The zebrafish embryo is a vertebrate well suited for visualizing nanoparticles at high resolution in live animals. Its optical transparency and genetic versatility allow noninvasive, real‐time observations of vascular flow of nanoparticles and their interactions with cells throughout the body. As a consequence, this system enables the acquisition of quantitative data that are difficult to obtain in rodents. Until now, a few studies using the zebrafish model have only described semiquantitative results on key nanoparticle parameters. Here, a MACRO dedicated to automated quantitative methods is described for analyzing important parameters of nanoparticle behavior, such as circulation time and interactions with key target cells, macrophages, and endothelial cells. Direct comparison of four nanoparticle (NP) formulations in zebrafish embryos and mice reveals that data obtained in zebrafish can be used to predict NPs' behavior in the mouse model. NPs having long or short blood circulation in rodents behave similarly in the zebrafish embryo, with low circulation times being a consequence of NP uptake into macrophages or endothelial cells. It is proposed that the zebrafish embryo has the potential to become an important intermediate screening system for nanoparticle research to bridge the gap between cell culture studies and preclinical rodent models such as the mouse.  相似文献

The locally defined growth of carbon nanofibers with lambda shape in an open flame process is demonstrated. Via the growth time, the geometry of the structures can be tailored to a Λ‐ or λ‐type shape. Microchannel cantilever spotting and dip‐pen nanolithography are utilized for the deposition of catalytic salt NiCl2 · 6H2O for locally controlled growth of lambda‐shaped carbon nanofibers. Rigorous downscaling reveals a critical catalytic salt volume of 0.033 µm³, resulting in exactly one lambda‐shaped carbon nanofiber at a highly predefined position. An empirical model explains the observed growth process.  相似文献

Super‐resolution fluorescence microscopy allows for unprecedented in situ visualization of biological structures, but its application to materials science has so far been comparatively limited. One of the main reasons is the lack of powerful dyes that allow for labeling and photoswitching in materials science systems. In this study it is shown that appropriate substitution of diarylethenes bearing a fluorescent closed and dark open form paves the way for imaging nanostructured materials with three of the most popular super‐resolution fluorescence microscopy methods that are based on different concepts to achieve imaging beyond the diffraction limit of light. The key to obtain optimal resolution lies in a proper control over the photochemistry of the photoswitches and its adaption to the system to be imaged. It is hoped that the present work will provide researchers with a guide to choose the best photoswitch derivative for super‐resolution microscopy in materials science, just like the correct choice of a Swiss Army Knife's tool is essential to fulfill a given task.  相似文献

Targeted delivery of nanomedicine/nanoparticles (NM/NPs) to the site of disease (e.g., the tumor or lung injury) is of vital importance for improved therapeutic efficacy. Multimodal imaging platforms provide powerful tools for monitoring delivery and tissue distribution of drugs and NM/NPs. This study introduces a preclinical imaging platform combining X‐ray (two modes) and fluorescence imaging (three modes) techniques for time‐resolved in vivo and spatially resolved ex vivo visualization of mouse lungs during pulmonary NP delivery. Liquid mixtures of iodine (contrast agent for X‐ray) and/or (nano)particles (X‐ray absorbing and/or fluorescent) are delivered to different regions of the lung via intratracheal instillation, nasal aspiration, and ventilator‐assisted aerosol inhalation. It is demonstrated that in vivo propagation‐based phase‐contrast X‐ray imaging elucidates the dynamic process of pulmonary NP delivery, while ex vivo fluorescence imaging (e.g., tissue‐cleared light sheet fluorescence microscopy) reveals the quantitative 3D drug/particle distribution throughout the entire lung with cellular resolution. The novel and complementary information from this imaging platform unveils the dynamics and mechanisms of pulmonary NM/NP delivery and deposition for each of the delivery routes, which provides guidance on optimizing pulmonary delivery techniques and novel‐designed NM for targeting and efficacy.  相似文献

The European Macroseismic Scale 1998 – (EMS 98) is an instrument for the natural hazard of earthquakes, which allows the explanation of observed differences in the behaviour of the prevailing construction types by introducing and implementing vulnerability classes. The EMS 98 enables also the identification of purely empirically justified ranges of scatter. The basic methodology is also applicable to other natural hazards such as flood, tsunami and wind. This permits the development of a unified methodology for the consideration of building vulnerability in the sense of a multi‐hazard approach. For the first time, a vulnerability‐oriented instrument is available to evaluate a building stock for different natural hazards according to criteria that have been standardised in terms of engineering. The multi‐hazard vulnerability with its possible scatters is examined and visualized on real building inventories. With the concept of ”LEGOisation“ the existing buildings, a novel approach is presented to allow the sub‐structuring of buildings into storeys (including roof, basement and floors) to further development of the classification towards specific damage characteristics and local vulnerability. A still to be developed ”Conceptual Simulation Tool“ is described as an outlook. This uses the tools and methods developed to simulate damage and losses as a result of various natural hazards and their sequences.  相似文献

The importance of direct tensile tests on solid and capillary bonded particulate agglomerates is investigated and compared to compression test measurements. The properties of wet agglomerates are varied by changing the contact angle by means of functionalization of the particle surface. Process conditions are considered by variation of ambient humidity. A qualitative evaluation of the results is performed by analyzing the measured force distance curves of different tensile tests. The results are quantitatively evaluated by calculating the breakage strength, mass related breakage energy and breakage probability showing that the ratio between tensile and compressive tests is highly dependent on the adjusted parameters. Next to the process parameter effect, also the influence of agglomerate size is considered. Tensile strength data are used to estimate the single bond forces between the primary particles of the agglomerates. Tensile and compressive test results are compared to numerical results (DEM) of agglomerate breakage using an elastic stiff bond model.  相似文献

The visual and mechanical quality of paperboard packaging components formed by deep drawing closely corresponds to the wrinkle distribution within the material wall section. Assessing this quality of paperboard packaging components currently relies on very slow or even subjective methods. Therefore, counting the number of wrinkles and measuring the mean distance between wrinkles proved to be a sensitive strategy for the evaluation of quality levels in deep drawing formed paperboard containers like cups or trays. A method introduced by Hauptmann proposes the measurement of the distances between 10 wrinkles in machine direction and 10 wrinkles in cross direction for each sample. However, this method is unsatisfactory because it can only observe a small part of each sample and is subject to deviation through the individual judgement of the person carrying out the survey. Further methods, as well as their respective application range, are listed in Table  1 . Hence, it is desirable to automate the wrinkle survey to be carried out without operator influences to the measurement. This paper introduces such an automated measuring method. The given method relies on recording the sample surface topography through laser‐distance measurement. An evaluation algorithm, which enables the detection of wrinkles in the topographic data, is proposed. Furthermore, it is shown that the method yields feasible results and is capable of generating reliable quality information in considerably reduced time frames.  相似文献

Nitride and oxy‐nitride HPPMS coatings for the application in the plastics processing (Part 2) In the previous issue three oxy‐nitridic hard coatings on CrAl‐basis were investigated. These coatings were deposited by physical vapour deposition (PVD) as protective coatings against adhesive and abrasive wear in polymer extrusion. The coatings were developed using a variation of the oxygen content to investigate the influence of the chemical composition on the coating properties as well as composite properties between the coating and the coated tool. Following up on these findings this article will focus on the application oriented system properties of the three investigated coating systems towards the polycarbonate melt.  相似文献
