This paper describes a two-stage process for separating milk proteins from pasteurized skim milk in three fractions: casein micelles, β-Lactoglobulin (β-Lg) and other large whey proteins, and α-Lactalbumin (α-La). Casein micelles were extracted in the retentate of a microfiltration using rotating ceramic disk membranes. α-La and β-Lg transmissions remained between 0.8 and 0.98. Their yields in permeate reached 81% for α-La and 76.6% for β-Lg at a VRR of 5.4. The separation between β-Lg and α-La was carried out by UF using a rotating disk module equipped with a 50 kDa PES circular membrane. Permeate fluxes were very high, remaining above 340 L h?1 m?2 at VRR = 5 and 40 °C. α-La transmission remained generally between 0.2 and 0.13 giving yields from 28% to 34%. β-Lg rejection was above 0.94, giving a maximum selectivity of 4.2. These data confirm the potential of dynamic membrane filtration for separating α-La and β-Lg proteins from skim milk.  相似文献

Experimental analysis concerning supercritical flow over a dentated sill in nonsubmerged conditions and the local scour downstream of this sill is presented. In this case, the energy dissipation is incomplete so that a scour control downstream of the hydraulic structure could be required. The use of this type of dissipation device could be needed in cases where the maximum flow depth upstream of the sill must be limited to satisfy physical or structural conditions. This could happen, for instance, in the restoration of stilling basin in gravity dams and at the outlet of diversion systems. Experimental and theoretical results, useful in the design of this type of stilling basin and scour control, are presented in the paper, with an application to the case of the Molato Dam in Italy.  相似文献

Together with the evolution of buildings and systems, energy assessment models have become more and more detailed, requiring a precise evaluation of the building loads and gains.For an accurate analysis of solar heat gains, technical standards introduce an external shading reduction coefficient of the incident solar radiation, called shading factor. Its value is provided for simple geometries, which are usually not suitable to properly describe a real environment. In addition, the relations adopted to evaluate the shading factor are based on simplified hypotheses, which cause non-negligible errors when compared to more complex algorithms.The objective of this study is the development of a calculation procedure of the shading factor under complex boundary conditions. The algorithms have been implemented in a software tool written in Matlab language. It can provide for the value of the shading factor on a generically oriented and tilted surface. After setting the site and the time for the simulation, generic-shaped windows can be modelled. The external environment, which can be imported from DXF files, can include a horizon profile, generic-shaped obstructions and vegetation. The calculation can be performed for every sky condition: clear, average or generic. In addition, the simulation can be run to obtain instantaneous, daily average or monthly average shading factor values.  相似文献

In insects, β-N-acetylglucosaminidase (GlcNAcase) participates in critical physiological processes such as fertilization, metamorphosis, and glycoconjugate degradation. Insects produce glycoproteins carrying paucimannosidic-type N-glycans, the terminal GlcNAc residue of which is cleaved by a GlcNAc-linkage specific GlcNAcase, also known as the fused lobes (FDL) protein. To obtain information on the structure of GlcNAcases and insight into their contribution to physiological processes, we cloned Bombyx mori FDL (BmFDL) from silkworm larvae. The full-length cDNA (1.9 kb) encoded a protein of 633 amino acids with 42% amino acid sequence identity to Drosophila melanogaster FDL (DmFDL). Recombinant BmFDL cleaved only β-1,2-linked GlcNAc residues from the α-1,3 branch of biantennary N-glycan. This substrate specificity was similar to that of DmFDL. Microsomal FDL activity was inhibited by anti-BmFDL antibodies. Taken together, our results suggest that BmFDL is a N-glycan-processing GlcNAcase in B. mori.  相似文献

BACKGROUND: Plums are a rich source of nutritive and bioactive compounds. The objective was to evaluate the impact of fruit tissue (flesh and peel), ripening stage and cultivar on the composition of phytochemicals in plums (Prunus domestica L.) and to analyse the relation between total antioxidant activity (TAA) and the measured variables. RESULTS: The content of total sugars and total organic acids was higher in flesh but total phenolic content (TPC) and TAA was statistically higher in plum peel. The composition of sugars depended on fruit tissue, ripening stage and the cultivar. Ripening and fruit tissue affected the composition and concentrations of organic acids. TPC of ‘Haganta’ peel increased during ripening but there was no significant time trend for ‘Jojo’ peel and flesh of both cultivars. The composition of phenolics in peel depended on the cultivar, but in flesh on the cultivar and ripening. Ripening resulted in increased total anthocyanins and TAA in peel. TAApeel was positively correlated with TPCpeel, total anthocyaninspeel, cyanidin 3‐glucosidepeel and peonidin 3‐rutinosidepeel in the cultivar ‘Haganta’. CONCLUSION: The composition of phytochemicals in P. domestica fruit depends on fruit tissue, ripening stage and the cultivar. BACKGROUND: Bioethanol is mostly produced from starchy parts of the corn grain kernel leaving significant amounts of valuable by‐products such as dried distillers' grains with solubles (DDGS) which can be used as a substitute for traditional feedstuff. The suitability of six maize hybrids from Serbia was investigated for bioethanol and DDGS production. The correlation between physical and chemical characteristics of the grain, bioethanol yield and quality of the corresponding DDGS was assessed. RESULTS: All hybrids had very different chemical composition and physical characteristics which could allow various applications. The highest bioethanol yield (94.5% of theoretical) and volumetric productivity (2.01 g l?1 h?1) were obtained with hybrid ZP 434 and the lowest with ZP 611k. Regarding chemical composition, all DDGS samples manifested good properties as feed components. Their protein content was higher compared to the kernel. In addition, the samples showed high digestibility and high mineral content, especially of calcium and phosphorus. CONCLUSION: A hybrid ZP 434 was selected as the most promising bioethanol producer. This property is attributed to the highest level of soft endosperm which is more susceptible to starch‐hydrolysing enzymes. A high yield potential per hectare makes it the best candidate for commercial bioethanol production. © 2012 Society of Chemical Industry  相似文献

For the first time new type of liquid single crystal elastomers forming the chiral smectic A* (SmA*) phase has been prepared using chiral lactic acid derivative as a co-monomer. The synthesis and the basic characterization of the ferroelectric liquid crystalline co-monomer based on differential scanning calorimetry and small/wide-angle X-ray scattering are reported and discussed. The monomer possesses the paraelectric SmA* and the ferroelectric SmC* phases over a broad temperature range. The preparation of new smectic liquid crystalline elastomers together with characterization of their mesomorphic and structural properties is also reported. New elastomers possess the orthogonal paraelectric SmA* phase over 50 K broad temperature range.  相似文献

We examined two muscle biopsy specimens from a proband and her mother with myoclonus epilepsy with ragged-red fibers (MERRF), both obtained at an interval of about 10 years, using histochemistry, in situ hybridization, and single-fiber polymerase chain reaction. Total (wild-type and mutant) mitochondrial DNAs (mtDNAs) were greatly increased in ragged-red fibers (RRF) over non-RRF in all muscle specimens analyzed. The proportion of mutant mtDNA was also significantly higher in RRF than in non-RRF. By comparing the first and second muscle biopsied specimens in each patient, we found that while the proportion of RRF, cytochrome coxidase deficient fibers, and mutant DNA in muscle changed over a 10-year period, the proportion of wild-type and mutant mtDNAs in RRF and in non-RRF was similar between the two specimens. These results suggest that the ratio of wild-type to mutant mtDNAs in RRF and non-RRF in MERRF is at a steady state level in each muscle fiber, without replicative advantage of mutant mtDNA.  相似文献
