The diversity of bacterial groups in activated sludge from large- and small-scale wastewater treatment plants was explored by barcoded pyrosequencing of 16S rRNA gene. Activated sludge samples (three small and 17 large scale) were collected from 12 wastewater treatment plants to clarify precise taxonomy and relative abundances. DNA was extracted, and amplified by 4 base barcoded 27f/519r primer set. The 454 Titanium (Roche) pyrosequences were obtained and analyses performed by Quantitative Insight Into Microbial Ecology (QIIME) with around 100,000 reads. Sequence statistics were computed, while constructing a phylogenetic tree and heatmap. Computed results explained total microbial diversity at phylum and class level and resolution was further extended to Operational Taxonomic Unit (OTU) based taxonomic assignment for investigating community distribution based on individual sample. Composition of sequence reads were compared and microbial community structures for large- and small-scale treatment plants were identified as major phyla (Proteobacteria and Bacteroidetes) and classes (Betaproteobacteria and Bacteroidetes). Also, family level breakdowns were explained and differences in family Nitrospiraceae and phylum Actinobacteria found at their species level were also illustrated. Thus, the pyrosequencing method provides high resolution insight into microbial community structures in activated sludge that might have been unnoticed with conventional approaches.  相似文献

A novel trilayered controlled‐release nitrogen, phosphorous, and potassium (NPK) fertilizer hydrogel was prepared by dipping the NPK fertilizer granules sequentially in 7% w v?1 poly(vinyl alcohol) (PVA) and 2% w v?1 chitosan (CS) solutions and then cross‐linking the CS layer (cross‐CS) via glutaraldehyde vapor deposition. Different NPK fertilizer hydrogels were then synthesized by inverse suspension polymerization of the dried PVA/cross‐CS bilayer‐coated fertilizer granules in various molar ratios of acrylamide (AM) and acrylic acid (AA) monomers, and polymerization with varying molar ratios of ammonium persulfate, N,N,N′,N′‐tetramethylethylenediamine and N,N′‐methylenebisacrylamide (N‐MBA). The water dissolution time of the obtained PVA/cross‐CS/poly (AA‐co‐AM) trilayer‐coated NPK fertilizer hydrogel granules was prolonged, while the water absorbency increased with increasing AA contents, and decreased with increasing N‐MBA contents in the outer poly(AA‐co‐AM) coating. The optimal trilayer‐coated NPK fertilizer hydrogel obtained released 84 ± 18, 63 ± 12, and 36 ± 15% of the N, P, and K nutrients, respectively, after a 30‐day immersion in water. The release phenomena of the N, P, and K nutrients of the fertilizer hydrogel obeyed both the Korsmeyer‐Peppas and Ritger‐Peppas models with a pseudo‐Fickian diffusion mechanism. © 2014 Wiley Periodicals, Inc. Accumulating evidence suggests that the gut microbiome influences the brain functions and psychological state of its host via the gut–brain axis, and gut dysbiosis has been linked to several mental illnesses, including major depressive disorder (MDD). Animal experiments have shown that a depletion of the gut microbiota leads to behavioral changes, and is associated with pathological changes, including abnormal stress response and impaired adult neurogenesis. Short-chain fatty acids such as butyrate are known to contribute to the up-regulation of brain-derived neurotrophic factor (BDNF), and gut dysbiosis causes decreased levels of BDNF, which could affect neuronal development and synaptic plasticity. Increased gut permeability causes an influx of gut microbial components such as lipopolysaccharides, and the resultant systemic inflammation may lead to neuroinflammation in the central nervous system. In light of the fact that gut microbial factors contribute to the initiation and exacerbation of depressive symptoms, this review summarizes the current understanding of the molecular mechanisms involved in MDD onset, and discusses the therapeutic potential of probiotics, including butyrate-producing bacteria, which can mediate the microbiota–gut–brain axis.  相似文献

ABSTRACT: The 80% ethanol extracts of 5 purple-fleshed sweet potato cultivars were separated into 2 fractions, anthocyanins- and phenolic compounds-rich fractions, to clarify the contribution of these constituents to the radical-scavenging activity. The separation was accomplished with an ethyl acetate liquid/liquid extraction. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity in each fraction and the contributors varied according to the cultivars. The dominant DPPH radical-scavengers in "Ayamurasaki" and "Kyushu-132" were anthocyanins rather than phenolic compounds, while those in "Miyanou-36" and "Bise" were phenolic compounds, such as chlorogenic acid. Furthermore, the high-performance liquid chromatography analysis of anthocyanins showed that "Ayamurasaki" and "Kyushu-132" were rich in anthocyanins with peonidin aglycon, whereas "Miyanou-36,""Bise," and "Tanegashimamurasaki" contained cyanidin aglycon.  相似文献

