Predicting the three-dimensional structure of proteins from amino acid sequences with only a few remote homologs,or de novo prediction,remains a major challenge in computational biology.The modeling of the protein backbone represents the initial phase of a protein structure prediction process.Using a parallel ant colony optimization based on sharing one pheromone matrix,this report proposes a parallel approach to predict the structure of a protein backbone.The parallel approach combines various sources of energy functions and generates protein backbones with the lowest energies jointly determined by the various energy functions.All free modeling targets in CASP8/9 are used to evaluate the performance of the method.For 13 targets in CASP8,two out of the predicted model1s selected by our approach are the best of the published CASP8 results,and seven out of the model1s are ranked in the top 10.For 29 targets in CASP9,20 out of the best models from our predictions are ranked in the top 10,and 11 out of the model1s are ranked in the top 10. 相似文献

For the existing problems of walking chair robot such as simple function,lower bearing capacity and not walking in complex environment,a novel varistructured quadruped / biped human-carrying walking chair robot is proposed.The proposed robot could be used as biped and quadruped walking chair robots.Considering the conversion of the walking chair robot from the quadruped to the biped or vice versa,6-UPS and 2-UPS+UP(U,P and S are universal joint,the prismatic pair,and sphere joint,respectively) parallel mechanisms are selected as the leg mechanism of the biped walking robot and quadruped walking robot,respectively.Combining the screw theory and theory of mechanism,the degrees of freedom of the leg mechanism and the body mechanism in diferent motion states are computed so as to meet the requirements of mechanism design.The motion characteristics of the 2-UPS+UP parallel mechanism which is the key part of the walking chair robot are analyzed.Then,the workspace of the moving platform is drawn and the efect of the structural parameters on the workspace volume is studied.Finally,it is found that the volume of the workspace of the moving platform is bigger when the side length ratio and the vertex angle ratio of the fxed platform and the moving platform which are isosceles triangles are close to 1.This study provides a theoretical foundation for the prototype development. 相似文献

Network user equilibrium or user optimum is an ideal state that can hardly be achieved in real traffic. More often than not, every day traffic tends to be in disequilibrium rather than equilibrium, thanks to uncertainties in demand and supply of the network. In this paper we propose a hybrid route choice model for studying non-equilibrium traffic. It combines pre-trip route choice and en-route route choice to solve dynamic traffic assignment (DTA) in large-scale networks. Travelers are divided into two groups, habitual travelers and adaptive travelers. Habitual travelers strictly follow their pre-trip routes which can be generated in the way that major links, such as freeways or major arterial streets, are favored over minor links, while taking into account historical traffic information. Adaptive travelers are responsive to real-time information and willing to explore new routes from time to time. We apply the hybrid route choice model in a synthetic medium-scale network and a large-scale real network to assess its effect on the flow patterns and network performances, and compare them with those obtained from Predictive User Equilibrium (PUE) DTA. The results show that PUE-DTA usually produces considerably less congestion and less frequent queue spillback than the hybrid route choice model. The ratio between habitual and adaptive travelers is crucial in determining realistic flow and queuing patterns. Consistent with previous studies, we found that, in non-PUE DTA, supplying a medium sized group (usually less than 50%) of travelers real-time information is more beneficial to network performance than supplying the majority of travelers with real-time information. Finally, some suggestions are given on how to calibrate the hybrid route choice model in practice to produce realistic results. 相似文献

