There is growing evidence that a large proportion of upper digestive tract tumors are ascribable to heavy alcohol drinking and tobacco consumption. The cancer-promoting action of ethanol is mediated by acetaldehyde, its first metabolite, also derived from the bacterial oxidation of alcohol by the oral microflora, classified by the International Agency for the Research on Cancer as a carcinogen. Acetaldehyde is also one of the major components of tobacco smoke. These findings suggest two different strategies to decrease the risk of alcohol-related oral cancers: the reduction of the levels of alcohol-derived acetaldehyde in saliva and the reduction of oral bacterial flora. Therefore, the aim of our study was to develop and characterize some buccal tablet formulations containing both 20 mg L-cysteine hydrochloride (able to chemically neutralize acetaldehyde) and 10 mg chlorhexidine diacetate (well-known antiseptic compound active against a large spectrum of oral microbes). One of these formulations, chosen on the basis of its favourable delivery kinetics of the active principles, was demonstrated to be able to reduce acetaldehyde concentration in an in vitro system and to lower its salivary levels in volunteers after ethanol contact. Our findings support the hypothesis that the application of buccal devices containing cysteine and chlorhexidine could reduce salivary acetaldehyde levels and thus the incidence of upper gastrointestinal cancer in drinkers and smokers.  相似文献

Consumers are oriented towards fresh-cut vegetables that provide phytonutrients useful for preventing stress-related diseases. The aim of this work was to evaluate the cut operations on the quality changes of lamb's lettuce ( Valerianella olitoria L.) cv. Trofy during storage at 4 °C for 8 days. Results showed that chlorophyll and carotenoids reduction was observed after 8 days of storage. In both treatments, total carotenoids after 8 days decreased from 20 to 16 mg g−1 FW. Free and total phenols increased with storage in both treatments. Total phenols were 23% higher in control (32 μmol g−1 FW) compared to cut leaves (25 μmol g−1 FW) after 8 days of storage. Anthocyanins increased after 8 days and reached 30 mg 100 g−1 FW without significant difference between treatments. Ascorbic acid (AsA) and dehydroascorbic (DHA) acid increased in cut leaves compared to control. After 1 day AsA concentration was 3 300 nmol g−1 FW in cut leaves, while in control leaves was 1 500 nmol g−1 FW. Analogously AsA + DHA was higher in cut leaves, 4 100 nmol g−1 FW, while in control leaves the mean was 3 000 nmol g−1 FW. After 5 days of storage the values of AsA returned to initial values, while AsA + DHA were lower.  相似文献

The preparation, characterization, and stability of lyophilized liposome-based formulation of mitoxantrone was investigated. Mitoxantrone was entrapped inside small, unilamellar liposomes composed of dioleoylphosphocholine (DOPC), cholesterol, and cardiolipin. The mean vesicle size and drug entrapment efficiency of the liposomes were ~ 150 nm and ~ 99%, respectively. Less than 1% of drug was lost and mean vesicle size remained unchanged after sterile filtration. The pre-lyophilized (pre-lyo) formulations were characterized by a differential scanning calorimetric (DSC) method. Results showed that the glass transition temperatures (Tg') increased as the molar ratios of sucrose:lipid and trehalose:lipid in the formulations were increased. The maximum Tg' of the pre-lyo formulations containing 10:1 sucrose:lipid and trehalose:lipid molar ratios were - 37°C and - 41°C, respectively. After reconstitution of the lyophilized cake of the sucrose-containing formulation, the mean vesicle size was comparable to pre-lyo liposome size. In vitro release studies showed that less than 2% of mitoxantrone was released after an extensive dialysis against phosphate buffered saline (PBS) at 37°C, indicating that the mitoxantrone was highly associated and retained inside the liposomes. Short-term stability studies of the sucrose-containing formulations revealed that the reconstituted and eight-fold diluted formulations were stable for up to 8 hours at room temperature. Long-term stability studies of lyophilized liposomal mitoxantrone showed that the lyophilized formulation was stable for up to 13 months after storage at refrigerated condition.  相似文献

The effects of adding mixtures of herbs such as garlic, laurel, and marjoram on selected chemical indices of olive oil from canned dried tomatoes were studied for various storage periods of up to 1 yr. Conventional analytical indices measured included acidity, PV, and p-anisidine value. Flavored samples showed kinetic constant values that were significantly (P=0.001) higher than unflavored ones, whereas oligopolymer and oxidized TG and DG contents were similar to or slightly higher than in the unflavored samples compared with those with herbs. The addition of the mixture of herbs slowed polymerization reactions but did not inhibit TG oxidation. Discrepancies between the results obtained by conventional analyses and high-performance size-exclusion chromatography indicated that the former approaches were insufficient to determine oxidative degradation of oil as a result of interferences from compounds in the food matrix.  相似文献

