Metamaterials with adjustable, sometimes unusual properties offer advantages over conventional materials with predefined mechanical properties in many technological applications. A group of metamaterials, called modular metamaterials or metastructures, are developed through the arrangement of multiple, mostly similar building blocks. These modular structures can be assembled using prefabricated modules and reconfigured to promote efficiency and functionality. Herein, a novel modular metastructure is developed by taking advantage of the high compliance of preprogrammable double-spirals. First, the mechanical behavior of a four-module metastructure under tension, compression, rotation, and sliding is simulated using the finite-element method. Then, 3D printing and mechanical testing are used to illustrate the tunable anisotropic and asymmetric behavior of the spiral-based metastructures in practice. The results show the simple reconfiguration of the presented metastructure toward the desired functions. The mechanical behavior of single double-spirals and the characteristics that can be achieved through their combinations make our modular metastructure suitable for various applications in robotics, aerospace, and medical engineering.  相似文献

Capillary electrophoresis (CE) has been combined with matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) and radionuclide detection to assay mass-limited biological samples. Nanovial sampling techniques enable injections into the CE capillary from 50 to 150-nL volume samples; after the separation, nanoliter fraction collection combines the CE effluent with a MALDI matrix and minimizes sample spreading, thus allowing both MALDI MS and radionuclide detection on the CE fractions. MALDI MS complements the elution time information of CE by providing accurate molecular mass data, and radionuclide detection provides zeptomole limits of detection with quantitative information. While MALDI MS detects all fully processed peptides at sufficient concentration, culturing the neuron in media containing 35S-Met provides selective radionuclide detection of newly synthesized methionine-containing peptides. The analysis and detection of the expected neuropeptides and hormones in a single 40-microm bag cell neuron from Aplysia californica with CE/MALDI MS/radionuclide detection demonstrates the ability of this hyphenated approach to work with chemically complex mass-limited samples.  相似文献

At present, much attention is paid to the use of antimicrobial peptides (AMPs) of natural and artificial origin to combat pathogens. AMPs have several points that determine their biological activity. We analyzed the structural properties of AMPs, as well as described their mechanism of action and impact on pathogenic bacteria and viruses. Recently published data on the development of new AMP drugs based on a combination of molecular design and genetic engineering approaches are presented. In this article, we have focused on information on the amyloidogenic properties of AMP. This review examines AMP development strategies from the perspective of the current high prevalence of antibiotic-resistant bacteria, and the potential prospects and challenges of using AMPs against infection caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2).  相似文献

The exhibit 10 Short Films About Fog, which was part of the Data & Matter Exhibition at Palazzo Bembo during the Venice Architecture Biennale in 2018, documents and expands experiments into the relationship between fog, textiles, and air. The project sought to probe whether the otherwise formless phenomena of fog could be explicitly formed, and how it might assume structures and textures. By revealing the characteristics of fog that fascinate us, yet are latent, the short films also transform the phenomena through magnification, thereby amplifying inherent behaviors and qualities.  相似文献

Significant antibacterial properties of non-thermal plasma (NTP) have converted this technology into a promising alternative to the widespread use of antibiotics in assisted reproduction. As substantial data available on the specific in vitro effects of NTP on male reproductive cells are currently missing, this study was designed to investigate selected quality parameters of human spermatozoa (n = 51) exposed to diffuse coplanar surface barrier discharge NTP for 0 s, 15 s, 30 s, 60 s and 90 s. Sperm motility characteristics, membrane integrity, mitochondrial activity, production of reactive oxygen species (ROS), DNA fragmentation and lipid peroxidation (LPO) were investigated immediately following exposure to NTP and 2 h post-NTP treatment. Exposure to NTP with a power input of 40 W for 15 s or 30 s was found to have no negative effects on the sperm structure or function. However, a prolonged NTP treatment impaired all the sperm quality markers in a time- and dose-dependent manner. The most likely mechanism of action of high NTP doses may be connected to ROS overproduction, leading to plasma membrane destabilization, LPO, mitochondrial failure and a subsequent loss of motility as well as DNA integrity. As such, our findings indicate that appropriate plasma exposure conditions need to be carefully selected in order to preserve the sperm vitality, should NTP be used in the practical management of bacteriospermia in the future.  相似文献

