Due to the already prevalent and increasing use of silver-nanoparticle (Ag-NP) products and the raised concerns in particular for the aquatic environment, analytical techniques for the characterization of such products are of need. However, because Ag-NP products are of different compositions and polydispersities, analysis especially of the size distribution is challenging. In this work, an asymmetric flow field flow fractionation (A4F) multidetector system (UV/vis, light scattering, inductively coupled plasma mass spectrometry - ICPMS), in combination with a method to distinguish and quantify the particle and dissolved Ag fractions (ICPMS after ultracentrifugation), for the characterization of Ag-NP products with different degrees of polydispersities is presented. For validation and to outline benefits and limitations, results obtained from batch dynamic light scattering (batch-DLS) and transmission electron microscopy (TEM) were compared. With the developed method a comprehensive understanding in terms of dissolved Ag and Ag-NP concentration as well as an element selective, mass- and number particle size distribution (PSD) was obtained. In relation to batch-DLS, the reliability of the data was improved significantly. In comparison to TEM, faster measurement times and the ability to determine the samples directly in dispersions are clearly advantageous. The proposed setup shows potential for a rapid- and reliable characterization method of virtually any polydisperse metallic NP dispersion, many of them available on the market already.  相似文献

The microwave (MW)‐assisted synthesis of one dimensional carbon systems is introduced as a promising approach to improve the speed and cost‐effectiveness of the fabrication process. Improved reaction conditions are generated by direct MW heating and synthesis under advanced reaction conditions. The influence of the reaction conditions is investigated and the importance of individual process parameters on the synthesis is discussed. Temperature and pressure data recorded during the irradiation process are analyzed in detail and allow the determination of essential process parameters. This leads to improved reaction conditions, better control of the one‐dimensional carbon nanosystems by tuning the catalyst materials, and allows expanding this approach to initiate the synthesis on a variety of different substrates, such as quartz glass and mica.  相似文献

The mechanical behavior of an electrode during implantation into neural tissue can have a profound effect on the neural connections and signaling that takes place within the tissue. The objective of the present work was to investigate the in vivo implant mechanics of flexible, silicon-based ACREO microelectrode arrays recently developed by the VSAMUEL consortium (European Union, grant #IST-1999-10073). We have previously reported on both the electrical [1]-[3] and mechanical [4], [5] properties of the ACREO electrodes. In this paper, the tensile and compression forces were measured during a series of in vivo electrode insertions into the cerebral cortex of rats (7 acute experiments, 2-mm implant depth, 2-mm/s insertion velocity). We compared the ACREO silicon electrodes (40 opening angle, 1-8 shafts) to single-shaft tungsten electrodes (3 degrees and 10 degrees opening angles). The penetration force and dimpling increased with the cross-sectional area (statistical difference between the largest and the smallest electrode) and with the number of shafts (no statistical difference). We consistently observed tensile (drag) forces during the retraction phase, which indicates the brain tissue sticks to the electrode within a short time period. Treating the electrodes prior to insertion with silane (hydrophobic) or piranha (hydrophilic) significantly decreased the penetration force. In conclusion, our findings suggest that reusable electrodes for acute animal experiments must not only be strong enough to survive a maximal force that exceeded the penetration force, but must also be able to withstand high tension forces during retraction. Careful cleaning is not only important to avoid foreign body response, but can also reduce the stress applied to the electrode while penetrating the brain tissue.  相似文献

The performance of a novel portable, tunable diode laser, resonant photoacoustic (TDL-PA) analyzer developed for field measurements of CH4 was compared to a commonly applied offline gas chromatographic (GC) method. This comparative studywas realized under normal field conditions parallel to long-term weekly GC monitoring of four different soil types with very different methane budgets. The method used for gas-exchange measurements was the well-known closed-chamber technique. The TDL-PA analyzer detects methane at 1650.957 nm [R (5) line of the 2v3 band], guaranteeing high precision without the need for correction procedures. The two techniques correlated well (R2 = 0.988) over the entire concentration range (0.15-33 ppmv CH4) tested at highly varying flux rates between -30 and -12 ppbv CH4 min(-1) for uptakes and between 2.5 and 362 ppbv CH4 min(-1) for emissions. The two analyzers proved to be interchangeable, leaving the online advantages to the TDL-PA. A suitable CH4 online GC solution for chamber measurement is not available as a portable system. Additionally, the data sampling rate of 2 Hz enables a direct coupling to other infrared gas analyzers with the high time resolution commonly required to determine plant CO2 assimilation rates or soil respiration rates.  相似文献

