We have built a fiber-optic confocal reflectance microscope capable of imaging human tissues in near real time. Miniaturization of the objective lens and the mechanical components for positioning and axially scanning the objective enables the device to be used in inner organs of the human body. The lateral resolution is 2 micrometers and axial resolution is 10 micrometers. Confocal images of fixed tissue biopsies and the human lip in vivo have been obtained at 15 frames/s without any fluorescent stains. With longitudinal diffusion as an unavoidable source of peak broadening, the peak efficiency (expressed by the plate number, N) in capillary zone electrophoresis depends on the ratio of electrophoretic mobility, mu, and tracer- or self-diffusion coefficient, D. Both parameters are functions of the ionic strength of the electrolyte solution. According to theory, the mobility is decreased with increasing ionic strength by the relaxation effect (depending on the relative permittivity) and the electrophoretic effect (depending on the relative permittivity and the viscosity of the solvent), whereas the diffusion coefficient is decreased only by the relaxation effect. This allows the theoretical predictions that the plate number, which is proportional to the ratio mu/D, decreases with increasing ionic strength and that the magnitude of this reduction depends on the solvent. Taking the values for relative permittivity and viscosity allows forecasting that, in general, water as a solvent exhibits the smallest lowering of the plate number, as compared to organic solvents. The theoretical predictions are confirmed by the data for the ratio calculated from measured mobilities and diffusion coefficients for iodide as the analyte ion in water, methanol, and acetonitrile with ionic strength of the background electrolyte varying between 0.005 and 0.080 mol L(-1). Whereas the experimentally observed plate number per volt is reduced from its "ultimate value" of about 20 (analyte charge number z = 1, zero ionic strength) in water by only 10%, the decrease at the same ionic strength in methanol and acetonitrile reaches 25 to 30%. Thus, the maximum plate number should read Nmax approximately equals 13 zU (with U being the effective voltage) for these solvents with ionic strengths normally applied in capillary electrophoresis. The report of work-related musculoskeletal pain may be related to worker evaluations of the design and management of work through two mechanisms: one biomechanical and the other stress-related. This study of engineering workers (n = 204) explored the validity of these mechanisms using sequential logistic regression. Analyses suggested that workers' ratings of the adequacy of the design and management of their work were related to their report of work-related musculoskeletal pain. However, the mechanisms appeared to be activated in certain conditions. The reporting of pain in the upper body was both biomechanically- and stress-related, whereas that in the lower body was only biomechanically-related. It is argued that the mechanism activated appeared to be determined by the anatomical location of the pain, and probably the variance shared between the different aspects of work design and management, on the one hand, and the mechanical load of the job, on the other. 相似文献

Negative emotion has been shown to reduce flexibility in cognition and behavior. We examined interpersonal flexibility during negative emotional episodes within parent-child interactions. Fifty-five mothers and early-adolescent daughters were observed during a positive discussion, a negative (conflict) discussion, and another positive discussion. Codes of moment-to-moment changes in emotion expression were used to create state space grids from which measures of emotional valence and flexibility were derived. As expected, mean flexibility was lowest during the conflict discussion when negative emotion peaked, suggesting that interpersonal flexibility decreases with increasing negative emotion. Sub-groups identified as low or high in stress were also compared. Dyads with girls reporting more stressful events showed lower flexibility during the first positive discussion. However, dyads expressing more negative emotion during the conflict discussion were also more flexible, suggesting that flexible dyadic styles permit more negative emotion.