A command filtered back-stepping attitude controller is exploited and analyzed to design a dynamic state-feedback controller for a generic Reentry Vehicle. A novel back-stepping control that obviates the need to compute analytic derivatives in the traditional back-stepping design is presented by combining command filtered back-stepping method, sliding-mode-based integral filters and inputto-state stability (ISS) analysis. The ISS-modular approach provides a simple and effective way for controlling non-linear Reentry Vehicle satisfying the strict-feedback form, simultaneously solving the problem of “explosion of complexity” in traditional back-stepping approach. The stability analysis of the closed-loop system and the convergence of the aerodynamic angles are verified based on the smallgain theorem. A nanofluidic preconcentrator with the capability of rapidly preconcentrating and precisely positioning protein bands in multiple microchannels has been developed for highly sensitive detection of biomolecules. A novel electrical resistive network model is developed to guide the design of the nanofluidic preconcentrator which consists of a PDMS slab bonded with a glass slide. In the prototype design, two microchannels (23 mm long, 25–50 μm wide, and 5–15 μm deep), one preconcentration microchannel and one ground microchannel are connected in the middle via 16 nanochannels (25–50 μm long, 25 μm wide, and 50–80 nm deep). With two sets of optimal voltage settings applied on the opposite ends of the nanofluidic chip, the ion depletion region and electrokinetic trapping were generated to carry out the preconcentration. With the optimal voltage settings (30–30 V) predicted by the model, the ionic current of the nanochannel in our optimized preconcentrator was adjusted to be greater than the threshold value (3.9 nA) needed for the occurrence of the preconcentration, and a preconcentration factor >105 was achieved in 5 min. The sample positioning capability of the preconcentrator was demonstrated by adjusting the applied voltages and moving the preconcentrated protein bands to multiple sites by a distance from several micrometers to several millimeters in the preconcentration channel. The multi-channel preconcentration capability was also demonstrated by preconcentrating two protein bands in two separate microchannels. In this work, the resistive network model was developed and validated to optimize nanofluidic preconcentrators for rapid, high throughput and highly sensitive sensing of low abundance analytes. 相似文献

A cylindrical gold-coated stainless steel rod was positioned at the center of a straight microchannel connecting two fluid reservoirs on either end. The microchannel was filled with 1 mM KCl containing 0.5 μm diameter carboxylate-modified spherical particles. Induced-charge electro-osmotic (ICEO) flow occurred around the metallic rod under a sinusoidal AC electric field applied using two platinum electrodes. The ICEO flows around the metallic rod were measured using micro particle image velocimetry (micro-PIV) technique as functions of the AC electric field strength and frequency. The present study provides experimental data about ICEO flow in the weakly nonlinear limit of thin double layers, in which, the charging dynamics of the double layer cannot be presented analytically. The measured ICEO flow pattern qualitatively agrees with the theoretical results obtained by Squires and Bazant (J Fluid Mech 509:217–252, 2004). Flow around the rod is quadrupolar, driving liquid towards the rod along the electric field and forcing it away from the rod in the direction perpendicular to the imposed electric field. The measured ICEO flow velocity is proportional to the square of the electric field strength, and depends on the applied AC frequency. 相似文献

Spatiotemporal deformations of the free charged surface of a thin electrolyte film undergoing a coupled electrokinetic flow composed of an electroosmotic flow (EOF) on a charged solid substrate and an electrophoretic flow (EPF) at its free surface are explored through linear stability analysis and the long-wave nonlinear simulations. The nonlinear evolution equation for the deforming surface is derived by considering both the Maxwell’s stresses and the hydrodynamic stresses. The electric potential across the film is obtained from the Poisson–Boltzmann equation under the Debye–Hückel approximation. The simulations show that at the charged electrolyte–air interface, the applied electric field generates an EPF similar to that of a large charged particle. The EOF near the solid–electrolyte interface and the EPF at the electrolyte–air interface are in the same (or opposite) directions when the zeta potentials at the two interfaces are of the opposite (or same) signs. The linear and nonlinear analyses of the evolution equation predict the presence of travelling waves, which is strongly modulated by the applied electric field and the magnitude/sign of the interface zeta potentials. The time and length scales of the unstable modes reduce as the sign of zeta potential at the two interfaces is varied from being opposite to same and also with the increasing applied electric field. The increased destabilization is caused by a reverse EPF near the free surface when the interfaces bear the same sign of zeta potentials. Flow reversal by EPF at the free surface occurs at smaller zeta potential of the free surface when the film is thicker because of less influence of the EOF arising at the solid–electrolyte boundary. The amplitude of the surface waves is found to be smaller when the unstable waves travel at a faster speed. The films can undergo pseudo-dewetting when the free surface is almost stationary under the combined influences of EPF and EOF. The free surface instability of the coupled EOF and EPF has some interesting implications in the development of micro/nano fluidic devices involving a free surface. 相似文献
