Dissociation between the development of myocardial contracture and heart damage in the "calcium paradox"

The interrelationship between contracture development and heart damage during the calcium paradox under different sodium concentration in Ca-free media was studied on isolated rat hearts. It had been shown that calcium paradox development accompanied contracture development, intensive membrane disruption and alteration of tissue energy state. We had not found relation between contracture magnitude and degree of myocardial alterations in calcium paradox. Our dates confirm so-called intracellular hypothesis of calcium paradox. The experiments had shown close correlation between transmembrane sodium gradient in Ca-free media and degree of cellular damage and energy state alterations during calcium readmission in solution.

On the limited value of fine-needle aspiration for the diagnosis of benign melanocytic proliferations of the skin

Ligation of the alpha2-macroglobulin signalling receptor (alpha2MSR) with alpha2-macroglobulin (alpha2M)-methylamine or a cloned and expressed receptor binding fragment (RBF) stimulates DNA synthesis. To examine the possible role of the Ras pathway in the mitogenic effects observed on ligating alpha2MSR, we studied the formation of p2 Ras-GTP in murine peritoneal macrophages upon treatment with alpha2M-methylamine and RBF, respectively. Both alpha2M-methylamine (50 pM) and RBF (50 pM) stimulated a 2-3-fold increase in the formation of the p21Ras-GTP complex compared with unstimulated cells. p21Ras-GT32P complex formation was both time and RBF concentration dependent and was comparable to p21Ras-GT32P complex formation induced by EGF (200 ng/mL) and platelet derived growth factor (50 mg/mL). Up-regulation of cells with phorbol dibutyrate prior to stimulation with RBF had no effect on p2 Ras-GT32P formation. However, treatment of macrophages with the tyrosine kinase inhibitor genestein drastically reduced RBF-induced formation of the p21 Ras-GT32P complex. Wortmannin, an inhibitor of phosphatidylinositol-3'-kinase (PI3K), had no effect on p21Ras-GT32P complex formation. It is concluded that the mitogenic effects of ligating alpha2MSR are mediated through a Ras-dependent pathway.     

Fluorogenic substrates for proteases based on intramolecular fluorescence energy transfer (IFETS)

A prospective class of intramolecular fluorescence energy transfer substrates (IFETS) is described. In contrast to the known chromogenic and fluorogenic substrates that are used widely in the scientific and medical research, IFETS allow to control the enzymatic cleavage at any point of the peptide chain and thus permit simultaneous studies of enzymes of different specificity. We discuss the main types of donor and acceptor, their advantages and drawbacks and the effectiveness of exited-state energy transfer between them. High sensitivity and selectivity of IFETS in the assay of proteinases was demonstrated. They prove to be a very useful and very promising instrument for enzymology and medicine.     

Microstructure and Superconducting Properties of TaC-Doped Bi-2223 Ceramics

The microstructure, phase composition, and superconducting properties of Bi-2223 ceramics doped with 0.05–0.5 wt % TaC were investigated. The materials were heat-treated at 840°C in three steps (10 + 24 + 24 h). By optimizing the TaC content and heat-treatment conditions, the 77-K critical current density of the Bi-2223 ceramics in zero field was raised by a factor of 2.

Experience in heptral treatment of diffuse liver diseases

AIM: Heptral trial in alcoholic, drug and viral diseases of the liver. MATERIALS AND METHODS: 67 patients with chronic diffuse liver diseases were treated with heptral. The examination covered AlAt, AP, HHTP, bilirubin, cholesterol, CT of the liver, esophagogastroduodenoscopy. Heptral was given by two steps: 14 days of intravenous drops (800 mg/day) followed by 14 days of oral use (2 tablets, 400 mg each) before meal at 8 a.m. and 2 p.m. RESULTS: In alcoholic disease of the liver, heptral relieved depression, reduced AlAT, AP, HHTP, bilirubin, density of the liver. The addition of heptral to interferon-alpha-2 treatment of chronic hepatitis C corrected intrahepatic cholestasis. In drug-induced liver damage heptral promoted normalization of the hepatic tests, improved general condition of the patients. CONCLUSION: Heptral is effective in patients with alcoholic and drug liver lesions, chronic hepatitis C.
