Duchenne muscular dystrophy (DMD) is an X-linked recessive neuromuscular disorder with a prevalence of approximately 1 in 3500–5000 males. DMD manifests as childhood-onset muscle degeneration, followed by loss of ambulation, cardiomyopathy, and death in early adulthood due to a lack of functional dystrophin protein. Out-of-frame mutations in the dystrophin gene are the most common underlying cause of DMD. Gene editing via the clustered regularly interspaced short palindromic repeats (CRISPR) system is a promising therapeutic for DMD, as it can permanently correct DMD mutations and thus restore the reading frame, allowing for the production of functional dystrophin. The specific mechanism of gene editing can vary based on a variety of factors such as the number of cuts generated by CRISPR, the presence of an exogenous DNA template, or the current cell cycle stage. CRISPR-mediated gene editing for DMD has been tested both in vitro and in vivo, with many of these studies discussed herein. Additionally, novel modifications to the CRISPR system such as base or prime editors allow for more precise gene editing. Despite recent advances, limitations remain including delivery efficiency, off-target mutagenesis, and long-term maintenance of dystrophin. Further studies focusing on safety and accuracy of the CRISPR system are necessary prior to clinical translation.  相似文献

We investigated the effect of tofogliflozin, a sodium-dependent glucose cotransporter 2 inhibitor (SGLT2i), on retinal blood flow dysregulation, neural retinal dysfunction, and the impaired neurovascular coupling in type 2 diabetic mice. Tofogliflozin was added to mouse chow to deliver 5 mg/kg/day and 6-week-old mice were fed for 8 weeks. The longitudinal changes in the retinal neuronal function and blood flow responses to systemic hyperoxia and flicker stimulation were evaluated every 2 weeks in diabetic db/db mice that received tofogliflozin (n =6) or placebo (n = 6) from 8 to 14 weeks of age. We also evaluated glial activation and vascular endothelial growth factor (VEGF) expression by immunofluorescence. Tofogliflozin treatment caused a sustained decrease in blood glucose in db/db mice from 8 weeks of the treatment. In tofogliflozin-treated db/db mice, both responses improved from 8 to 14 weeks of age, compared with vehicle-treated diabetic mice. Subsequently, the electroretinography implicit time for the oscillatory potential was significantly improved in SGLT2i-treated db/db mice. The systemic tofogliflozin treatment prevented the activation of glial fibrillary acidic protein and VEGF protein expression, as detected by immunofluorescence. Our results suggest that glycemic control with tofogliflozin significantly improved the impaired retinal neurovascular coupling in type 2 diabetic mice with the inhibition of retinal glial activation.  相似文献

This study investigated whether hydrogels comprising hydrophilic cationic polymers have similar bactericidal effects. Bacteria were seeded on hydrogels and agar and their viability was assessed with time. Cationic hydrogels displayed bactericidal effects upon long-term bacterial contact. Furthermore, we assessed the areal density of cationic monomer unit of the cationic hydrogels, water content, and the initial elastic modulus. We examined correlations between each factor and bacterial death ratios; consequently, the bacterial death ratios were strongly correlated with the areal density of cationic hydrogel monomers. Elastic energy (Wel) generated at the cytomembrane ion-binding region and the cationic hydrogel and the cytomembrane interfacial energy (Wf) were estimated; consequently, Wel exceeded Wf at higher contact areas. The cationic hydrogel may extract cytomembranes with a reasonable adsorption area. Therefore, cationic hydrogels may be used as probes for ultrasonic echo to sterilize medical equipment.  相似文献

Persistent acid production by Lactobacillus delbrueckii subsp. bulgaricus during refrigerated storage is a major cause of reduced viability of probiotic strains such as Bifidobacterium breve in yoghurt. It was established that H+ -ATPase-defective mutants of lactic acid bacteria have reduced growth and metabolism in low pH environments. Therefore, the aim of this study was to evaluate inhibition of post-acidification and maintenance of B. breve viability in yoghurt fermented by L. delbrueckii subsp. bulgaricus mutants with reduced membrane-bound H+ -ATPase activity during refrigerated storage. Spontaneous neomycin mutants of L. delbrueckii subsp. bulgaricus that had a significantly (P < or = 0.05) reduced H+ -ATPase activity were successfully isolated. Yoghurt fermented using L. delbrueckii subsp. bulgaricus SBT0164 No. 55-1 (mutant) starter culture had markedly reduced post-acidification and maintained viability (> or = 10(8) CFU/ml) of both Bifidobacteruim breve JCM 1192(T) and Bifidobacteruim breve JCM 7017 during storage at 10 degrees C for 21 days. These results clearly showed that yoghurt fermented by mutants of L. delbrueckii subsp. bulgaricus with reduced membrane-bound H+ -ATPase activity has reduced post-acidification that prolongs viability of B. breve in yoghurt during refrigerated storage.  相似文献

