimal option by considering the balance between the exposure and the engineering, economic, and social feasibility of the option. This methodology is helpful for providing clear reasons why an optimal disposal system design is selected by combining quantitative information on the exposure and the feasibility of each option. 相似文献

This paper focuses on a maximum‐power‐point tracking method of photovoltaics via use of the short‐current pulse. It has been reported that the optimum operating current is proportional to the short current and that maximum‐power‐point tracking can be performed by detecting the short current. The proposed method utilizes the intermittent short‐current pulse to estimate the optimum operating current and its operating characteristics have experimentally been verified. Also, an adaptive mechanism to identify the parameter between the optimum current and the short current is discussed.
