High hydrostatic pressure (HHP) processing was used to determine its ability to induce protein gelation. Isolated hake myofibrils were processed by HHP at 0, 150, 250, and 500 MPa (10?ºC/10 min) and/or by heating (90?ºC/20 min). The results were analyzed by Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), determination of sulfhydryl group contents, and dynamic rheometry measurements. FTIR data indicated that secondary protein structures exhibited a reduction in α-helix together with an increase in β-sheet as a result of protein denaturation caused by HHP. DSC showed that HHP induced a reduction in myosin denaturation temperature (T peak) indicating protein unfolding. Protein gelation after HHP is based on physical (non-covalent) interactions which make more sulfhydryl groups available, while after heating, it is based on the formation of covalent (disulfide) bonds as a consequence of protein denaturation reducing the sulfhydryl groups. The combination of HHP and heating, particularly the latter, improved network stabilization. These results were reflected in the rheological changes, in which heated gels showed more elastic, cohesive, and time-stable networks than pressurized (non-heated) gels. The HHP effect provided softer, more flexible networks. The gel at 500 MPa was the most elastic and time-stable and exhibited the highest level of connectivity. 相似文献

Background
It has been demonstrated that statins can increase intestinal sterol absorption. Augments in phytosterolemia seems related to cardiovascular disease.Objective
We examined the role of soluble fiber intake in endogenous cholesterol synthesis and in sterol absorption among subjects under highly effective lipid-lowering therapy.Design
In an open label, randomized, parallel-design study with blinded endpoints, subjects with primary hypercholesterolemia (n = 116) were assigned to receive during 12 weeks, a daily dose of 25 g of fiber (corresponding to 6 g of soluble fibers) plus rosuvastatin 40 mg (n = 28), rosuvastatin 40 mg alone (n = 30), sinvastatin 40 mg plus ezetimibe 10 mg plus 25 g of fiber (n = 28), or sinvastatin 40 mg plus ezetimibe 10 mg (n = 30) alone.Results
The four assigned therapies produced similar changes in total cholesterol, LDL-cholesterol, and triglycerides (p < 0.001 vs. baseline) and did not change HDL-cholesterol. Fiber intake decreased plasma campesterol (p < 0.001 vs. baseline), particularly among those patients receiving ezetimibe (p < 0.05 vs. other groups), and β-sitosterol (p = 0.03 vs. baseline), with a trend for lower levels in the group receiving fiber plus ezetimibe (p = 0.07). Treatment with rosuvastatin alone or combined with soluble fiber was associated with decreased levels of desmosterol (p = 0.003 vs. other groups). Compared to non-fiber supplemented individuals, those treated with fibers had weight loss (p = 0.04), reduced body mass index (p = 0.002) and blood glucose (p = 0.047).Conclusion
Among subjects treated with highly effective lipid-lowering therapy, the intake of 25 g of fibers added favorable effects, mainly by reducing phytosterolemia. Additional benefits include improvement in blood glucose and anthropometric parameters.994.

The maximal upstroke of transmembrane voltage (dV(m)/dt(max)) has been used as an indirect measure of sodium current I(Na) upon activation in cardiac myocytes. However, sodium influx generates not only the upstroke of V(m), but also the downstroke of the extracellular potentials V(e) including epicardial surface potentials V(es). The purpose of this study was to evaluate the magnitude of the maximal downstroke of V(es) (|dV(es)/dt (min)|) as a global index of electrical activation, based on the relationship of dV(m)/dt(max) to I(Na). To fulfill this purpose, we examined |dV(es)/dt(min)| experimentally using isolated perfused mouse hearts and computationally using a 3-D cardiac tissue bidomain model. In experimental studies, a custom-made cylindrical "cage" array with 64 electrodes was slipped over mouse hearts to measure V(es) during hyperkalemia, ischemia, and hypoxia, which are conditions that decrease I(Na). Values of |dV(es)/dt(min)| from each electrode were normalized (|dV(es)/dt (min)|(n)) and averaged (|dV(es)/dt(min)|(na)). Results showed that |dV(es)/dt(min)|(na) decreased during hyperkalemia by 28, 59, and 79% at 8, 10, and 12?mM [K(+)](o), respectively. |dV(es)/dt(min)| also decreased by 54 and 84% 20 min after the onset of ischemia and hypoxia, respectively. In computational studies, |dV(es)/dt(min)| was compared to dV(m)/dt(max) at different levels of the maximum sodium conductance G(Na), extracellular potassium ion concentration [K(+)](o), and intracellular sodium ion concentration [Na(+)](i), which all influence levels of I(Na). Changes in |dV(es)/dt(min)|(n) were similar to dV(m)/dt (max) during alterations of G(Na), [K(+)](o), and [Na(+)](i). Our results demonstrate that |dV(es)/dt(min)|(na) is a robust global index of electrical activation for use in mouse hearts and, similar to dV(m)/dt(max), can be used to probe electrophysiological alterations reliably. The index can be readily measured and evaluated, which makes it attractive for characterization of, for instance, genetically modified mouse hearts and drug effects on cardiac tissue. 相似文献

