In general, the stiffened plates consisting of steel plate elements are unavoidably accompanied by initial imperfections such as residual stresses and initial deflections, which have considerable effects on their ultimate strength. Therefore, it is needed for designing them to develop more rational method taking the ultimate strength influenced by initial imperfections into account rather than the conventional design method being on the basis of the linear elastic buckling theory.From this point of view, this study aims to evaluate rigorously the ultimate strength of orthogonally stiffened plate with initial imperfections under uniaxial in-plane compression. The elasto-plastic finite element method is applied to attain this purpose. By a happy combination of modal analytical technique and conventional finite element method, much reduction of the degree of freedom can be expected to be realized herewith. The dynamic moduli, E′ and E″, and tan δ for nylon–CR and PET–CR composites with unidirectional short fibers were studied as a function of temperature by using a Rheovibron. The temperature dependence of tan δ showed two dispersion peaks for nylon–CR composite. The peak at ?28°C corresponded to the main dispersion of CR and the peak at 100°C to the α-dispersion of nylon 6. For a PET-CR composite, in addition to the individual dispersion of CR and PET, a small and broad peak was observed at about 90°C. The angular dependence of E′ indicated that the short fibers assumed good orientation. The storage modulus for the composites was given by the parallel model as E′ = Vf′Ef + VmE′m., where E′c, E′f and E′m were the storage modulus for the composite, fiber, and matrix and Vf and Vm were the volume fraction of fiber and matrix, respectively. In the transverse direction of fiber, the peak values of tan δ at ?28°C were given by the following equation; tan δc = tan δm ? δVf, where tan δc and tan δm are the loss tangent for the composite and matrix, respectively, and α is coefficient depending on fiber type. This is the first report of glycoside hydrolase family 43 β-xylosidase from Aspergillus oryzae. To characterize this enzyme, the recombinant enzyme was expressed in Escherichia coli. This paper presents the synergistic enhancement of the refolding yield of denatured and reduced lysozyme by using detergents as aggregation inhibitors and water-miscible organic cosolvents as modulators for the detergents. Adding only cetyltrimethylammonium bromide (CTAB) led to a slight increase in the refolding yield (up to 13%). Further addition of dimethylsulfoxide (DMSO) with CTAB drastically increased the refolding yield up to 35%, a value which was higher than the simple sum of the refolding yields in the presence of only CTAB or DMSO. The synergistic enhancement was also observed in the coexistence of other detergents, such as polyethylene glycol monooleyl ether (n = 50) and N-tetradecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate, and cosolvents, such as N,N-dimethylformamide and N,N-dimethylacetamide. Experimental data and a kinetic analysis revealed the guideline for selecting a couple of additives; detergents which can adequately inhibit the aggregation of proteins by binding to hydrophobic surfaces of refolding intermediates should be employed as an aggregation inhibitor, and cosolvents which can properly prevent both protein–protein and protein–detergent interactions act as effective modulators for the aggregation inhibitor, resulting in a desirable balance between folding and aggregation rates. 相似文献

We have previously identified and cloned a human gene, D40, that is preferentially expressed in testis among normal organs, while it is widely expressed in various human tumor cell lines and primary tumors derived from different organs. In this report, we have examined the expression and localization of this protein in human testis with an antibody specific to D40 protein. In Western analyses, the anti-D40 antibody recognized a major band with a molecular mass of 300 kDa and a minor band of 250 kDa. These bands were not observed in the testis lysates from patients with Sertoli-cell-only syndrome and with Kleinfelter syndrome, who lack germ cells of the testis, indicating that D40 protein is expressed in the germ cells of normal testis. Immunohistochemical studies have revealed that D40 protein is highly expressed in spermatocytes and in the pre-acrosome of round spermatids. In the acrosome, D40 protein expression is observed not inside but outside the acrosome membrane. This is consistent with the finding that the amino-acid sequence at the amino terminal of the D40 protein lacks a hydrophobic signal peptide that is required for proteins to translocate to the membrane. Expression of D40 protein is observed in the acrosome of ejaculated spermatozoa as well, although the level is low compared with that in the pre-acrosome of spermatids. These results suggest that D40 protein plays important roles in spermatogenesis, especially in the formation and maintenance of the acrosome. 相似文献

Thermal and nonthermal effects originating when a system is subjected to a microwave radiation field in the TiO2-photocatalyzed transformation of model substances containing various functional groups (e.g., benzoic acid, phthalic acid, o-formylbenzoic acid, phthalaldehyde, succinic acid, dimethyl phthalate, diethyl phthalate, and phenol) have been examined under simultaneous irradiation by ultraviolet (UV) and microwave (MW) radiations. Characteristics of the microwave effects and the fate of each substrate during the microwave-assisted photocatalytic process were monitored by UV absorption spectroscopy, HPLC methods, total organic carbon assays, and identification of intermediates using electrospray mass spectral techniques. Microwave thermal and nonthermal effects were delineated by comparing results from MW-generated internal heat versus conventional external heating, and at constant ambient temperature under a microwave field. Factors involved in the nonthermal component of the microwave radiation were inferred for the initial adsorption of the substrate and its subsequent degradation occurring on the surface of TiO2 particles. Microwave effects bear on the mechanism through which a model substrate undergoes oxidative degradation. Mulberry leaf extracts were generated using four concentrations of ethanol (50%, 60%, 70%, and 95% v/v). A 60% ethanolic mulberry leaf extract (60E) yielded a high total phenolic content (TPC) and antioxidant activity using 1, 1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging and a ferric reducing antioxidant power (FRAP) assay. Coating materials were derived using a combination of soy protein isolates (SPI) and low methoxyl (LM) pectin in a 1:1 ratio. The effect of various parameters on microencapsulation, such as pH (3.5, 4.0, and 4.5) and the concentration of coating materials (2.5, 5.0 and 7.5% w/v), was studied. Microcapsules produced using 60E as a core material at pH 4.0 with 7.5% of coating material showed a high encapsulation yield, encapsulation efficiency, TPC and antioxidant activity. 相似文献

The identification of 20 strains of yeasts isolated from foods by means of DNA sequence analysis with two kinds of universal primers for the rDNA region was examined, and the results were compared with those of the conventional phenotyping test using API 20C AUX. In the analysis of the 26S region, all 20 yeast strains tested were identified at the species level. In the ITS1 region, 16 strains were also classified at the species level. In addition, all results of DNA sequence analysis were consistent with those of the phenotyping test at the genus level. Furthermore, DNA sequence analysis was able to identify causative yeasts observed in two suspect foods, though phenotyping tests alone failed to identify them. 相似文献
