We have determined the time course, the spatial spread in brain tissue, and the intracellular distribution of biotin- and fluorescein-labeled phosphorothioate oligodeoxynucleotides (ODNs) following single injections into the rat striatum or the lateral ventricle. These time and space parameters were correlated with the ability of c-fos phosphorothioate antisense ODNs to suppress the induction of Fos protein by cocaine. A rapid and dose-dependent tissue penetration of labeled ODNs was observed following either intrastriatal or intraventricular injections of a constant sample volume. Inspection of tissue sections by confocal microscopy uncovered a distinct change in the intracellular disposition of labeled ODNs during the 24 h post-injection period. At 1, 6 and 12 h, the vast majority of the fluorescent signal was confined to the interstitial spaces throughout the zone penetrated by ODNs. Neuronal nuclei displayed faint labeling along the outer portion of the nucleus at 1 and 6 h post-injection. At these time-points, ODNs were not detected in the cytoplasm. By 16 h, ODNs were barely detectable in the extracellular space and absent from neuronal nuclei. Instead, ODNs were seen in large cytoplasmic granules of neurons throughout the tissue zone penetrated by the ODNs. Experiments with intrastriatal injections of antisense ODNs to c-fos mRNA revealed Fos suppression between 3 and 12 h, but not at 16 and 24 h. This combined analysis has revealed that (1) restricted tissue penetration by ODNs limits their antisense effects on protein expression, and (2) depletion of extracellular ODNs and sequestration of c-fos antisense ODNs into large intracellular granules coincides with the loss of their biological activity. 相似文献

In the growth stage of a product life cycle, the demand rate is usually unstable and follows an increasing pattern. The traditional inventory policies, which have been developed for stationary demand pattern, are not appropriate to this situation. Although there exist some researches in the past dealing with inventory policy for the case of increasing demand pattern, most of them focused on the inventory systems in which shortages are not allowed. In reality, the presence of shortages is sometimes economically preferable when holding cost is significant as compared with shortage cost. The aim of the research presented in this paper is, therefore, to develop a replenishment policy for inventory systems with nonlinear increasing demand pattern and shortage allowance in such a way that the total demand during a predefined planning horizon can be exactly met. A heuristic technique to help determine the operational parameters for the inventory policy is then developed. In the proposed heuristic technique, the consecutive improvement method developed by Wang (Comput Oper Res, 29:1819–1825, 2002) will first be used to help determine replenishment times. And then, a new concept of reduction cost, which is defined as the difference between the holding cost when shortage is allowed and the incurred shortage cost, is introduced and applied to help find the optimal shortage starting point in each replenishment cycle. The process of hot water extraction of tar sand was modified and adapted for removal of heavy oil from bottom tank petroleum sludges, and was submitted to a laboratory feasibility study. This process can also be utilized to clean beach sands contaminated by accidental heavy oil spills. In the case of oil contaminated sands, a single stage extraction has yielded a 99% recovery of hydrocarbons and clean sands (containing less than 0.1% of hydrocarbons) which are thus safe to be returned to the environment. In the case of petroleum bottom tank sludge, it was necessary to proceed with a double stage extraction with the addition of a wetting agent. A dosage of a Na2 Si O3 aqueous solution of 1% by weight has proven efficient, allowing an 82% recovery of hydrocarbons, with only 0.5% of hydrocarbons content in the solid residues. 相似文献

In the present work, we report first results about a technology using a conjugated copolymer poly(5-hydroxy-1,4-naphthoquinone-co-5-hydroxy-2-carboxyethyl-1,4-naphthoquinone) acting both as immobilizing and transducing element for reagentless immunosensor, and its application for the detection of HPV infection. It was shown that the reagentless immunosensor was able to detect the interaction between antigenic peptide L1 from HPV-16 major capsid protein, a dominant epitope involved in viral infection as well as in prophylactic vaccine, and the relevant antibody. 相似文献

Increased discharge capacity of a Li-air activated carbon cathode produced by preventing carbon surface passivation 总被引:1,自引:0,他引:1
A significant discharge capacity increase (larger than 3 times) for the gas-diffusion-electrode (GDE) used in Li-air cells was demonstrated through modification of the carbon surface with long-chain hydrophobic molecules. The capacity loss of the Li-air activated carbon cathode was found to be caused by the formation of undesired surface passivation. The mechanism of such passivation was identified as the formation of dense Li oxide films directly on the surface of the carbon during the oxygen reduction reaction. Such dense layers of Li oxide are here identified as the root cause of the undesired passivation, which blocks electrochemical reactions, increases the impedance and drops the discharge voltage rapidly. This investigation reveals that the capacity for the gas-diffusion-electrode can be substantially increased, if the activated carbon is modified by attaching long-chain hydrophobic molecules onto the surface. The carbon surface modification significantly delays the formation of the dense Li oxide layers. Therefore, the discharge capacity for the GDE is substantially increased. 相似文献

Commercially available, multi-walled carbon nanotubes grown by CVD are usually inherently entangled, but can be separated by cutting. However, most cutting methods both cause damage to the nanotubes and involve a lengthy work-up procedure. The use of abrupt, repeated exposure to oxidising conditions in air proved to be an efficient (68% yield) means of producing material with open ends, moderate functionalisation, and enhanced solvent dispersibility; the average lengths were reduced from over 5 μm to approximately 650 nm. Additionally, the character of the surface oxides can be tuned to have either an acidic or basic character by using a simple thermal treatment. These approaches could be deliberately integrated into conventional CVD processes, but also have implications for the products of standard nanotube syntheses. Raman spectroscopy and electron microscopy were used to study the impact of cutting on the intrinsic graphitic structure and the length distribution. X-ray photoelectron spectroscopy was used to determine the extent of functionalisation. The cut carbon nanotubes were dispersed in dimethylformamide (DMF), a Lewis basic solvent, and chloroform, a Lewis acidic solvent, using mild sonication. Through the use of an experimentally determined extinction coefficient (ε = 35.10 ml mg−1 cm−1), the relative dispersibility of the cut and functionalised carbon nanotubes in DMF and chloroform was determined. 相似文献

In this paper, a zinc oxide (ZnO) nanotube, fabricated by the hydrothermal growth method on triple-junction (T-J) solar cell devices to enhance efficiency, is investigated. Compared to those of bare T-J solar cells (without antireflection (AR) coating) and solar cells with Si3N4 AR coatings, the experimental results show that the T-J solar cells, which use a ZnO nanotube as an AR coating, have the lowest reflectance in the short wavelength spectrum. The ZnO nanotube has the lowest light reflection among all experimental samples, especially in the range of 350 to 500 nm from ultraviolet (UV) to visible light. It was found that a ZnO nanotube can enhance the conversion efficiency by 4.9%, compared with a conventional T-J solar cell. The Si3N4 AR coatings also enhance the conversion efficiency by 3.2%.The results show that a cell with ZnO nanotube coating could greatly improve solar cell performances. 相似文献
