Poly(ethylene oxide)‐g‐gellan polysaccharide nanocarriers for controlled gastrointestinal delivery of simvastatin 下载免费PDF全文
Herein, a novel gellan polysaccharide‐based amphiphilic copolymer was synthesized for the development of simvastatin‐loaded micellar nanoparticles. The nanoparticles were explored for their controlled drug release and improved pharmacodynamic potentials. The copolymer was characterized by Fourier transform infrared spectroscopy (FTIR) and elemental analysis. The onset of copolymer micellization was detected by fluorescence spectroscopy. Simvastatin was loaded into micellar particles by solvent evaporation method and the particles were then characterized by microscopic and light scattering techniques. The physical state of drug was studied by X‐ray diffraction analysis. Pharmacodynamic assessment of the micellar preparations was done on rabbit models. The copolymer formed micellar nanoparticles in water. Critical micellar concentration was 9.12mg/l. The micellar particles (426.8–912.6nm) entrapped a maximum of 18.86% drug. Higher negative zeta potential indicated physical stability of micellar systems. A simple diffusion mechanism was operative in the event of comparatively faster drug release in pH6.8 phosphate buffer solution. No significant drug‐copolymer interaction was traced by FTIR spectroscopy. The amorphization of drug into micellar particles reduced LDL‐cholesterol level by ~45% in hyperlipidemic rabbits and this was about 2.5 times higher than pure drug dispersion. Copolymer micellar nanoparticles of simvastatin could control cholesterol level in hyperlipidemic rabbits and thus had potential in drug delivery applications. © 2015 Wiley Periodicals, Inc. J. Appl. The methodology of response reactions (RERs) introduced earlier from thermodynamic and kinetic considerations is used in this work to develop a new algorithm for the classification and enumeration of unique/direct reaction routes (RRs) and overall reactions (ORs). According to the RERs approach, a unique set of both RRs and ORs may be generated starting from any conceivable set of linearly independent RRs and ORs. In particular, the direct ORs may be most conveniently enumerated starting from the formula matrix of the terminal species (reactants and products), i.e., without any relation to the elementary reactions comprising the detailed mechanism. Depending on the type of ORs produced by the RRs one can distinguish between two distinct types of direct RRs. Namely, one option is to define a direct RR by specifying the intermediate species that need to be eliminated. This type of RR is referred to as Milner RRs. Polyurethane (PU) recycling is a topic of growing interest due to the increasing amount of polyurethane waste. The main purpose of polyurethane chemical recycling is to recover the starting polyol. In this study, a method of polyurethane thermochemical recycling, glycerolysis by means of crude glycerin, is proposed. This work presents a comparative study of commercial catalysts used in order to accelerate the decomposition process, namely triethylamine (TEA), potassium acetate (KAc), 1,4‐diazabicyclo[2.2.2]octane (DABCO), sodium hydroxide (NaOH), dibutyltin dilaurate (DbDl), and stannous octoate (StOc).The effect of used catalyst on the chemical structure and rheological properties was studied. The type of catalyst does not have significant influence on the chemical structure, but causes different course of reaction: split‐ and single‐phase in applied conditions. Glycerolysates were measured by Brookfield Rheometer. It was found that repolyols can be described by the Herschel–Bulkely mathematical model in the best accuracy. Effect of plasticizer type on the kraft lignin–natural rubber composite microstructure and selected properties was determined. The composites were prepared with addition of a commonly used naphthenic oil plasticizer to study the decomposition product of polyurethane (glycerolysate) and its characteristics. Kraft lignin powder was incorporated into the natural rubber matrix in amounts of 10 and 40 parts per 100 parts of natural rubber (phr). The reference samples were prepared without any lignin present. The chemical interaction between the filler particles and natural rubber macromolecules was analyzed by Fourier transform infrared spectroscopy (FTIR) and the adhesion was characterized by scanning electron microscopy (SEM). The results of the adhesion measurements confirmed poor distribution of lignin particles into the natural rubber matrix with increasing filler content. Optimal lignin content in the composites was 10 phr in the case of both plasticizers. Moreover, the results of FTIR verified the formation of non-covalent bonds and the need for modification of the filler to enhance the reinforcing effect in the natural rubber matrix. Dynamic mechanical analysis (DMA) and