Preparative synthesis with P450 monooxygenases is hampered in cell-free systems by the requirement for cofactors such as NAD(P)H as reduction equivalents. A validated medium-throughput screening system was designed for improving P450 monooxygenases by mediated electron transfer with zinc/cobalt(III)sepulchrate (Zn/Co(III)sep) as an alternative and cost-effective cofactor system. The monooxygenase P450 BM-3 F87A was used as a model system for developing the screening system in a 96-well format. A coefficient of variation of less than 10% was achieved under optimized screening conditions. The mediator evolution screen was validated by comparing the activity of P450 BM-3 to P450 BM-3 F87A and by screening a saturation mutagenesis library at amino acid position R47. For mediated electron transfer, two double mutants P450 BM-3(F87A R47F) and P450 BM-3 (F87A R47Y) were identified with a two-threefold increased catalytic efficiency (up to 32 microM(-1) min(-1) for P450 BM-3(F87A R47F) and 34 microM(-1) min(-1) for P450 BM-3 (F87A R47Y)) compared to P450 BM-3 F87A. The kinetic constants of the double mutants are, in contrast to those of P450 BM-3 F87A, dependent on Co(III)sep concentration in the presence of NADPH. kcat increases from 145 min(-1) (0.25 mM Co(III)sep) to 197 min(-1) (0.5 mM Co(III)sep), and Km decreases simultaneously from 7.0 microM to 3.7 microM, for P450 BM-3 (F87A R47F). For P450 BM-3 (F87A R47Y), kcat increases from 138 min(-1) (0.25 mM Co(III)sep) up to 187 min(-1) (0.5 mM Co(III)sep), and Km decreases from 8.2 microM to 4.2 microM. Due to lower Km values, the catalytic efficiencies were improved six times for P450 BM-3 (F87A R47F) and three times for P450 BM-3 (F87A R47Y), when comparing catalytic efficiencies of the mediated electron-transfer system to the natural reduction equivalent NADPH.  相似文献

X-ray crystallographic and functional analysis of the class I DNA photolyase from Thermus thermophilus revealed the binding of flavin mononucleotide (FMN) as an antenna chromophore. The binding mode of FMN closely coincides with the binding of a deazaflavin-like chromophore in the related class I DNA photolyase from Anacystis nidulans. Compared to the R46E mutant, which lacks a conserved arginine in the binding site for the antenna chromophore, the FMN-comprising holophotolyase exhibits an eightfold higher activity at 450 nm. The facile incorporation of the flavin cofactors 8-hydroxy-deazariboflavin and 8-iodo-8-demethyl-riboflavin into the binding site for the antenna chromophore paves the way for wavelength-tuning of the activity spectra of DNA photolyases by using synthetic flavins.  相似文献

Silicon nanowires have been identified as important components for future electronic and sensor nanodevices. So far gold has dominated as the catalyst for growing Si nanowires via the vapour-liquid-solid (VLS) mechanism. Unfortunately, gold traps electrons and holes in Si and poses a serious contamination problem for Si complementary metal oxide semiconductor (CMOS) processing. Although there are some reports on the use of non-gold catalysts for Si nanowire growth, either the growth requires high temperatures and/or the catalysts are not compatible with CMOS requirements. From a technological standpoint, a much more attractive catalyst material would be aluminium, as it is a standard metal in Si process lines. Here we report for the first time the epitaxial growth of Al-catalysed Si nanowires and suggest that growth proceeds via a vapour-solid-solid (VSS) rather than a VLS mechanism. It is also found that the tapering of the nanowires can be strongly reduced by lowering the growth temperature.  相似文献
