5 antigens are identified in ascitic fluid from patients with ovarian tumours--AgA, AgB, AgV, AgG and AgD. Specific antisera are produced in rabbits by immunization with different protein and glycoprotein fractions of this fluid and by simultaneous injections of gamma-globulin fraction from antihuman plasma protein and pool of 25 ascitic fluids to rabbits. By the double immunodiffusion technique with standard test-systems for these antigens. It is found that all of them are absent in donor blood serum, but are present in certain normal and fetal tissues. AgA and AgD are also found in some extracts of lung, stomach, colon, and connective tissue tumours. The immunohistochemical and high-sensitive serological methods are necessary for determination of the usefulness of these antigens as tumour markers. 相似文献

A considerable blastomogenic effect of metabolite serotonin 5-methoxytryptamine (5-MOT) subcutaneously administered for a long time to C57BL/6 mice was established. This effect was decreased noticeably if the further metabolism of 5-MOT into 5-methoxyindolyl-3-acetic acid (5-MIAA) was blocked by pyrazidol. These results explain the fact that the blastomogenic effect of 5-MOT is not direct but is caused by the transformation of 5-MOT into its final carcinogenic metabolite 5-MIAA. 相似文献

The indirect immunofluorescent technique has been used to study the localization and distribution of one of the ascitic fluid antigens (AgD) in patients with ovary carcinoma on sections of normal and tumour tissues. 155 samples of human malignant tumours were studied by the immunofluorescent method and the antithymidine antibodies index-labelling (IL--% of tumour cells in the S-phase). Wide individual variability (0.1-73%) of IL of the studied tumours was shown to be present and to be preserved inside the tumours which were homogeneous by the histological type and developmental stage. Three benzisoxazole haptens designed to elicit antibody binding sites with widely differing polarity have been synthesized and used to induce antibodies in mice. Monoclonal antibodies were prepared using hybridoma technology, and screened for catalysis of the ring-opening isomerization and/or decarboxylation of a series of related benzisoxazoles and their 3-carboxy-derivatives. No catalysis of decarboxylation was observed, but 4 of a total of 47 antibodies obtained against the three haptens catalyzed the isomerization process. Of 12 antibodies raised against the 3-acetylbenzisoxazole structure 5 none was catalytically active; but one of 24 raised against a 3-isopropenylbenzisoxazole 6 increased the rate of ring-opening of 6-nitrobenzisoxazole, and 5 of 11 antibodies raised against a benzisoxazole 7 with a 3-amidinium group were moderately active against either 6-nitro or 6-acylaminobenzisoxazoles. Competitive binding studies suggest that at least some of the antibodies induced by the isopropenyl hapten do possess a recognizably hydrophobic binding site. 相似文献

The distribution of human embryonic prealbumin-1 (EPA-1) has been studied in 52 different malignant tumours using indirect immunofluorescent technique and two types of antibodies to different antigenic determinants: "EPA-1" and "EPA-1S". In mesenchymal tumours (except for desmoids and chondrosarcomas) this glycoproteid was observed in the cytoplasm of tumour cells, but "EPA-1S"--in the connective-tissue fibers only. Localization of glycoproteid EPA-1 depends on the type of the used primary antibodies. EPA-1 is also observed in the connective-tissue stroma of most of epithelial tumours. A conclusion is made that transformation of EPA-1 glycoproteid synthesis from definitive to fetal form occurs in malignant tumours. 相似文献

The possibility of poly(vinyl chloride) (PVC) plasticization and modification by regenerated PVC/polyurethane (PU) blends (PVC/PUR), obtained after partial thermochemical destruction of the PU foam‐backed PVC‐sheet (soundproof materials waste), has been studied. The partial thermochemical destruction of the PU foam‐backed PVC‐sheet was carried out using alkanolamine in a one‐screw extruder. The destruction product was a thermoplastic mixture of PVC and regenerated linear PUs, applicable for reuse without any purification and fractionation. The influence of processing conditions on structure‐property relationships for the PVC/PUR blends was studied using the methods of differential scanning calorimetry (DSC), dynamic mechanical thermal analysis (DMTA), size exclusion chromatography (SEC), and mechanical testing. It has been established that PVC/PUR blends can be reused as an effective polymer plasticizer for PVC resin (PVCr) and as a modifier for plasticized PVC (PVCp). POLYM. ENG.
