A simple and rapid liquid chromatographic method was developed for the simultaneous determination of twelve benzimidazole anthelmintics in livestock foods using reversed-phase high-performance liquid chromatography with photodiode array detection (PDA). A sample was homogenized with acetonitrile and n-hexane, and centrifuged. The acetonitrile phase was isolated and evaporated. The residue was dissolved in 0.1 mol/L carbonate buffer solution (pH = 9.1), sonicated, and then subjected to clean-up on a Bond Elut LRC-C18 cartridge. The benzimidazole compounds were separated isocratically on a Capcell Pak C18 UG 120 (5 microns, 150 x 4.6 mm i.d.) column and detected by PDA at 295 and 313 nm. Mixtures of acetonitrile and 0.05 mol/L ammonium acetate in mixing ratios of (20:80) and (40:60) were used as the mobile phase, and the flow-rate was 1.0 mL/min at 40 degrees C. The mean recoveries (n = 3) from 0.1-0.5 microgram/g added samples were 72.6-97.2% with coefficients of variation of 0.3-8.5%. The detection limits were 0.01-0.05 microgram/g.  相似文献

We report a new application of affinity capture-elution electrospray mass spectrometry (ACESI-MS) to assay the enzymes phosphomannomutase (PMM) and phosphomannose isomerase (PMI), which when deficient cause congenital disorders of glycosylation CDG-type Ia and type Ib, respectively. The novel feature of this mass-spectrometry-based assay is that it allows one to distinguish and quantify enzymatic products that are isomeric with their substrates that are present simultaneously in complex mixtures, such as cultured human cell homogenates. This is achieved by coupled assays in which the PMM and PMI primary products are in vitro subjected to another enzymatic reaction with yeast transketolase that changes the mass of the products to be detected by mass spectrometry. The affinity purification procedure is fully automated, and the mass spectrometric analysis is multiplexed in a fashion that is suitable for high-throughput applications.  相似文献

Behaviors of bubble(s) exposed to an ultrasonic vibration are focused. Size of the bubbles interested in the present study is of O(1 mm) in diameter. The bubbles were injected through the micro syringe to the test fluid (water or water/surfactant mixture) filled in the rectangular tank. Ultrasonic vibration was irradiated to the bubble(s) after the detachment of the bubble from the tip of the syringe; thus the bubbles were exposed to the periodic oscillation in rising the test fluid. The authors clearly detect radial and shape oscillations under the large-amplitude vibration by use of high-speed camera. Preferable mode number of the shape oscillation, and the transition process from the radial to the shape oscillation are discussed.  相似文献

Photochemical diodes were prepared using a thin film of TiO2 made from titanium isopropoxide by a sol-gel method. A half part of a conductive SnO2 film plated on a quartz plate was coated with the TiO2 film, and the other parts of the SnO2 substrate were coated with a thin Pt film. The photochemical diode thus prepared shows photocatalytic activity for hydrogen evolution from aqueous ethanol solutions containing NaOH or HClO4. No other products are detected in the gas phase. Hydrogen evolution from the Pt part is definitely observed, indicating electron transfer from the TiO2 film to the Pt film through the SnO2 film. Another type of photochemical diode, the TiO2 film coated on a Pt plate, is found to show higher activity than the TiO2/SnO2/Pt device for the hydrogen photo-evolution without any support electrolyte. Hydrogen evolution on this device, however, occurs on the TiO2 side but not on the Pt side. These results are discussed in terms of a photo-electrochemical mechanism at semiconductor surfaces.  相似文献

Polycyclic aromatic hydrocarbons (PAHs), widespread environmental pollutants, were recently reported to show photomutagenesis. As contaminants in the environment are usually exposed to sunlight, a way to evaluate the phototoxic characteristics of pollutants is required. We have previously found that phosphorylation of histone H2AX (gamma-H2AX), which accompanied the induction of DNA double strand breaks (DSBs), was significantly induced by low concentrations of benzo[a]pyrene (10(-9)-10(-7) M) and UVA (0.6 J/cm2) in CHO-K1 cells. Higher concentrations have been required for the detection of DSBs. The aim of the present study is to investigate the applicability of gamma-H2AX in a new phototoxicity assay of PAHs. The human keratinocytes, HaCaT, were treated with four model PAHs (naphthalene, phenanthrene, pyrene, and benzo[a]pyrene, 10(-11)-10(-7) M) and/or UVA (5 J/cm2), and the induction of gamma-H2AX was assessed. Furthermore, DSBs were directly detected using a biased sinusoidal field gel electrophoresis, and the cell viability was examined as a general assay of phototoxicity. The induction of gamma-H2AX was detected in the presence of all the PAHs except naphthalene at concentrations of 10(-9)-10(-7) M, whereas neither DSBs nor cell death could be detected at those concentrations, and higher concentrations were required for the detection. Naphthalene showed no phototoxicity in any of the three different assays. These findings suggest that histone H2AX is a potential moleculartargetfor detecting the phototoxicity of PAHs more sensitively than the detection of cell viability and DSBs.  相似文献

PURPOSE: To report a patient with acute retinal pigment epitheliitis examined less than 24 hours after onset of symptoms. METHOD: One day after the onset of blurred vision in her left eye, a 33-year-old woman had a best-corrected visual acuity of LE, 20/60 -2. The left eye had classic uniform golden-colored nodules in a honeycomb pattern in the foveal retinal pigment epithelium. Intravenous fundus fluorescein angiography disclosed staining of the foveal pigment epithelium. RESULTS: One month after initial examination, visual acuity was LE, 20/20, and fine subfoveal pigmentary clumping was present. CONCLUSION: The pigmentary maculopathy of acute retinal pigment epitheliitis may be nonspecific, resulting from more than one type of primary foveal inflammation.  相似文献

We present a zero-power magnetic levitation technique using a composite of magnetostrictive and piezoelectric materials. The composite is bonded to iron yokes with an attached permanent magnet, by which the magnetic force exerted on movable yoke via air gap is controlled by the applied voltage on the piezoelectric material. The magnetic force control is based on the inverse magnetostrictive effect of the magnetostrictive material, i.e., the magnetization is varied with mechanical stress. The advantage of the composite is zero power consumption, because no current flows in static operation as a result of the capacitive property of the piezoelectric material. This feature will be useful in high-precision stage or conveyor systems using magnetic levitation where heat generation and power consumption should be avoided. The zero power characteristic of the composite is valid at any reference gap or load, whereas that of the conventional electromagnetic type is valid only at the equilibrium gap. We performed two levitation experiments: one using the composite to demonstrate the zero power advantage, and the other combining the composite to adjust the bias gap and electromagnet to stabilize the motion of the levitated yoke. The composite driven by a small dc-dc converter successfully varied the gap and maintained it constant with zero power consumption.  相似文献
