In lethal and sublethal ammonia toxicity tests, we examined differences in tolerance of three species of freshwater amphipods, one native and two invasive in Ireland. The native Gammarus duebeni celticus was slightly less tolerant to ammonia than the invasive G. pulex (96 h LC50= 1.155 and 1.544 mg l(-1), respectively), while another invader, Crangonyx pseudogracilis, had the lowest tolerance (LC50= 0.36 mg l(-1)). Parasitism of G. pulex by the acanthocephalan Echinorhynchus truttae greatly reduced the tolerance of the invader to ammonia (LC50= 0.381 mg l(-1)). Further, precopula pair disruption tests indicated that G. d. celticus was more sensitive to ammonia than G. pulex at sublethal levels. We discuss these results in the context of the ecological replacements of native by invader amphipods.  相似文献

Cytotoxic T lymphocytes (CTL) specific for human minor histocompatibility (H) antigens can be isolated from the blood of major histocompatibility complex (MHC)-matched allogeneic bone marrow transplant (BMT) recipients and may play a prominent role in the graft-versus-host (GVH) and graft-versus-leukemia (GVL) reactions (Tsoi et al, J Immunol 125:2258, 1980; Tsoi et al, Transplant Proc 15:1484, 1983; Goulmy et al, Nature 302:159, 1983; Irle et al, Transplantation 40:329, 1985; and Niederwieser et al, Blood 81:2200, 1993). The identification of minor H antigens that are expressed in hematopoietic cells, including leukemic cells, but not in fibroblasts and other tissue types has suggested that such tissue-restricted antigens could potentially serve as targets for T-cell immunotherapy to enhance GVL activity without inducing GVH disease (de Bueger et al, J Immunol 149:1788, 1992; van der Harst et al, Blood 83:1060, 1994; and Dolstra et al, J Immunol 158:560, 1997). To explore the feasibility of this strategy, donor CD3+CD8+ CTL clones specific for recipient minor H antigens were isolated and characterized from allogeneic BMT recipients. CTL clones were obtained from the majority of donor/recipient pairs. Seventeen distinct minor H antigens distinguishable by their MHC-restricting allele, population frequency, and/or distribution of tissue expression were defined by 56 CD3+CD8+ CTL clones isolated from these patients. The MHC-restricting alleles for these CTL clones included HLA-A2 and HLA-B7, which had previously been shown to present minor H antigens to CTL, as well as HLA-A3, -A11, -B8, -B53, and -Cw7, which had not previously been described to present minor H antigens to CTL. Estimated phenotype frequencies for these 17 distinct minor H antigens range from 0.17 to 0.92. In vitro cytotoxicity assays using hematopoietic cells and fibroblasts as target cells showed that 5 of the 17 minor H antigens were expressed in both hematopoietic cells and fibroblasts. However, 12 were presented for CTL recognition only by hematopoietic cells and not by dermal fibroblasts derived from the same donors. These results significantly extend the spectrum of CTL-defined human minor H antigens that could potentially serve as target antigens for cellular immunotherapy to promote GVL activity after allogeneic BMT.  相似文献

Recent advances in neutron and X-ray sources and instrumentation, new and improved scattering techniques, and molecular biology techniques, which have permitted facile preparation of samples, have each led to new opportunities in using small-angle scattering to study the conformations and interactions of biological macromolecules in solution as a function of their properties. For example, new instrumentation on synchrotron sources has facilitated time-resolved studies that yield insights into protein folding. More powerful neutron sources, combined with molecular biology tools that isotopically label samples, have facilitated studies of biomolecular interactions, including those involving active enzymes.  相似文献

Previous reports concerning the location of D18S44 with respect to the centromere have been ambiguous. Also, it has not been possible, based on formerly reported markers, to show that contigs WC18.0 and WC18.1 overlap. However, the data presented here definitively show, using FISH technology, that D18S44 (located on WC18.0) maps to proximal 18q. Furthermore, inter-Alu fingerprinting shows a clear overlap between WC18.0 and WC18.1, thereby establishing a complete contig between D18S44 and markers from WC18.1.  相似文献

BACKGROUND & AIMS: Patients with inflammatory bowel disease have symptoms of irritable bowel syndrome (IBS) with a higher than expected prevalence. Stress is an important factor in the pathogenesis of IBS. Thus, previous inflammation may predispose to IBS by rendering the bowel more susceptible to the impact of stress. The aim of this study was to examine the effect of previous colitis on stress-induced responses in rats. METHODS: Acute colitis was induced in rats by intrarectal administration of trinitrobenzene sulfonic acid (TNBS), and the rats were allowed to recover for 6 weeks before application of mild restraint stress for 3 consecutive days. In vitro measurements included myeloperoxidase activity, plasma corticosterone levels, interleukin 1 beta messenger RNA expression, and [3H]noradrenaline release from the myenteric plexus. RESULTS: Six weeks after administration of TNBS, stress caused a significant increase in myeloperoxidase activity in TNBS-treated rats but not in stressed controls; plasma corticosterone responses were similar. Stress also caused an exaggerated and significant suppression of [3H]noradrenaline release in TNBS-treated stressed rats compared with stressed controls. This was accompanied by a significant decrease in interleukin 1 beta messenger RNA expression in the colon. CONCLUSIONS: Previous colitis rendered the colon more susceptible to effects of stress on enteric nerve function and also increased some parameters of inflammation in response to stress.  相似文献

