Chronic white adipose tissue (WAT) inflammation has been recognized as a critical early event in the pathogenesis of obesity-related disorders. This process is characterized by the increased residency of proinflammatory M1 macrophages in WAT. However, the lack of an isogenic human macrophage-adipocyte model has limited biological studies and drug discovery efforts, highlighting the need for human stem cell-based approaches. Here, human induced pluripotent stem cell (iPSC) derived macrophages (iMACs) and adipocytes (iADIPOs) are cocultured in a microphysiological system (MPS). iMACs migrate toward and infiltrate into the 3D iADIPOs cluster to form crown-like structures (CLSs)-like morphology around damaged iADIPOs, recreating classic histological features of WAT inflammation seen in obesity. Significantly more CLS-like morphologies formed in aged and palmitic acid-treated iMAC-iADIPO-MPS, showing the ability to mimic inflammatory severity. Importantly, M1 (proinflammatory) but not M2 (tissue repair) iMACs induced insulin resistance and dysregulated lipolysis in iADIPOs. Both RNAseq and cytokines analyses revealed a reciprocal proinflammatory loop in the interactions of M1 iMACs and iADIPOs. This iMAC-iADIPO-MPS thus successfully recreates pathological conditions of chronically inflamed human WAT, opening a door to study the dynamic inflammatory progression and identify clinically relevant therapies.  相似文献

Burning rate characteristics and sensitivity characteristics of energetic mixtures composed of metal particles and oxidizers, the so called ‘pyrolants’, were studied experimentally. The pyrolants tested were made of various particle sizes of magnesium (Mg) and polytetrafluoroethylene (TF). Mg/TF pyrolant produces high combustion flame temperature, so it is used as heat sources and igniter pyrolants. The maximum flame temperature (Tf) is 3271 K at 0.1 MPa, and 3483 K at 1 MPa. These values are obtained when Mg concentration is 30%. When the Mg concentrations are less than 50% the pyrolants are not able to burn stably at one atmosphere. The burning rate increases with increasing the concentration of Mg and decreases with increasing the mean diameter of Mg particles at constant pressure. Explosive energy evaluated with drop hammer test decreased with increasing burning rate, so there is strong relationship between burning rate and explosive energy.  相似文献

Three types of glass/nylon 6 intermediate material forms-film stacking, uncommingled yarn, and commingled yarm-were selected study the correlations between the impregnating property and mechanical properties. The size of the glass fiber block to be filled with matrix and the porosity in glass fiber bundles by spearing out the fiber bundle was different in these materials. Unidirectional glass fiber reinforced thermoplastic composites were fabricated by compression molding. The being test was performed by using the three-point loading system, and the fracture behavior and the degree of impregnation were observed to examine the influence of processing conditions on the bending properties, relative to the form of the intermediate material. Bending strength increased, in accordance with the impregnating property, least in the film stacking form, second most in uncommingled yarn, and most in commingled yarn. The impregnating property was affected by the size of fiber blocks and the porosity in fiber bundles, because bending strength was improved by spreading out the fiber bundles. Commingled yarn is an excellent intermediate materials, which has both the fineness of matrix/fiber mixing and large porosity in fiber bundles.  相似文献