The effects of difructose anhydride III (di-D-fructofuranose-1,2':2,3'-dianhydride; DFA III) administration (3% DFA III for 4 weeks) on rat intestinal microbiota were examined using denaturing gradient gel electrophoresis (DGGE). According to DGGE profiles, the number of bacteria related to Bacteroides acidofaciens and uncultured bacteria within the Clostridium lituseburense group decreased, while that of bacteria related to Bacteroides vulgatus, Bacteroides uniformis and Ruminococcus productus increased in DFA III-fed rat cecum. In the cecal contents of DFA III-fed rats, a lowering of pH and an increase in short chain fatty acids (SCFAs), especially acetic acid, were observed. The DFA III-assimilating bacterium, Ruminococcus sp. M-1, was isolated from the cecal contents of DFA III-fed rats. The strain had 98% similarity with R. productus ATCC 27340T (L76595), and mainly produced acetic acid. These results confirmed that the bacteria harmful to host health were not increased by DFA III administration. Moreover, DFA III stimulated the growth of Ruminococcus sp. M-1 producing acetic acid, which may alter the intestinal microbiota towards a healthier composition. It is expected that DFA III would be a new candidate as a prebiotic.  相似文献

We previously reported that a copolymer consisting of N-isopropylacrylamide (NIPAM) and benzophenone (BP) units, behaves as a photosensitizer showing temperature-controlled oxygenation activity in water (J. Am. Chem. Soc.2006, 128, 8751). This polymer shows a heat-induced oxygenation enhancement at low temperature region (5-20 °C), while showing a heat-induced oxygenation suppression at high temperature region (20-60 °C), resulting in an off-on-off activity profile against the temperature window. This is driven by a heat-induced phase transition of the polymer from coil to micelle and then to globule states. In the present work, effects of adding an amine component (N-[3-(dimethylamino)propyl]acrylamide: DMAPAM) to the polymer on the sensitization activity were studied, where the relationship between the phase transition behavior and the activity was clarified by several spectroscopic analyses. The polymers, poly(NIPAMx-co-BPy-co-DMAPAMz), show activity controlled by temperature and pH. The off-on-off activity profile shifts to higher temperature with a pH decrease. This is because protonation of the DMAPAM units leads to an increase in the polymer polarity and, hence, the polymer aggregates at higher temperature. In addition, increase in the DMAPAM content of the polymer leads to further shift of the activity profile. In contrast, at pH < 8, no activity enhancement is observed because complete protonation of the DMAPAM units suppresses polymer aggregation.  相似文献

A luminescent Eu, Dy: SrAl2O4 glass‐ceramics with high transparency in the visible region was successfully synthesized using the frozen sorbet technique with the control of O2 partial pressure () for the oxidation of Eu2+ ions. The glass‐ceramics include Eu2+, Eu3+, and Dy3+ ions, and thus exhibits three characteristic types of emission bands, 4f–5d at around 520 nm (Eu2+ ions), 4f–4f at 610 nm (Eu3+ ions), and 480 nm (Dy3+ ions). The Eu, Dy: SrAl2O4 glass‐ceramics provide remarkable long‐persistent luminescence under dark condition. The glass‐ceramics also exhibits color‐changing luminescence in the visible region based on their remarkable light storage properties. The luminescent Eu, Dy: SrAl2O4 glass‐ceramics using the frozen sorbet technique with control of are promising materials for application in novel photonic and light storage materials.  相似文献

Active oxidation behavior of CVD-SiC in CO─CO2 atmospheres was investigated using a thermogravimetric technique in the temperature range between 1823 and 1923 K. The gas pressure ratio, P CO2/ P CO, was controlled between 10−4 and 10−1 at 0.1 MPa. Active oxidation rates (mass loss rates) showed maxima at a certain value of P CO2/ P CO, ( P CO2/ P CO )*, In a P CO2/ P CO region lower than the ( P CO2/ P CO)* a carbon layer was formed on the SiC surface. In a P CO2/ P CO region higher than the ( P CO2/ P CO)*, silica particles or a porous silica layer was observed on the SiC surface.  相似文献

The aluminum aryloxide was prepared via the reaction of phenolic antioxidant, 3‐(3,5‐di‐t‐butyl‐4‐hydroxyphenyl)‐N‐octadecylpropionamide, with triethyl aluminum. Propylene polymerization using supported Ziegler–Natta catalyst systems was carried out in the presence of the antioxidant or its aluminum aryloxide. Although the antioxidant gave rise to decrease in catalyst yield and change in hydrogen response, the aluminum aryloxide had no influence on the catalytic polymerization behavior, and thus the obtained polymer characteristics such as molecular weight, polydispersity, and meso pentad as a stereoregularity were comparable to that polymerized without the antioxidant and the aluminum aryloxide. Polypropylene obtained in the presence of the aluminum aryloxide was well stabilized for oxidation and its stability was over 1000 h at 100°C (estimated to be over 30 years at room temperature). © 2005 Wiley Periodicals, Inc.