The fire-retardant performances of various surface coatings for naval ship interior applications have been evaluated by a number of small-scale fire test methods. The coatings were eveluated as topcoats by cone calorimetry and limiting oxygen index methods and as primer/topcoat systems by radiat heating, direct flame impingement and reverse panel heating procedures. The coatings investigated were (1) a solvent-based epoxy polyester, (2) a solvent-based fireretardant alkyd,(3) a solvent-based chlorimated alkyd and (4) a water-based acrylic. In terms of the measured parameters, the chlorinated alkyd has superior fire properties while the epoxy polyserter, the fire-retardant alkyd and parameters, the chlorinated alkyd has superior fire properties while the epoxy polyester, the fire-retardant alkyd and the acrylic have significat disadvantages in relation to fire propagation. The relatively low amount of hydrogen chloride (~ 0.3%—mass per initial mass) evolved by the chlorinated alkyd during cone calorimetric combustion and its outstanding fire-retardant properties have resulted in it being considered acceptable for naval ship interior applications.  相似文献

Individual differences in taste perception may influence dietary habits, nutritional status, and ultimately nutrition-related chronic disease risk. Individual differences in sweetness intensity perception and the relationship between perceived sweetness intensity, food behaviors, and dietary intake was investigated in 85 adults. Subjects (body mass index [BMI]= 21 ± 3, 21 ± 4 y) completed a food and diet questionnaire, food variety survey, 2 24-h food records, and a perceived sweetness intensity measurement using the general labeled magnitude scale (gLMS). There was interindividual variation in perceived sweetness intensity (0 to 34 gLMS units, mean 10 ± 7). One-way analysis of variance (ANOVA) revealed no difference between perceived sweetness intensity and degree of importance placed on not adding sugar to tea or coffee (P = 0.2) and the degree of importance placed on avoiding sugar-sweetened or fizzy drinks (P = 1.0). Independent t-test analysis revealed no significant association between perceived sweetness intensity and the food variety measure for sugar and confectionary intake (P = 0.6) and selected fruit and vegetable intake (P = 0.1 to 0.9). One-way ANOVA also demonstrated no difference between tertiles of sweetness intensity and BMI (P = 0.1), age (P = 0.3), and food variety score (P = 0.5). No correlation was observed with regards to perceived sweetness intensity and mean total energy (kJ) intake (r = 0.05, P = 0.7), percent energy from total fat, saturated fat, protein, carbohydrate, and grams of fiber (r =-0.1 to 0.1, P = 0.2 to 0.8) and also for intake of the micronutrients: folate, magnesium, calcium, iron, and zinc (r = 0.1 to 0.2, P = 0.1 to 0.4). Only modest correlations were observed between sodium (r = 0.3, P < 0.05), vitamin C (r = 0.3, P < 0.05), and potassium (r = 0.2, P < 0.0) intake and perceived sweetness intensity. Overall, perceived sweetness intensity does not appear to play a role in food behaviors relating to sugar consumption and dietary intake in adults.  相似文献

In two experiments with Holstein cows effects of dietary supplementation of niacin to periparturient cattle on subsequent milk production and ketosis were examined. In Experiment 1, 20 cows were assigned to two groups. The treatment group received niacin at 6 g per head per day for 2 wk prepartum and 12 g per head per day for 4 wk postpartum. The control group received no niacin. In Experiment 2, 40 cows were assigned to four groups of 10 cows. One group served as the control and the other three received 3, 6, or 12 g niacin per head per day for 10 wk postpartum. In both experiments niacin-supplemented groups produced slightly more milk than controls. There were no consistent changes of milk composition with treatment. Glucose concentration in blood serum was higher for cows receiving niacin. Concentrations of beta-hydroxybutyrate and nonesterified fatty acids in blood and plasma were generally lower for niacin-supplemented groups. Niacin supplementation had no effect on feed intake or body weight changes. In Experiment 1 there was a significant reduction of niacin concentration in red blood cells of postpartum cows not receiving supplemental niacin. Insulin concentration in blood serum in Experiment 2 was unaffected by niacin supplementation.  相似文献

ABSTRACT

Surface energy balance models such as the surface energy balance system (SEBS) assume that the influence of soil moisture and biophysical parameters on evapotranspiration (ET), are implicitly encompassed in model input variables such as the land surface temperature. This simplification may generally be suitable for environments in which the available energy constrains ET but may prove to be inadequate during suboptimal environmental conditions. To address this limitation, the SEBSESF approach was proposed to better represent the influence of environmental stress on surface energy fluxes, using an empirically derived calibration factor to adjust the evaporative fraction previously determined in SEBS. SEBSESF was evaluated by comparing the modelled ET estimates against Eddy Covariance ET measurements (ECET) at two sites, with contrasting environmental conditions. Comparisons between ECET and modelled ET indicated that the implementation of SEBSESF resulted in an improved correlation between ECET and modelled ET estimates, as well as an increase in the percentage of modelled ET estimates within an acceptable accuracy range (± 15 to 30%). Overall, the results of the study demonstrate how potential improvements in the mapping of ET within arid and semi-arid environments can be achieved by accounting for environmental stress. This in turn provides an opportunity to better understand and quantify energy and water fluxes, using satellite-based ET estimation approaches.  相似文献