The grafting of the methyl methacrylate (MMA) monomer onto natural rubber using potassium persulfate as an initiator was carried out by emulsion polymerization. The rubber macroradicals reacted with MMA to form graft copolymers. The morphology of grafted natural rubber (GNR) was determined by transmission electron microscopy and it was confirmed that the graft copolymerization was a surface‐controlled process. The effects of the initiator concentration, reaction temperature, monomer concentration, and reaction time on the monomer conversion and grafting efficiency were investigated. The grafting efficiency of the GNR was determined by a solvent‐extraction technique. The natural rubber‐g‐methyl methacrylate/poly(methyl methacrylate) (NR‐g‐MMA/PMMA) blends were prepared by a melt‐mixing system. The mechanical properties and the fracture behavior of GNR/PMMA blends were evaluated as a function of the graft copolymer composition and the blend ratio. The tensile strength, tear strength, and hardness increased with an increase in PMMA content. The tensile fracture surface examined by scanning electron microscopy disclosed that the graft copolymer acted as an interfacial agent and gave a good adhesion between the two phases of the compatibilized blend. © 2001 John Wiley & Sons, Inc. This paper describes the daily concentrations of trace metals and ionic constituents in the aerosol of Beijing, China from March 2001 to August 2003. Daily PM10 concentrations were also measured from September 2001 to August 2003. The daily average PM10 concentration at Beijing, China from September 2001 to August 2003 was 171+/-117 microg m(-3) (n = 673), which is 5-fold higher than at Yokohama, Japan. Trace metal concentrations were analyzed by using inductively coupled plasma mass spectrometry equipped with a laser ablation sample introduction (LA/ICP-MS), which is a rapid and simultaneous method for multi-element analysis. The daily average metal concentrations in TSP in Beijing from March 2001 to August 2003 were: Al: 3.5+/-2.4 (n = 727), Ti: 0.47+/-0.35 (n = 720), V: 0.013+/-0.010 (n = 716), Cr: 0.019+/-0.015 (n = 618), Mn: 0.24+/-0.16 (n = 730), Fe: 5.5+/-3.9 (n = 728), Co: 0.0046+/-0.0055 (n = 629), Ni: 0.022+/-0.024 (n = 680), Cu: 0.11+/-0.11 (n = 660), Zn: 0.77+/-0.60 (n = 726), As: 0.048+/-0.047 (n = 731), Se: 0.010+/-0.010 (n = 550), Cd: 0.0068+/-0.0082 (n = 709), Sb: 0.033+/-0.036 (n = 687), and Pb: 0.43+/-0.50 (n = 728) (unit, microg m(-3)). All the metal concentrations in TSP in Beijing, China were 1.7-21.8 times higher than those in TSP in the center of Tokyo, Japan. Notably, As concentrations in TSP in Beijing were 20-fold higher than those in Tokyo. Source identification of aerosols in Beijing was carried out by using the chemical mass balance (CMB) receptor model, with the daily concentration of metals in the aerosol. The major primary sources of the aerosol of Beijing were considered to be soil dust and coal combustion. Vehicle exhaust contribution tended to increase.  相似文献

This paper presents results of a design study on a semiconductor coupled surface acoustic wave (SAW) convolver in which bi-directionally propagating SAWs, on a piezoelectric substrate with a high coupling coefficient, couple with bonded semiconductor diodes through multistrips. To obtain convolution signals with a high efficiency, we adopted a diode-balanced bridge structure for the nonlinear operation. We also found that the tapping pitches of the multi-strip electrodes have robustness against operation frequency variation and temperature-dependent variation on the delay of the SAW. We verified the effectiveness of the device in. a circuit simulation and an experiment on a test circuit, which was fabricated by using an epitaxial lift-off film-bonding process.  相似文献

This paper proposes a novel framework for developing adaptive and scalable network services. In the proposed framework, a network service is implemented as a group of autonomous agents that interact in the network environment. Agents in the proposed framework are autonomous and capable of simple behaviors (e.g., replication, migration, and death). In this paper, an evolutionary adaptation mechanism is designed using genetic algorithms (GAs) for agents to evolve their behaviors and improve their fitness values (e.g., response time to a service request) to the environment. The proposed framework is evaluated through simulations, and the simulation results demonstrate the ability of autonomous agents to adapt to the network environment. The proposed framework may be suitable for disseminating network services in dynamic and large-scale networks where a large number of data and services need to be replicated, moved, and deleted in a decentralized manner.  相似文献

Oligoribonucleotides containing four G clusters, (UGnU)4 (n = 3-5), were synthesized and characterized by electrophoretic analysis and resistance to nucleases. Electrophoretic analyses indicate that the oligomers except for (UG3U)4 can form antiparallel-stranded monomeric quadruplexes in the presence of potassium cation. The oligomers used in this study are protected from endonuclease P1 and venom phosphodiesterase degradation. In addition, the oligomers indicate resistance to nucleases in human serum and E. coli S-100 fraction.  相似文献
