The aim of this study was to analyze the expression of peroxisome proliferator-activated receptor γ (PPARγ) and retinoid X receptor α (RxRα), a binding heterodimer playing a pivotal role in the successful trophoblast invasion, in the placental tissue of preeclamptic patients. Furthermore, we aimed to characterize a possible interaction between PPARγ and H3K4me3 (trimethylated lysine 4 of the histone H3), respectively H3K9ac (acetylated lysine 9 of the histone H3), to illuminate the role of histone modifications in a defective trophoblast invasion in preeclampsia (PE). Therefore, the expression of PPARγ and RxRα was analyzed in 26 PE and 25 control placentas by immunohistochemical peroxidase staining, as well as the co-expression with H3K4me3 and H3K9ac by double immunofluorescence staining. Further, the effect of a specific PPARγ-agonist (Ciglitazone) and PPARγ-antagonist (T0070907) on the histone modifications H3K9ac and H3K4me3 was analyzed in vitro. In PE placentas, we found a reduced expression of PPARγ and RxRα and a reduced co-expression with H3K4me3 and H3K9ac in the extravillous trophoblast (EVT). Furthermore, with the PPARγ-antagonist treated human villous trophoblast (HVT) cells and primary isolated EVT cells showed higher levels of the histone modification proteins whereas treatment with the PPARγ-agonist reduced respective histone modifications. Our results show that the stimulation of PPARγ-activity leads to a reduction of H3K4me3 and H3K9ac in trophoblast cells, but paradoxically decreases the nuclear PPARγ expression. As the importance of PPARγ, being involved in a successful trophoblast invasion has already been investigated, our results reveal a pathophysiologic connection between PPARγ and the epigenetic modulation via H3K4me3 and H3K9ac in PE.  相似文献

Shipwrecks and dumped munition continue to be a major hazard, both in the North Sea but also on a global scale. Research within the EU Interreg project North Sea Wrecks (NSW), in cooperation with the German Aerospace Centre, Institute for the Protection of Maritime Infrastructures (DLR), is generating new insights into the status of wrecks, the potential leakage of pollutants from remaining munitions loads and the effects of contamination on exposed marine organisms in the North Sea environment. Further, historical documents are generated from archives to describe ship's history and sinking scenario. These historical findings were compared to models and images of the visual inspections of the wrecks. Further, samples of water, sediment and organisms are being analysed for traces of explosives. Combining the results of these different fields of research allows for a better understanding of the environmental risks deriving from these wrecks. This process is shown below by focusing on the wreck of the German light cruiser SMS MAINZ, which sank in 1914. Data were compared to three additional wrecks situated also within the southern German Bight. Available data about the wrecks were preliminary assessed using a wreck risk model. Finally, wrecks were ranked according to their potential environmental risk.  相似文献

Methods for postirradiation characterization of bulk (cm3) irradiated materials or even spent nuclear fuels are sparse due to their extremely radioactive nature. While several methods exist to characterize smaller volumes (<?1 mm3) of such samples, selecting these volumes from larger samples is challenging. X-ray-based methods are prohibitive due to the strong γ-radiation from the sample flooding the detectors. Neutron-based methods available in the proximity of irradiation reactors allow for thermal neutron radiography or computed tomography using a small reactor source, but one cannot assess isotope distributions or microstructural features such as phases, texture, or strain from diffraction measurements due to flux limitations. We present herein a pathway to provide pulsed neutron characterization of bulk irradiated samples using time-of-flight neutron diffraction for microstructural characterization and energy-resolved neutron imaging for assessment of isotopic densities and distributions. Ultimately, laser-driven pulsed neutron sources may allow deployment of these techniques pool-side at irradiation reactors.