The importance of the non-singular terms of the series representation for the stresses in the crack tip region has been underlined in recent papers. In the present work it is recognized that the stress-intensity factors KI and KII are no longer sufficient to define fracture crisis for the plane problem. Some fracture criteria are applied introducing the third parameter λ, which is connected with the stress collinear to the crack's line. The respective fracture loci in the KI−KII−λ space are discussed; they depend on the distance from the crack tip at which the crisis conditions are checked. Finally an application of the above mentioned loci to the uniaxial loading condition is shown.
Résumé Le problème de la mécanique de la rupture a été étudie jusqu'ici en considérant, dans la solution élastique, valable autour des extrémités de fissure, le seul premier terme du développement en autofonctions de Williams. Ce terme de la série qui représente le champ de contrainte est inversement proportionnel à la racine carrée de la distance radiale de l'extrémité de l'axe de fissuration. Cependant, dans cette étude, on considère aussi le deuxième terme de cette série, indépendant de la distance radiale. L'omission du deuxième terme, comme on le souligne dans des travaux assez récents, équivaut à négliger l'effet des tensions normales colinéaires à l'axe de fissuration. Si l'on considère aussi le terme non singulier, on admet que la rupture dépend d'un paramètre ultérieur lié à la contriante normale colinéaire à l'axe de fissuration. Par conséquent, les facteurs d'intensité de contrainte KI et KII, introduits par Irwin, ne suffisent plus à décrire la ramification de la fissure. Les endroits de rupture, comme on le montre dans cette étude, sont représentés par des surfaces dans l'espace tridimensionnel KI−KII−λ, et non plus par des courbes dans le plan KI−KII. Conformément à ces hypothèses, les surfaces varient en fonction de la distance radiale de l'extrémité de la fissure d'après laquelle on estime les conditions de crise (déterminant la rupture). Cette distance est caractéristique du matériau considéré, puisqu'elle est liée à la dimension de la zone plastique à l'extrémité de la fissure. Dans la partie finale de l'étude, on montre ensuite une application dans un cas de sollicitation sous charge monoaxiale en indiquant les résultats expérimentaux obtenus sur des plaques de plexiglass entaillées.

In this paper we report the fabrication of thiolene‐based microstructured reactors (MRs) that have been specifically designed to include solid‐supported reagents within the microchannels network. We propose a convenient solution to realize reversible press‐fit, leak‐proof interconnects that greatly simplify the MR coupling to the external environment such as capillary tubing, sample reservoirs and pumps. The MRs have been used to carry out the oxidation of α‐terpinene and methionine using [60]fullerene, covalently linked to Tentagel® and silica gel matrices, as a singlet oxygen sensitizer. High conversions have been observed for both substrates although, in the case of α‐terpinene, a partial photodegradation of the endo‐peroxide product was detected. Interestingly, in the case of methionine, a quantitative conversion to the corresponding sulfoxides was achieved in about 40 seconds, using low‐power, white LED illumination. The reaction time is considerably shorter when compared to the batch procedure that requires, for the same process, about one hour illumination and the use of a 300‐W tungsten halogen lamp.  相似文献

An investigation was carried out to evaluate the use of High Performance Size-Exclusion Chromatography (HPSEC) of polar compounds of refined, mild deodorized, extra virgin olive oils as well as of their blends, in attempting to reveal significant differences in the amounts of the substance classes constituting polar compounds among these oils. Two sets of blends were prepared by mixing an extra virgin olive oil with both refined and mild deodorized olive oils in increasing amounts. The obtained data highlighted that the triacylglycerol oligopolymers were absent or present in traces in the extra virgin olive oil, while their mean amount was equal to 0.04 g/100 g and 0.72 g/100 g in mild deodorized and refined olive oils, respectively. Oxidized triacylglycerols and diacylglycerols were more abundant in mild deodorized oil and refined oil than in extra virgin olive oil. The Factorial Discriminant Analysis of the data showed that the HPSEC analysis could reveal the presence of refined/mild deodorized oils in extra virgin olive oils. In particular, the classification functions obtained allowed designation of mixtures containing at least 30 g/100 g of mild deodorized oil and all those containing refined olive oil as deodorized oil, therefore as oils subjected to at least a mild refining treatment.  相似文献