BACKGROUND: Consumption of broccoli is increasing steadily worldwide because of the interest in its bioactive composition and nutritive value for health promotion. Novel broccoli cultivars to be established under current adverse conditions in production areas (aggressive environmental conditions and saline irrigation waters) need to maintain physical and nutritional quality for consumption and year‐round supply to the markets. The newly introduced cultivars ‘Naxos’ and ‘Parthenon’ have been selected as potential candidates to replace the currently underperforming ‘Nubia’ variety. We aimed to compare the physical and phytochemical quality (glucosinolates, hydroxycinnamic acids, flavonoids, vitamin C and minerals), as well as the in vitro antioxidant capacity of these three cultivars under conditions of environmental stress. RESULTS: ‘Parthenon’ showed equal productivity and nutritional composition to ‘Nubia’, whereas ‘Naxos’ presented in general the best results when compared to ‘Nubia’ and ‘Parthenon’. For phenolic compounds ‘Nubia’ presented the highest contents, although ‘Naxos’ seemed better adapted to saline stress conditions, as suggested by the lowest degree of variation in the contents of healthy phytochemicals, including phenolic compounds, when grown under such conditions. CONCLUSION: ‘Naxos’ broccoli performed best and is a suitable candidate to replace ‘Nubia’ for marketable, nutritive and phytochemical quality, especially in areas of production under adverse conditions as found in Mediterranean southeast Spain (semiarid climate with saline irrigation water). Object
This study demonstrates that 3T SV-MRS data can be used with the currently available automatic brain tumour diagnostic classifiers which were trained on databases of 1.5T spectra. This will allow the existing large databases of 1.5T MRS data to be used for diagnostic classification of 3T spectra, and perhaps also the combination of 1.5T and 3T databases.Materials and methods
Brain tumour classifiers trained with 154 1.5T spectra to discriminate among high grade malignant tumours and common grade II glial tumours were evaluated with a subsequently-acquired set of 155 1.5T and 37 3T spectra. A similarity study between spectra and main brain tumour metabolite ratios for both field strengths (1.5T and 3T) was also performed.Results
Our results showed that classifiers trained with 1.5T samples had similar accuracy for both test datasets (0.87 ± 0.03 for 1.5T and 0.88 ± 0.03 for 3.0T). Moreover, non-significant differences were observed with most metabolite ratios and spectral patterns.Conclusion
These results encourage the use of existing classifiers based on 1.5T datasets for diagnosis with 3T 1H SV-MRS. The article presents the results on the investigation of the channeling and volume reflection effects in a bent silicon crystal with 13 GeV/c positive and negative hadrons (mainly π+, p and π−) at the CERN PS T9 line. In particular, this is the first study carried out on volume reflection at this energy providing a deflection angle of 69.4 ± 4.7 μrad and an efficiency of 92.7 ± 3.3%, with positive particles.The measurements have been carried out on a bent silicon strip crystal, using a high precision tracking system based on microstrip silicon detectors; this setup is allowed to trigger on the desired beam portion and to select the incoming particle angular range. In this paper we present the biaxial failure curve of a chopped glass-reinforced polyester composite. The curve has been obtained combining the results of experimental tests with the results of numerical simulations developed by means of the Finite Element Method. The experimental tests have been performed applying perpendicular loads to cruciform specimens. Then, taking into account the numerical results, we have constructed the failure envelope in the tensile–tensile quadrant. Moreover we propose to modify the cruciform geometry to obtain, varying the widths of the loaded arms and the tapered central zone, points of the failure curve in which the two components of the plane stress tensor are different. 相似文献

Transport measurements on the cuprates suggest the presence of a quantum critical point (QCP) hiding underneath the superconducting dome near optimal hole doping. We provide numerical evidence in support of this scenario via a dynamical cluster quantum Monte Carlo study of the extended two-dimensional Hubbard model. Single-particle quantities, such as the spectral function, the quasi-particle weight and the entropy, display a crossover between two distinct ground states: a Fermi liquid at low filling and a non-Fermi liquid with a pseudo-gap at high filling. Both states are found to cross over to a marginal Fermi-liquid state at higher temperatures. For finite next-nearest-neighbour hopping t', we find a classical critical point at temperature T(c). This classical critical point is found to be associated with a phase-separation transition between a compressible Mott gas and an incompressible Mott liquid corresponding to the Fermi liquid and the pseudo-gap state, respectively. Since the critical temperature T(c) extrapolates to zero as t' vanishes, we conclude that a QCP connects the Fermi liquid to the pseudo-gap region, and that the marginal Fermi-liquid behaviour in its vicinity is the analogue of the supercritical region in the liquid-gas transition. 相似文献

Nanostructures are potentially useful as building blocks to complement future electronics because of their high versatility and packing densities. The fabrication and characterization of particular nanostructures and the use of new theoretical tools to describe their properties are receiving much attention. However, the integration of these individual systems into general schemes that could perform simple tasks is also necessary because modern electronics operation relies on the concerted action of many basic units. We review here new conceptual schemes that can allow information processing with ligand or monolayer protected metallic nanoclusters (MPCs) on the basis of the experimentally demonstrated and theoretically described electrical characteristics of these nanostructures. In particular, we make use of the tunnelling current through a metallic nanocluster attached to the electrodes by ligands. The nanostructure is described as a single electron transistor (SET) that can be gated by an external potential. This fact permits exploiting information processing schemes in approximately defined arrays of MPCs. These schemes include: (i) binary, multivalued, and reversible logic gates; (ii) an associative memory and a synchronization circuit; and (iii) two signal processing nanodevices based on parallel arrays of MPCs and nanoswitches. In each case, the practical operation of the nanodevice is based on the SET properties of MPCs reported experimentally. We examine also some of the practical problems that should be addressed in future experimental realizations: the stochastic nature of the electron tunnelling, the relatively low operation temperatures, and the limited reliability caused by the weak signals involved and the nanostructure variability. The perspectives to solve these problems are based on the potentially high degree of scalability of the nanostructures. 相似文献