Plant U-box armadillo repeat (PUB-ARM) ubiquitin (Ub) ligases have important functions in plant defense through the ubiquitination of target proteins. Defense against pathogens involves vesicle trafficking and the formation of extracellular vesicles. The PUB-ARM protein SENESCENCE ASSOCIATED UBIQUITIN E3 LIGASE1 (SAUL1) can form patches at the plasma membrane related to tethering multi-vesicular bodies (MVBs) to the plasma membrane. We uncovered the structure of a full-length plant ubiquitin ligase and the structural requirements of SAUL1, which are crucial for its function in patch formation. We resolved the structure of SAUL1 monomers by small-angle X-ray scattering (SAXS). The SAUL1 model showed that SAUL1 consists of two domains: a domain containing the N-terminal U-box and armadillo (ARM) repeats and the C-terminal ARM repeat domain, which includes a positively charged groove. We showed that all C-terminal ARM repeats are essential for patch formation and that this function requires arginine residue at position 736. By applying SAXS to polydisperse SAUL1 systems, the oligomerization of SAUL1 is detectable, with SAUL1 tetramers being the most prominent oligomers at higher concentrations. The oligomerization domain consists of the N-terminal U-box and some N-terminal ARM repeats. Deleting the U-box resulted in the promotion of the SAUL1 tethering function. Our findings indicate that structural changes in SAUL1 may be fundamental to its function in forming patches at the plasma membrane.  相似文献

Epigenetics play a vital role in early embryo development. Offspring conceived via assisted reproductive technologies (ARTs) have a three times higher risk of epigenetic diseases than naturally conceived children. However, investigations into ART-associated placental histone modifications or sex-stratified analyses of ART-associated histone modifications remain limited. In the current study, we carried out immunohistochemistry, chip-sequence analysis, and a series of in vitro experiments. Our results demonstrated that placentas from intra-cytoplasmic sperm injection (ICSI), but not in vitro fertilization (IVF), showed global tri-methylated-histone-H3-lysine-4 (H3K4me3) alteration compared to those from natural conception. However, for acetylated-histone-H3-lysine-9 (H3K9ac) and acetylated-histone-H3-lysine-27 (H3K27ac), no significant differences between groups could be found. Further, sex -stratified analysis found that, compared with the same-gender newborn cord blood mononuclear cell (CBMC) from natural conceptions, CBMC from ICSI-boys presented more genes with differentially enriched H3K4me3 (n = 198) than those from ICSI-girls (n = 79), IVF-girls (n = 5), and IVF-boys (n = 2). We also found that varying oxygen conditions, RNA polymerase II subunit A (Polr2A), and lysine demethylase 5A (KDM5A) regulated H3K4me3. These findings revealed a difference between IVF and ICSI and a difference between boys and girls in H3K4me3 modification, providing greater insight into ART-associated epigenetic alteration.  相似文献

Summary: Superhydrophobic surfaces are generated by a simple one‐step method of electrospinning of fluorinated homopolymers and copolymers of PFS. The hydrophobicity and superhydrophobicity can be changed by simply changing the surface morphology, which is possible by changing the electrospinning conditions. The appropriate combination of surface morphology and fluorinated materials led to the formation of super‐water‐resistant coatings showing the ‘water‐roll’ effect at an angle of 0°, i.e. placement of water droplets on such surfaces was not possible as they immediately rolled away. The effect is compared with the corresponding nonfluorinated PS and found to be clearly distinct in terms of water‐roll effect. Incorporation of about 30 mol‐% PFS onto the PS backbone could also convert hydrophobic PS surfaces to superhydrophobic surfaces. The effect is generalized by also using a new fluorinated poly(p‐xylylene) derivative. The molecular weight of the polymers has no noticeable effect on hydrophobicity/superhydrophobicity behavior.

Boiling phenomena are an important aspect in security and efficiency for technical applications with high heat flux like nuclear reactors. This study presents optical coherence tomography (OCT) as a novel modality for three-dimensional and two-dimensional time resolved imaging of nucleate boiling on heated surfaces on a microscopic scale with high spatial (<10 μm) and temporal (>25 frames per second) resolution. Within this study, a borosilicate glass plate coated with an optically transparent and electrically conductive indium tin oxide (ITO) layer with a thickness of approximately 100 nm was used as heating surface. The combination of these two properties allows optical inspection of the nucleate boiling from the backside by OCT focused on the formation, growth and detachment of single bubbles. We demonstrated for the first time that OCT is an excellent tool to acquire two-dimensional and three-dimensional images of the base of vapor bubbles from the backside of the heated surface. The acquired images allow for instance the temporally resolved measurement of the bubble diameter, diameter of the bubble base and the contact angle. Exploiting the phase information of the acquired OCT signal stacks allows imaging the movement of the bubble surrounding fluid. We think that OCT will provide many new insights into the boiling phenomena at the bubble base. The recent enhancement of the acquisition rates of OCT systems will facilitate four-dimensional imaging of single bubble evaporation procedures in the nearer future.  相似文献