The growth in the number of chronic non-communicable diseases in the second half of the past century and in the first two decades of the new century is largely due to the disruption of the relationship between the human body and its symbiotic microbiota, and not pathogens. The interaction of the human immune system with symbionts is not accompanied by inflammation, but is a physiological norm. This is achieved via microbiota control by the immune system through a complex balance of pro-inflammatory and suppressive responses, and only a disturbance of this balance can trigger pathophysiological mechanisms. This review discusses the establishment of homeostatic relationships during immune system development and intestinal bacterial colonization through the interaction of milk glycans, mucins, and secretory immunoglobulins. In particular, the role of fucose and fucosylated glycans in the mechanism of interactions between host epithelial and immune cells is discussed.  相似文献

Eight iron aluminide alloys with different contents of V and Cr were prepared up to 25 at. pct of both elements. The effect of the V and Cr concentration on the microstructure and mechanical properties was investigated by several complementary techniques. This investigation revealed that the microstructure of all the investigated alloys was comparable regardless of their chemical composition. All the alloys were in a solid solution condition without any major chemical inhomogeneity. For all alloys, a comparable grain size and D03 crystallographic structure was observed. In situ X-ray diffraction measurement revealed that the crystallographic structure was stable up to 1073 K (800 °C) regardless of the chemical composition. Mechanical testing showed that the compressive yield stress significantly increased with the increasing total sum of V plus Cr. Much higher values of yield stress were measured for symmetric concentrations of V and Cr when compared to non-symmetric ones. Eventually, it was shown that the formation of at least a rough system of the lattice positions occupation by four types of atoms in four sub-lattices derived from D03 is the most probable strengthening factor for alloys with symmetric concentrations of V and Cr.  相似文献

Structural integrity and functional characteristics of biomacromolecules are largely defined by electrostatic forces between ionized moieties, which are often altered at interfaces. Unraveling these changes requires access to charge state and structure of surface-confined biopolymers in aqueous environments. We therefore combined electrokinetic measurements of interfacial electrical potentials with the simultaneous determination of the optical layer thickness by reflectometric interference spectroscopy. Two examples are summarized to demonstrate the resulting options: The pH-switching of grafted poly(l-glutamic acid) layers caused by dissociation-dependent helix-coil transitions was studied; potential distribution and ion mobility within the grafted polyelectrolyte were unraveled using an new theoretical model for the charging of polyelectrolyte layers. The charge-driven modulation of biopolymers at interfaces was furthermore analyzed in the adsorption of fibronectin onto polymer substrates with varied charge density; the results permit us to reach a conclusion about the relevance of electrostatic matching for orientation and anchorage of the protein. Altogether, the introduced methodology was found suitable to follow the electrosurface characteristics of biomacromolecules in situ.  相似文献

The peptide content of individual mammalian cells is profiled using matrix-assisted laser desorption/ionization (MALDI) time-of-flight mass spectrometry. Both enzymatic and nonenzymatic procedures, including a glycerol cell stabilization method, are reported for the isolation of individual mammalian cells in a manner compatible with MALDI MS measurements. Guided microdeposition of MALDI matrix allows samples to be created with suitable analyte-to-matrix ratios. More than 15 peptides are observed in individual rat intermediate pituitary cells. The combination of accurate mass data, expected cleavages by proteolytic enzymes, and postsource decay sequencing allows identification of 14 of these peptides as pro-opiomelanocortin prohormone-derived molecules. These protocols permit the classification of individual mammalian cells by peptide profile, the elucidation of cell-specific prohormone processing, and the discovery of new signaling peptides on a cell-to-cell basis in a wide variety of mammalian cell types.  相似文献
