Nanoparticles are playing an increasing role in biomedical applications. Excitotoxicity plays a significant role in the pathophysiology of neurodegenerative diseases, such as Alzheimer’s or Parkinson’s disease. Glutamate ionotropic receptors, mainly those activated by N-methyl-D-aspartate (NMDA), play a key role in excitotoxic death by increasing intraneuronal calcium levels; triggering mitochondrial potential collapse; increasing free radicals; activating caspases 3, 9, and 12; and inducing endoplasmic reticulum stress. Neutral phosphorous dendrimers, acting intracellularly, have neuroprotective actions by interfering with NMDA-mediated excitotoxic mechanisms in rat cortical neurons. In addition, phosphorous dendrimers can access neurons inside human brain organoids, complex tridimensional structures that replicate a significant number of properties of the human brain, to interfere with NMDA-induced mechanisms of neuronal death. Phosphorous dendrimers are one of the few nanoparticles able to gain access to the inside of neurons, both in primary cultures and in brain organoids, and to exert pharmacological actions by themselves. 相似文献

When working on the synthesis of substituted cyclodextrins (CDs), the main challenge remains the analysis of the reaction media content. Our objective in this study was to fully characterise a complex isomers mixture of Lipidyl-βCDs (LipβCD) obtained with a degree of substitution 1 (DS = 1) from a one-step synthesis pathway. The benefit of tandem mass spectrometry (MS/MS) and ion mobility separation hyphenated with mass spectrometry (IM-MS) was investigated. The MS/MS fragment ion‘s relative intensities were analysed by principal component analysis (PCA) to discriminate isomers. The arrival time distribution (ATD) of each isomer was recorded using a travelling wave ion mobility (TWIM) cell allowing the determination of their respective experimental collision cross section (CCSexp). The comparison with the predicted theoretical CCS (CCSth) obtained from theoretical calculations propose a regioisomer assignment according to the βCD hydroxyl position (2, 3, or 6) involved in the reaction. These results were validated by extensive NMR structural analyses of pure isomers combined with molecular dynamics simulations. This innovative approach seems to be a promising tool to elucidate complex isomer mixtures such as substituted cyclodextrin derivatives. 相似文献

Toxoplasma gondii is unable to synthesize purines de novo, instead salvages them from its environment, inside the host cell, for which they need high affinity carriers. Here, we report the expression of a T. gondii Equilibrative Nucleoside Transporter, Tg244440, in a Trypanosoma brucei strain from which nucleobase transporters have been deleted. Tg244440 transported hypoxanthine and guanine with similar affinity (Km ~1 µM), while inosine and guanosine displayed Ki values of 4.05 and 3.30 µM, respectively. Low affinity was observed for adenosine, adenine, and pyrimidines, classifying Tg244440 as a high affinity oxopurine transporter. Purine analogues were used to probe the substrate-transporter binding interactions, culminating in quantitative models showing different binding modes for oxopurine bases, oxopurine nucleosides, and adenosine. Hypoxanthine and guanine interacted through protonated N1 and N9, and through unprotonated N3 and N7 of the purine ring, whereas inosine and guanosine mostly employed the ribose hydroxy groups for binding, in addition to N1H of the nucleobase. Conversely, the ribose moiety of adenosine barely made any contribution to binding. Tg244440 is the first gene identified to encode a high affinity oxopurine transporter in T. gondii and, to the best of our knowledge, the first purine transporter to employ different binding modes for nucleosides and nucleobases. 相似文献

The stereochemical features displayed by the N-glycosidic linkagein crystalline N-linked glycoproteins are analyzed. From thestatistical analysis of 44 different glycosylation sites belongingto 26 glycoproteins of the Brookhaven Protein Data Bank, a meanstandard geometry for the GlcNAc moiety, along with a rationalizationof its confornia-tional behavior, can be proposed. As for theglycopeptide linkage, the distribution of observed conformationshas been analyzed on the basis of molecular mechanics calculations.The rotamer distribution of the Asn side chains conforms tothat observed on non-glycosylated structures, and it agreeswith the pattern of flexible conformations gathered from NMRmeasurements. In characterizing the protein-glycan interactions,some hydrogen bonds occur. Stacking between the amphiphilicmoiety of the glycan and some surrounding aromatic, or at leasthydrophobic, amino acid residues is also found. When lookingat the secondary structure of the glycosylated peptide, only25% of the glycosylation sites correspond to situations whereAsn is located at the top of a ß-turn. Other typesof secondary structure exist which fulfil the spatial requirementof having the glycan exposed at the surface of the protein.These data can be compared with the most recent studies on thepeptide conformation which would be required for glycosylation. 相似文献

Wood sawdust gasification experiments were performed in a steam fluidized bed at 800 °C between 2 and 10 bar. An evolution of gas yields with time was measured during the tests, and especially an increase of hydrogen and carbon dioxide yields. This test duration effect was ascribed to char build-up in the bed. As tests proceed, the contribution of char steam gasification to gas yield increases, and the catalytic effect of char on hydrocarbons and tar conversion and on water-gas shift reaction is enhanced.As total pressure increases from 2 to 10 bar, hydrogen, carbon dioxide and methane yields increase by 16%, 53% and 38% respectively, whereas carbon monoxide yield decreases by 33%. The changes in gaseous yields with pressure can be partly explained by the influence of pressure on gas phase reactions (acceleration of water-gas shift kinetics and change in hydrocarbon reactions). The increase of methane yield with pressure is rather suggested to be linked to a change in secondary pyrolysis reactions scheme under high pressure. 相似文献

Experimental results on electrical conductivity and thermoelectric power for FeCl3-doped polyvinyl alcohol-polypyrrole composites, with polypyrrole content exceeding the percolation threshold, are presented. The electrical conductivity is analyzed in terms of the hopping theory, whereas the thermoelectric power displays a metallic-like dependence. A model involving both the intrinsic resistance of the fibrils (V.R.H. hopping mechanisms) and the resistance of the junctions between fibrils (fluctuations-induced tunneling conduction) has been retained. 相似文献
