Two detoxification mechanisms working in the marine diatom Thalassiosira weissflogii to cope with mercury toxicity were investigated. Initially, the effect of mercury on the intracellular pool of non-protein thiols was studied in exponentially growing cultures exposed to sub-toxic HgCl2 concentrations. T. weissflogii cells responded by synthesizing metal-binding peptides, named phytochelatins (PCs), besides increasing the intracellular pool of glutathione and γ-glutamylcysteine (γ-EC). Intracellular Hg and PC concentrations increased with the Hg concentration in the culture medium, exhibiting a distinct dose-response relationship. However, considerations of the PCs-SH:Hg molar ratio suggest that glutathione could also be involved in the intracellular mercury sequestration. The time course of the non-protein thiol pool and Hg intracellular concentration shows that PCs, glutathione and γ-EC represent a rapid cellular response to mercury, although their role in Hg detoxification seems to lose importance at longer incubation times. The occurrence of a process of reduction of Hg(II) to Hg° and subsequent production of dissolved gaseous mercury (DGM) was also investigated at lower Hg concentrations, at which the PC synthesis doesn't seem to be involved. The significant (P < 0.01) correlation between the cellular density in solution and the production of DGM suggests that this diatom is capable of directly producing DGM, both in light and dark conditions. This finding has been confirmed by the absence of DGM production in the culture media containing formaldehyde-killed cells. The 70% ethanolic extracts from eight neglected fruits; Muntingia calabura, Leucaena leucocephala, Spondias dulcis, Syzygium jambos, Mangifera caesia, Ardisia elliptica, Cynometra cauliflora and Ficus auriculata were evaluated for their 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, α-glucosidase inhibitory activities as well as total phenolic content. The results of this study revealed that M. caesia fruit extract demonstrated the most potent radical scavenging activity. Among the fruits examined for α-glucosidase inhibitory activity, M. calabura and F. auriculata exhibited strong activity with no significant difference. The Pearson correlation indicated that the activities of M. caesia and F. auriculata contributed by phenolic compounds. A total of 65 metabolites were tentatively identified by using ultra-high-performance liquid chromatography tandem mass spectrometry (UHLPC-MS/MS). Transmission of pathogenic microorganisms on textile in the hospital settings may contribute to nosocomial infection. This study was aimed to develop a natural antimicrobial finishing on textile that can prevent transmission of pathogenic microorganisms that meets American Association of Textiles Chemists and Colorists (AATCC) standard. Punica granatum L ethanolic extract was shown to have broad spectrum antimicrobial efficacy with bis(6-methylheptyl)-benzene-1,2-dicarboxylate and ethyl-pentadecanoate as the main bioactive constituents. Textile samples developed with the extract finishing exhibited excellent antimicrobial efficacy with a growth reduction of 99.9% on Hoheinstein Challenge Test. A review on phthalate esters or phthalic acid esters (PAEs), chemicals of concern since a few decades ago that are widely used as plasticizers in food processing and packaging, is presented taking into account the background of such compounds, the metabolism, human exposure to PAEs, the sources and occurrence in food as well as the toxicological aspects and human health effects. In addition, 45 novel research articles that were published between 2002 and 2017 were identified and their results were tabulated showing the PAEs analysed, food matrix of PAEs, methods of sample preparation/extraction, methods of instrumental analysis and quantitation, percentage recovery and limit of detection (LOD) of the instrument for ease of comparison and referencing. In general, it was found that in the last 15 years, the number of PAEs analysed has increased from the commonly analysed 8 PAEs, namely dimethyl phthalate (DMP), diethyl phthalate (DEP), diisobutyl phthalate (DIBP), di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), dicyclohexyl phthalate (DCHP), di-n-octyl phthalate (DNOP) and di-(2-ethylhexyl) phthalate (DEHP) to as many as 23 PAEs. The methods of sample preparation have also progressed from the simple liquid-liquid extraction using organic solvents to solid-phase microextraction techniques to the more recent head-space or direct immersion solid-phase microextraction methods. Whereas for the analysis of PAEs, gas chromatography and liquid chromatography are still the preferred methods with improved LOD of analysis ranging from approximately 10 ppm for fatty foods to 1–60 ppt for water, juices and cooking oil samples. 相似文献

Antimicrobial behavior of lactic acid bacteria (LAB) has been explored since many years to assess their ability to produce bacteriocin, a natural preservative, to increase the shelf life of food. This study aims to characterize bacteriocin producing strains of lactic acid bacteria isolated from acidic to slightly acidic raw vegetables including tomato, bell pepper and green chili and to investigate their potential to inhibit food related bacteria. Among twenty nine LAB screened for antimicrobial activity, three exhibited antagonism against closely related bacterial isolates which was influenced by varying temperature and pH. They were identified up to strain level as Lactococcus lactis subsp. lactis TI-4, L. lactis subsp. lactis CE-2 and L. lactis subsp. lactis PI-2 based on 16S rRNA gene sequence. Their spectrum of inhibition was observed against food associated strains of Bacillus subtilis and Staphylococcus aureus. Moreover, L. lactis subsp. lactis PI-2 selected on the basis of higher antimicrobial activity was further evaluated for bacteriocin production which was detected as nisin A and nisin Z. These findings suggest the possible use of L. lactis strains of vegetable origin as protective cultures in slightly acidic as well as slightly alkaline food by the bio-preservative action of bacteriocins. 相似文献

Resistant starch (RS) can be generated through heat moisture treatment (HMT). The HMT was conducted by modifying starch using different ratio of moisture content, high temperature and heating time. A number of studies showed that the effects of HMT on RS contents in cereals, pulses, tubers and fruits were inconsistent. This study aimed to analyse the impact of HMT on RS level in various carbohydrate sources through a meta-analysis approach. Study selection was conducted with the PRISMA method. There were 21 relevant studies and 67 data used for meta-analysis. The database was analysed by using Hedges’ d. The results showed that there was a significant impact of HMT on RS level of cereals, especially wheat. The highest increase in RS levels for various carbohydrate sources in starch was influenced by the interaction of treatment between water content at 15 ≤ x < 25%, heating time at 0.25 < x ≤ 6 h and temperature at 120 ≤ x ≤ 130 °C. 相似文献

The global energy consumption is likely to grow faster than the population growth. The fuel consumption was growing from 6630 million tons of oil equivalents (Mtoe) in 1980 to 11,163 Mtoe in 2009. This projected consumption will increase 1.5% per year until 2030 and reach 16,900 Mtoe and the main drivers of this growth are mostly developing countries in Asia. Indonesia is one of the developing countries and energy supply is an important factor for all-around development. The country's energy consumption still depends on non-renewable energy such as crude oil, coal and natural gas as sources of energy. Utilization of fossil fuel continuously contributes to huge amount of greenhouse gases emission that leads to climate change. Facing such an unfavorable situation, the government of Indonesia prioritizes on energy supply securities by diversification of energy resources. The energy mixes in Indonesia based on five main resources; these are crude oil, natural gas, coal, hydropower, and renewable energy. Although the country encourages utilizing renewable energy, the contribution is only around 3%. Considering natural condition and geography, this country is blessed with great potential of renewable energy such as solar energy, wind energy, micro hydro and biomass energy. Noting the potential of renewable and sustainable energy resources in the country, the government must pay more attention on how to utilize it. Many efforts have been done to promote renewable energy such as to create energy policy and regulations, yet it still did not give any satisfactory result. Government, non-government agencies and the public should take a more proactive step to promote and use renewable energy in order to achieve the secure and environmentally sustainable energy resources. 相似文献