mechanical measurements proved that the specimen containing 10 phr of lignin with the use of glycerolysate as plasticizer displayed the highest mechanical performance. It was demonstrated that glycerolysate and naphthenic oil as plasticizing agents showed similar effect on the thermal properties of the prepared composites. The study aimed to investigate the effectiveness of porous calcium phosphates viz., hydroxyapatite (HAp) and a bi-phasic calcium phosphate (BCP) with predominately β-tricalcium phosphate (β-TCP) prepared by aqueous solution combustion method impregnated with cefuroxime axetil for the treatment of experimental osteomyelitis and compared with parenteral treatment. In vitro release of the drug was tested for its sustained elution characteristics for 21 days in PBS (pH 7.2) and measured by HPLC. In the in vivo study, bone infection was induced in tibia of rabbits by inoculation of 1 ml (3 × 106) CFU Staphylococcus aureus. On the 21st day, after the development of osteomyelitis, six animals were treated by filling the cavity with cefuroxime-impregnated HAp blocks (Group II), six animals with the same drug-impregnated β-TCP (Group III) and in six others, only cefuroxime (15 mg/kg twice daily) was injected parenterally 6 weeks (Group IV). Group I with six animals was kept untreated. Histologically, the signs of infection were found to subside by 3 and 6 weeks. Radiological evaluation with cefuroxime-impregnated HAp and β-TCP pointed out the disappearance of sequestrum and existence of newly formed bony specules. Concentration of cefuroxime in bone and serum as estimated by HPLC showed highest value on day 21 itself which reduced marginally by day 42 in both the groups and these values were higher than minimum inhibitory concentration (MIC) against S. aureus. Our findings suggest that bi-phasic calcium phosphates with predominately β-TCP content is a very efficient carrier material for antibiotic compounds even for refractory infections by S. aureus. 相似文献

The effects of the algicides terbutryn and copper sulfate on the potential for reducing the gaseous loss of NH3 from urea applied to rice were examined in experiments with 2 methods of N fertilizer management, 2 or 3 N rates, and 3 algicide treatments. The experiments were conducted during the 1986 dry and wet seasons in an experimental field at Pila, Laguna, Philippines.Copper sulfate had little effect as an algicide at the rate used, but terbutryn immediately reduced algal growth. The populations of species resistant to terbutryn probably increased, but terbutryn had no long-term effect on the total number of colony-forming units of algae. There was some evidence that terbutryn reduced photodependent N2 fixation as estimated by acetylene reduction assay.Terbutryn, when applied with urea 10 days after transplanting, reduced the maximum floodwater pH by 0.9 units or more for 7 d in the DS and by about 0.5 units for 8 d in the WS. Terbutryn increased the ammoniacal-N (AN) concentration in the floodwater 100% or more in the DS and 60% in the WS. The combined effect of terbutryn on the floodwater pH and AN concentration was reduced photodependent NH3 partial pressure (NH3), about 25% in the DS and 38% in the WS.

Combination heating of food using microwave and jet impingement has been simulated by coupling Maxwell’s equations of electromagnetics with energy equation and using experimentally measured heat transfer coefficient values for jet impingement in a novel domestic oven. Transient food temperatures from the model and experiment for each separate heating mode and their combination revealed the characteristic nature of each of the heating modes. Contour plots of temperature show that with combination heating, surface can be heated faster (for crispness) and edge over-heating can be partially avoided. Measures of non-uniformity in temperatures in the heated food are developed using coefficient of variation and middle 80-percentile range as the parameters. Using these measures, it is shown that combination heating leads to more uniform heating, without compromising the speed or convenience. A 22–30% increase in uniformity has been observed for combination microwave–jet impingement heating over microwave-only heating. Jet impingement is a good complement to microwave heating as it has different spatial and time variation of heating rates. During the initial period, jet impingement dominates over microwave heating near the surface, with microwave heating being more significant in the interior. At later times, the roles switch with microwaves becoming more dominant on the surface while jet impingement takes a more significant role in heating the interior of the food. These findings should help the product, process and equipment designer achieve the balance between speed and uniformity of heating in a more precise manner. 相似文献
